Characterization of Citrus tristeza virus isolates from grapefruit (Citrus paradisi Macf.) accessions of Citrus Active Germplasm Bank
Corazza-Nunes, Maria Júlia;Machado, Marcos Antonio;Stach-Machado, Dagmar Ruth;Nunes, William Mário Carvalho;Carvalho, Sérgio Alves de;Müller, Gerd Walter;
Summa Phytopathologica , 2006, DOI: 10.1590/S0100-54052006000400002
Abstract: citrus tristeza virus (ctv) isolates from 35 grapefruit accessions belonging to citrus active germplasm bank of the "instituto agron?mico de campinas" located at the "centro apta citros sylvio moreira", cordeirópolis, s?o paulo state, brazil, were characterized and evaluated through symptoms in the trees, biological indexing, immunological diagnosis with different monoclonal antibodies and sscp analysis (single-strand conformation polymorphism) of the coat protein gene. symptomatology indicated that, in general, the group of plants with smaller canopy volume and severe stem pitting differed significantly from the group that presented greater vegetative development and mild to moderate stem pitting. however, the isolates from most of the accessions induced mild reaction on mexican lime. the serological evaluation through the das-elisa using monoclonal antibodies did not reveal any association between virus titer in the plant tissue and symptoms. the reaction with different monoclonal antibodies and the distinct electrophoresis patterns obtained through sscp showed that there is a high degree of diversity among the isolates that infect these grapefruit accessions. high complexity within the same isolate was also observed in the sscp profiles. this finding indicates that the ctv isolates from these plants are a complex mixture of ctv haplotypes. similar sscp banding patterns were observed among some plants with strong stem pitting symptoms, and among some plants with weak or moderate stem pitting symptoms.
Expression profile of oxidative and antioxidative stress enzymes based on ESTs approach of citrus
Peroni, Luis Antonio;Ferreira, Renato Rodrigues;Figueira, Antonio;Machado, Marcos Antonio;Stach-Machado, Dagmar Ruth;
Genetics and Molecular Biology , 2007, DOI: 10.1590/S1415-47572007000500016
Abstract: plants not only evolve but also reduce oxygen in photosynthesis. an inevitable consequence of this normal process is the production of reactive oxygen species (ros). plants are adequately protected by the presence of multiple antioxidative enzymes in the cytosol and also in the different cell organelles such as chloroplasts, mitochondria, and peroxisomes. traditionally, ros were considered to be only a toxic byproduct of aerobic metabolism. however, recently it has become apparent that plants actively produce these molecules which may control many different physiological processes such as abiotic and biotic stress response, pathogen defense and systemic signaling. the search results using the citrus genome program in brazil (citest) for oxidative stress and the antioxidant enzyme system in citrus sinensis variety ?pera iac? indicated that the multiple ros-scavenging enzymes were expressed throughout all citrus tissues. the analyses demonstrated the ubiquitous expression of metallothioneins, probably indicating a constitutive expression pattern. oxalate oxidase has been identified as the most abundant expressed gene in developing fruits, which suggests a specific function in the ripening of citrus fruit. moreover, infected leaves with xylella fastidiosa and leprosis citri showed a massive change in their ros gene expression profile which may indicate that the suppression of ros detoxifying mechanisms may be involved in the induction of the diseases.

Abstract: the objective of this study was to verify the notification of information system for oral clefts in the live births declaration (dnv), studying the 3 items of dnv related to malformation. all the patients' medical records registered in oral cleft reference centers were checked. the inclusion criteria were: a) to have any oral cleft and b) to be born from 01/01/1999 to 12/31/2004. next step was to raise the information in the vital data sector of municipal health secretary, where all the dnvs identified were subjects with oral clefts (63 children). the first item presented a large subnotification (only 53.3 of the dnvs showed the register of malformation) and all types of clefts were not notified in any scale. highest sub notification was observed for cleft palate (70 %). second item (description of malformation) showed similarly a high number of errors referred to palate cleft, identified only in 25 % of the cases. all the documents showed no information about the third item (code of malformation). there was no statistical difference between professionals responsible for filling out the document. in conclusion, the dnv was inefficient to register the oral cleft cases due to imperfections related to 3 items studied, especially in the absence of information about cid-10 codes.

Abstract: the purpose of the present study was to determine the prevalence of oral clefts in the city of campos dos goytacazes-rj/brazil, in children born from january 01, 1999 to december 31, 2004. age group, cleft type, sex and race of the child were considered, as well as the marital status, education and origin of the child's mother at the time of birth. in this manner, the records of patients enrolled in reference services for treatment of craniofacial anomalies in the cities of campos dos goytacazes (rj), rio de janeiro (rj) and in bauru (sp) were reviewed with the purpose of selecting those whose birth and/or residence was in the city of campos dos goytacazes, within the target period. after the initial selection, a search in the islb was conducted to define the universe of the study, which was: finding the children who were born alive between 1999 and 2004 whose mothers were living in campos dos goytacazes. in this period, using as a reference the mother's residence, the number of children born in the city was of 46,707, of which 63 had cleft lip with or without cleft palate, representing a prevalence of 1.35 cases in every 1,000 live births. the analysis of the data revealed that 55.6% of the patients were male; the most common type of cleft was isolated cleft palate and cleft lip and palate (34.9%); 61.3% of the cases were white children but the percentage of cases within each race did not differ statistically in relation to the distribution of births in the population; most of the mothers were single or married with little education.
On the origin of the regioselective hydrolysis of a naphthoquinone diacetate: a molecular orbital study
Longo, Ricardo L.;Nunes, Ruth L.;Bieber, Lothar W.;
Journal of the Brazilian Chemical Society , 2001, DOI: 10.1590/S0103-50532001000100007
Abstract: the regioselectivity found in the mild basic hydrolysis of the 2,5-dimethyl-1,4-naphthohydroquinone diacetate (nunes, r.l.; bieber, l.w.; longo, r.l. j. nat. prod. 1999, 62, 1600) has been studied with ab initio and semiempirical molecular orbital methods. in the gas phase (isolated systems), these methods were not able to provide results that could explain the observed selectivity. however, when the solvent effects were included in the am1 method using the discrete solvation model it was possible to establish that this selectivity is due to the relative stability of the tetrahedral intermediates and their transitions states. the origin of this relative stability and thus of the observed selectivity is due to the repulsive interactions between the 2-methyl substituent in the naphthalene ring and the methyl group in the 4-acetate substituent, as well as their hindrance towards the hydration of the ionic group in the tetrahedral intermediates.

Psychology (PSYCH) , 2014, DOI: 10.4236/psych.2014.55051
Abstract:

On April 15, 2008, six students (aged 16 years) and one teacher (aged 29 years) from a New Zealand school lost their lives in a river canyoning tragedy. The present study investigated the school principal’s perspective of how he led his school through the tragedy, and the role of faith in the school’s coping. The school principal was interviewed two years after the event. The school’s Christian foundation was the fundamental source of strength and guidance for the principal, as well as for students, staff, teachers, and families in the immediate aftermath of the tragedy and in the two years following (i.e., to the time of the present study), the Christian culture of the school guiding decision-making. Support from outside the school (e.g. critical incident support; teaching support from other schools; social support from community agencies and civic leaders) also played an important role in assisting the school through the tragedy, particularly in the immediate aftermath of the event. Further studies are required that allow the voices of children, families and school staff to be heard regarding leadership strategies that impact on them through a disaster.
