Auto-Ubiquitination-Induced Degradation of MALT1-API2 Prevents BCL10 Destabilization in t(11;18)(q21;q21)-Positive MALT Lymphoma

Abstract: Background The translocation t(11;18)(q21;q21) is the most frequent chromosomal aberration associated with MALT lymphoma and results in constitutive NF-κB activity via the expression of an API2-MALT1 fusion protein. The properties of the reciprocal MALT1-API2 were never investigated as it was reported to be rarely transcribed. Principal Findings Our data indicate the presence of MALT1-API2 transcripts in the majority of t(11;18)(q21;q21)-positive MALT lymphomas. Based on the breakpoints in the MALT1 and API2 gene, the MALT1-API2 protein contains the death domain and one or both immunoglobulin-like domains of MALT1 (~90% of cases) - mediating the possible interaction with BCL10 - fused to the RING domain of API2. Here we show that this RING domain enables MALT1-API2 to function as an E3 ubiquitin ligase for BCL10, inducing its ubiquitination and proteasomal degradation in vitro. Expression of MALT1-API2 transcripts in t(11;18)(q21;q21)-positive MALT lymphomas was however not associated with a reduction of BCL10 protein levels. Conclusion As we observed MALT1-API2 to be an efficient target of its own E3 ubiquitin ligase activity, our data suggest that this inherent instability of MALT1-API2 prevents its accumulation and renders a potential effect on MALT lymphoma development via destabilization of BCL10 unlikely.
Nodular Lymphocyte Predominant Hodgkin Lymphoma and T Cell/Histiocyte Rich Large B Cell Lymphoma - Endpoints of a Spectrum of One Disease?

Abstract: In contrast to the commonly indolent clinical behavior of nodular lymphocyte predominant Hodgkin lymphoma (NLPHL), T cell/histiocyte rich large B cell lymphoma (THRLBCL) is frequently diagnosed in advanced clinical stages and has a poor prognosis. Besides the different clinical presentations of these lymphoma entities, there are variants of NLPHL with considerable histopathologic overlap compared to THRLBCL. Especially THRLBCL-like NLPHL, a diffuse form of NLPHL, often presents a histopathologic pattern similar to THRLBCL, suggesting a close relationship between both lymphoma entities. To corroborate this hypothesis, we performed gene expression profiling of microdissected tumor cells of NLPHL, THRLBCL-like NLPHL and THRLBCL. In unsupervised analyses, the lymphomas did not cluster according to their entity. Moreover, even in supervised analyses, very few consistently differentially expressed transcripts were found, and for these genes the extent of differential expression was only moderate. Hence, there are no clear and consistent differences in the gene expression of the tumor cells of NLPHL, THRLBCL-like NLPHL and THRLBCL. Based on the gene expression studies, we identified BAT3/BAG6, HIGD1A, and FAT10/UBD as immunohistochemical markers expressed in the tumor cells of all three lymphomas. Characterization of the tumor microenvironment for infiltrating T cells and histiocytes revealed significant differences in the cellular composition between typical NLPHL and THRLBCL cases. However, THRLBCL-like NLPHL presented a histopathologic pattern more related to THRLBCL than NLPHL. In conclusion, NLPHL and THRLBCL may represent a spectrum of the same disease. The different clinical behavior of these lymphomas may be strongly influenced by differences in the lymphoma microenvironment, possibly related to the immune status of the patient at the timepoint of diagnosis.

Abstract: Este artigo aborda o combate que empreendeu Gustavo Cor o à juventude católica brasileira nas páginas do Diário de Notícias e d’ O Globo entre as décadas de 1950 e de 1960. No caso em tela, interessa-nos entender as rea es de Gustavo Cor o às mudan as na sociedade e na Igreja Católica entre 1957 e 1964. A hipótese é que as críticas de Cor o explicitam o conflito entre duas gera es de católicos: uma conservadora; a dele; e outra progressista, a das organiza es de jovens católicos, mais especificamente, a da Juventude Universitária Católica (JUC). Apresentaremos a disputa a partir do exame da seguinte quest o: o papel que a juventude brasileira deveria desempenhar naqueles anos. Abstract: This paper discusses the fight of Brazilian Catholic Youth Gustavo Cor o in the newspapers Diário de Notícias and O Globo in the 1950s and 1960s. In case we are interested in understanding the reactions of Gustavo Cor o to changes in society and the Catholic Church between 1957 and 1964. The hypothesis is that the criticism of Cor o explains the conflict between two generations of Catholics: a conservative and other progressive, represented by the Catholic youth organizations, more specifically, the Youth Catholic University. We are going to examine the question: the role that Brazilian youth should play in those years. Therefore, the aim is to show that his criticism of the political action undertaken by the young catholic organizations should be read by the key of generation conflict which has marked the Brazilian Catholic field. A note on quantum algorithms and the minimal degree of epsilon-error polynomials for symmetric functions

Abstract: The degrees of polynomials representing or approximating Boolean functions are a prominent tool in various branches of complexity theory. Sherstov recently characterized the minimal degree deg_{\eps}(f) among all polynomials (over the reals) that approximate a symmetric function f:{0,1}^n-->{0,1} up to worst-case error \eps: deg_{\eps}(f) = ~\Theta(deg_{1/3}(f) + \sqrt{n\log(1/\eps)}). In this note we show how a tighter version (without the log-factors hidden in the ~\Theta-notation), can be derived quite easily using the close connection between polynomials and quantum algorithms.
Genuine Correlations in Hadronic Z$^0$ Decays

Abstract: Correlations among hadrons with the same electric charge produced in Z$^0$ decays are studied using the high statistics data collected from 1991 through 1995 with the OPAL detector at LEP. Normalized factorial cumulants up to fourth order are used to measure genuine pa rticle correlations as a function of the size of phase space domains in rapidity, azimuthal angle and transverse momentum. tein correlations. Some of the recently proposed algorithms to simulate Bose-Einstein effects, implemented in the Monte Carlo model \PYTHIA, reproduce reasonably well the me asured second- and higher-order correlations between particles with the same charge as well as those in all-charge particle multiplets.
