Abstract:

Presented in several types of food, bioactive amines are described as organic bases of low molecular weight. They have vasoactive, psychoactive and toxicological characteristics and constitute a potential health risk. The concentration of amines formed in foods depends on the type of microorganisms present, the action of decarboxylase enzymes produced by microorganisms on specific amino acids and favorable conditions for enzymatic activity. The presence of these chemical metabolites has been suggested as a quality indicator in routine analyzes for food production and marketing monitoring. The detection of bioactive amines can be performed by chromatographic methods, fluorometric and enzymatic kits. Bioactive amine formation can be prevented mainly through the adoption of good manufacturing practices, but the industry can also use other methods such as temperature control in the production chain, modified atmosphere packaging and food irradiation. This review aims to address the formation of bioactive amines in foods, emphasizing the formation and classification of these metabolites, aspects related to health, acceptable limits, detection methods and control methods used in the industry to ensure food safety and quality. The success of this approach is linked to the importance of bioactive amines as quality indicators, as well as the discussion on the development of methodologies for determining these substances and discussion of acceptable parameters in food.

Quality of Semi-Prepared Products from Rainbow Trout Waste (Onchorynchus mykiss) by Using Different Technological Strategics  [PDF]

Abstract:

The consumption of freshwater fish and fish products has gradually grown worldwide over the last decades, generating a proportional waste increase. The objective of the present study was to assess the chemical and bacteriological quality of restructured fish product, meatball-type, prepared with rainbow trout (Onchorynchus mykiss) waste added of 1% transglutaminase (MTG), 4% textured soy protein (TSP) and replacing part of the sodium chloride with potassium chloride (75%/25%) as described below: T1—starch addition (control); T2—MTG addition (1%); T3—soy protein addition (4%); T4—soy protein addition (4%) and MTG addition (1%); T5—soy protein addition (4%), MTG addition (1%) and partial replacement of salt (75% NaCl/25% KCl). Total aerobic mesophilic bacteria (TAMB), 2-thiobarbituric acid reactive substances (TBARS), pH determination and quantification of biogenic amines were performed on the day after manufacturing (P0) and after 60 days of storage (P1) at -25℃ ± 2℃. The results showed that there was no significant difference (p < 0.05) of microbiological quality, TBARS and pH after storage. T4 presented the lowest total biogenic amine content (256.84 mg/kg) whereas T3 and T5 had the highest value (791.36 and 707.19 mg/kg, respectively) in this parameter. Putrescine was the biogenic amine that presented the highest concentration (504.00 mg/kg) in T3 and cadaverine that presented the smallest concentration (0.36 mg/kg) in T4. The use of technological strategies for developing new products with non-commercial fillets kept the most standards, having changes only in some biogenic amines.

Abstract: The objective of this research was to evaluate the effect of packaging in 100% air and vacuum, combined with radiation (2 kGy and 3 kGy), on chicken breast fillet shelf-life by evaluation of bacterial deteriorants growth and pH variation parameters. The vacuum packaging increased shelf-life, when compared to commercial packaging in 100% air, both in non-irradiated and irradiated to 3 kGy fillets. The irradiation increased the shelf-life of fillet samples at both doses. The lactic acid bacteria were the organisms that most developed in irradiated samples, showing greater radioresistance, as compared to other microorganisms studied, while the enterobacteria showed greater sensitivity to treatment with ionizing radiation. The vacuum packaging, combined with irradiation, can be used to improve the safety of chicken breast fillets and to extend its shelf-life. O objetivo deste experimento foi avaliar o efeito do uso de embalagens, em 100% ar e a vácuo, combinadas com a radia o gama (2 kGy e 3 kGy), no aumento do prazo de validade comercial de filé de peito de frango resfriado, avaliando-se os parametros de desenvolvimento de bactérias deteriorantes e a varia o do pH das amostras. A embalagem a vácuo aumentou o prazo de validade comercial, quando comparada à embalagem em 100% ar, tanto nos filés n o irradiados, como nos irradiados a 3 kGy. A irradia o das amostras, nas duas doses utilizadas, aumentou, consideravelmente, a validade comercial desse alimento. As bactérias láticas foram os micro-organismos que mais se desenvolveram nas amostras irradiadas, confirmando, assim, uma maior radiorresistência, quando comparada com os outros micro-organismos estudados, enquanto as enterobactérias demonstraram maior sensibilidade ao tratamento com radia o ionizante. A embalagem a vácuo, combinada com a tecnologia de irradia o, pode ser utilizada para melhorar a seguran a de filés de peito de frango resfriados e para estender seu prazo de validade comercial. Abstract: this paper investigated the effects of modified atmosphere packing (60% n2 and 40% co2) and irradiation (with a 1.5kgy dose) either used alone or combined on the shelf life of fillets of refrigerated nile tilapia. a total of 120 fillets were used in the experiments in order to test four different treatments and included samples: packed in air (control), packed in modified atmosphere, packed in air and irradiated, and finally, packed in modified atmosphere and irradiated. regular monitoring of physicochemical parameters (tvb, ph, ammonia and tbars), bacteriological conditions (heterotrophic aerobic mesophilic and psychrotrophic bacteria) and sensory acceptance were performed. the results consistently indicated an increasing deterioration of the fillets with time, however those treated with irradiation, modified atmosphere or both combined had longer shelf lives (around two weeks) in comparison with the control (4 days only).
Generalized composite interval mapping offers improved efficiency in the analysis of loci influencing non-normal continuous traits

Abstract: in genetic studies, most quantitative trait loci (qtl) mapping methods presuppose that the continuous trait of interest follows a normal (gaussian) distribution. however, many economically important traits of agricultural crops have a non-normal distribution. composite interval mapping (cim) has been successfully applied to the detection of qtl in animal and plant breeding. in this study we report a generalized cim (gcim) method that permits qtl analysis of non-normally distributed variables. gcim was based on the classic generalized linear model method. we applied the gcim method to a f2 population with co-dominant molecular markers and the existence of a qtl controlling a trait with gamma distribution. computer simulations indicated that the gcim method has superior performance in its ability to map qtl, compared with cim. qtl position differed by 5 cm and was located at different marker intervals. the likelihood ratio test values ranged from 52 (gcim) to 76 (cim). thus, wrongly assuming cim may overestimate the effect of the qtl by about 47%. the usage of gcim methodology can offer improved efficiency in the analysis of qtls controlling continuous traits of non-gaussian distribution.
Frequency of the BCR/ABL rearrangements and associated alterations detected by FISH during monitoring of patients taking imatinib mesylate in isolation

Abstract: the introduction of imatinib mesylate as treatment of chronic myelogenous leukemia has saved many patients, but the success of therapy is hampered by resistance and possible non-destruction of the malignant clone. this article describes the cytogenetic responses and abnormal cytogenetic patterns involving the abl and bcr genes detected by fish in patients who use exclusively imatinib. the results showed that other alterations involving the bcr and abl genes do not seem to be related to resistance to the drug as they occur in low frequencies and can not be associated to the cytogenetic response or to the time of treatment. moreover, the response to imatinib seems to be individual and unpredictable, independent of the time of treatment and of its initiation after diagnosis.
L-Arginine Supplementation and Nitric Oxide Production:No Additional Effect When Associated to Exercise  [PDF]

Abstract:

L-arginine is an amino acid semiessencial considered a precursor of nitric oxide, a gas mainly produced in endothelial cells. Nutritional supplements based on the amino acid L-arginine have been broadly marketed in order to increase vasodilation and the blood supply to muscle in order to optimize metabolic responses induced by exercise. The main objective of this study was to evaluate the effect of L-arginine supplementation on nitric oxide production in response to exercise. Furthermore, the biochemical parameters of muscle fatigue were assessed. Fourteen trained runners were divided in two groups, supplemented with L-arginine (ARG) and placebo (PLA). Blood samples were collected before supplementation (T0), immediately after the first exercise session (T1), immediately after the second exercise session (T2), and after 20 minutes of rest (T3). Plasma cyclic guanosine monophosphate was assessed as a marker of nitric oxide production. The biochemical parameters of muscle fatigue analyzed were plasma lactate and ammonia. There was significant increase in plasma cyclic guanosine monophosphate in both groups in response to exercise: ARG (T0: 3.6 ± 1.4; T1: 17.9 ± 5.8; T2: 15.9 ± 5.3; T3: 7.3 ± 2.5 pmol/mL) and PLA (T0: 4.1 ± 1.1; T1: 18.8 ± 9.9; T2: 16.1 ± 3.5; T3: 9.3 ± 3.7 pmol/mL). A significant reduction in plasma lactate and ammonia were observed in the recovery period after exercise (T3). However, no significant difference was observed between groups in the variables studied. Therefore, L-arginine supplementation was unable to increase the effects of exercise on nitric oxide production and did not improve the metabolic responses to exercise in runners.

Chemical Quality Indexes of Mullet (Mugil platanus) Stored on Ice  [PDF]

Abstract: The present study was conducted aiming at establishing chemical quality parameters to assess ice stored mullet (0 ± 1) through the evaluation of nucleotide (adenosine monophosphate [AMP], inosine [HxR] and hypoxanthine [Hx]) degradation, biogenic amine (histamine [HI], putrescine [PU], cadaverine [CA] and tyramine [TI]) quantification and mesophilic and psychrotrophic bacteria count monitoring. The microbial load of 7 log CFU·g1 established as maximum acceptable limit was attained after the 20th day of ice storage. IMP concentration declined during the storage period to levels below the detection limit. HxR content increased only up to time T3 and then declined. Hx level increased during all the storage period. CA and HI content increase was not observed, on the other hand, PU and TI contents significantly increased (p < 0.05) at time T5. We concluded that IMP and Hx concentrations can be adequate parameters to assess mullet quality under the study conditions. HxR proved to be adequate to evaluate the freshness of mullet in the first days of storage while the amines, PU and TI, can be used to assess loss of quality. Mullet obtained in conditions similar to those of the present study and maintained at 0 ± 1can be consumed up to the 20th storage day.
