Correlation between electrochemical and theoretical studies on the leishmanicidal activity of twelve Morita-Baylis-Hillman adducts

Abstract: enzymatic bioreduction of nitro groups plays an important role on the activity of biologically active nitroaromatic compounds. electrochemical methods are useful tools to simulate in vivo metabolic processes. this work presents electrochemical studies in aprotic media (n, n-dimethylformamide (dmf) plus tetrabutylammonium perchlorate (tbap) 0.1 mol l-1) using cyclic voltammetry (cv), differential pulse voltammetry (dpv) and square wave voltammetry (swv) of twelve morita-baylis-hillman adducts (mbha) with significant leishmanicidal activity. to facilitate the analysis, the molecules were grouped in four classes according to the side chain. cv studies show three up to four reduction waves, in which the first two waves are related to nitro group reduction. the other waves (presenting more negative potential) refer to the reduction of the α,β-unsaturated carbonyl or to the nitrile activated olefin side-chain. ortho adducts present facilitated reduction in comparison to the other isomers (meta and para) possibly due to hydrogen bond formation between the benzylic-oh and the nitro group, which stabilizes the reduction product (anion radical nitro) more efficiently than the original compound. ortho derivatives also present higher leishmanicidal activity upon comparison to the other derivatives of each class. conformational studies using hf/6-31+g(d)/pcm as a calculation level highlight this effect. molecular hardness (n) and atomic charges (qn) values corroborate the obtained experimental data.

Abstract: Este artigo apresenta diretrizes para realizar a gest o de requisitos durante o processo de projeto de ambientes construídos (PPAC). Uma vers o preliminar das diretrizes foi desenvolvida a partir de uma revis o da literatura e um estudo exploratório. Tal vers o foi parcialmente aplicada e refinada em um estudo de caso, no qual foram enfatizados os requisitos ambientais de um projeto urbanístico. A aplica o no estudo de caso exigiu o uso de múltiplas fontes de dados, tais como aproximadamente 12 horas de entrevistas, consulta a documentos (por exemplo, do licenciamento ambiental) e observa es de reuni es relacionadas ao PPAC. As diretrizes propostas têm uma vis o holística da GR, contemplando todas as suas etapas ao longo do processo de projeto, e houve a disponibiliza o de mecanismos que permitem controlar inevitáveis mudan as nos requisitos. Os dados coletados também possibilitaram avaliar as diretrizes em termos de sua utilidade e facilidade de uso. Dentre os resultados do estudo de caso, ficou evidenciada contribui o das diretrizes na coleta, organiza o e visibilidade dos requisitos ambientais, o que beneficia a tomada de decis o. This article introduces guidelines for managing requirements during the building of the environment design process. A preliminary version of these guidelines was developed based on a literature review and an exploratory study. Such version was partially applied and enhanced in a case study of an urban project, where the environmental requirements were emphasized. Multiple sources of data were adopted in the case study, such as 12 hours of interviews, analysis of documents (e.g., environmental licenses) and observation of meetings related to the design process. The proposed guidelines have a holistic view of requirements management and involve all its phases along the design process. In addition, techniques that allow the control of change were indicated. The data available were also useful for assessing the guidelines from the perspective of their usability and effectiveness. Among the insights obtained from the case study, the contribution of the guidelines was evident in the elicitation, organization and visibility of environmental requirements, which benefits the decision-making.

Abstract: The main goal is to compare the antioxidant potential of rosemary and oregano natural extracts in precooked beef burger by assessing the lipid oxidation extent and sensory analysis. Five formulations (F) of hamburger were prepared from beef and mechanically separated as follows: meat containing sodium erythorbate (F1); deodorized rosemary extract (F2); oregano extract (F3); rosemary plus oregano extracts (F4) and without antioxidant addition, denominated control formulation (CF). The samples were frozen at ?18°C for 24 hours, then submitted to heat treatment in an electric oven with internal controlled temperature of 75°C, and again frozen for a period of 30 days. The lipid oxidation extent (determined by thiobarbituric acid reactive substances— TBRS) was evaluated at 0, 15 during 30 days. After heating in plate to temperature of 75°C, the samples were submitted to color, taste and odor evaluation by 40 untrained tasters. The formulations F1, F2 and F4 presented lower concentrations of TBRS, whereas CF at day zero already showed very high values, indicating oxidation of the product. The samples showed good acceptance in the sensorial analysis.

Abstract: introduction: schizophrenia is a chronic severe psychiatric disease causing major psychological, social and working deficiencies. typical neuroleptics are widely used, but often produce incomplete responses, toxicity and adverse effects on motricity. clozapine was a significant development. it is effective in treating 30-61% of the psychotic symptoms with minimal adverse effects. methods: clinical experience of 56 patients with refractory schizophrenia under treatment at the psychiatry service of the hospital de clínicas de porto alegre, included in the program to supply clozapine free of charge, promoted by the department of health and the environment of the state of rio grande do sul. results: the mean score for the brief psychiatric rating scale (bprs) was initially 77.9 (sd=16.1) and, at the end, 41.1 (sd=16.2). two patients left the program and one was excluded because he developed agranulocytosis. there were 4 hospitalizations. discussion: despite its well-established efficacy and applicability, clozapine is not free from adverse effects: postural hypotension, tachycardia, cloudy vision, dry eyes, hypersalivation, constipation and sedation frequently occur. hematological alterations occur in 0.05-2.8%. conclusions: there was a significant and lasting improvement of the symptoms in the patients enrolled. diseases with a longer evolution had worse responses, probably related to neurochemical and neurophysiological damage. the importance of early treatment and the need for state intervention, offering psychosocial and financial support to optimize the treatment of this population should be highlighted.

Abstract: INTRODU O: A Esquizofrenia é transtorno psiquiátrico cr nico grave que acarreta importantes déficits psicológicos, sociais e vocacionais. Os antipsicóticos clássicos s o amplamente usados como tratamento, mas comumente produzem respostas incompletas, toxicidade e efeitos extrapiramidais. A clozapina foi importante aquisi o na terapia antipsicótica. Mostrou-se com eficácia legítima em 30-61% das psicoses e baixa incidência de efeitos adversos. MéTODOS: Relato da experiência clínica de 56 pacientes com Esquizofrenia Refratária incluídos no programa de fornecimento gratuito da clozapina pela Secretaria da Saúde e do Meio Ambiente do Estado do Rio Grande do Sul em convênio com o Hospital de Clínicas de Porto Alegre. RESULTADOS: A média dos escores da Brief Psychiatric Rating Scale (BPRS) foi inicialmente 77,9 (DP=16,1) e, após noventa e três meses de tratamento, 41,1 (DP=16,2). Dois pacientes abandonaram o programa e um foi excluído por agranulocitose. Houve quatro interna es. DISCUSS O: Apesar de sua comprovada eficácia e aplicabilidade, a clozapina n o está livre de efeitos adversos: hipotens o ortostática, taquicardia, vis o turva, xerostomia, hipersaliva o, constipa o e seda o s o comuns. As altera es hematológicas ocorrem em 0,05 a 2,8% dos casos. Ainda, o alto custo restringe seu uso. CONCLUS ES: Houve melhora significativa e mantida dos pacientes que participaram do programa. Doen as com maior tempo de evolu o obtiveram menor resposta, provavelmente relacionada a altera es neurofisiológicas e neuroquímicas. Vale ressaltar a importancia do início precoce do tratamento e a necessidade da participa o do Estado, no sentido de oferecer suporte psicossocial e financeiro para a otimiza o do tratamento desses pacientes.

Abstract: Este artigo é fruto do enício de uma pesquisa que busca investigar as representa es presentes acerca do trabalho para o adolescente cortador de cana. Realizando as primeiras aproxima es com esses adolescentes (protagonista de sua história) algumas quest es come am a ser levantadas. Como forma de suscitar o debate, pretende-se com estas reflex es contribuir para que a busca da compreens o desses fen menos presentes na regi o norte-noroeste do estado do RJ fa a parte dos estudos nos diferentes espa os, seja na academia, nos sindicatos ou na elabora o de políticas oúblicas. Este trabalho pretende sinalizar quest es presentes na rela o educa o/trabalho do adolescente cortador de cana. Primeiramente será situada a rela o educa o/trabalho frente aos desafios das novas tecnologias, num contexto no qual as rela es de produ o se encontram t o arcaicas, como é o caso específico do trabalho na lavoura de cana de a úcar. Educa o, neste caso, longe de significar um instrumento concreto de mobilidade social e de amplia o de expectativas. Em seguida será analisado as rela es que o adolescente cortador de cana com o trabalho e a escola. Finalmente, ser o apresentadas as propostas que vêm sendo construídos no debate teórico-prático.
Visually Impaired Individuals in Brazil and Portugal and Learning about Drugs through a Board Game  [PDF]

Abstract: Objective: To compare the learning of visually impaired individuals after the use of the educational game “Drugs: playing it clean”. Method: Quasi-experimental, comparative, before-after study. Results: The participants’ mean age in Brazil was lower than in Portugal; a significant difference in information acquisition was found between the pre and post-test for the low-complexity (Brazil p = 0.018 and Portugal p = 0.002), without a difference in the number of correct answers for the medium/high-complexity questions between the two countries (p = 0.655 and p = 0.0792); when comparing the number of correct answers before and after the game intervention, an increase was found in Brazil and Portugal, respectively (21.8% - 61.1%; 11.2% - 38.9%); a significant difference was found in the number of correct answers between the low and medium/high-complexity questions (p = 0.030). Conclusion: The educational game permits information access and can be used as a teaching-learning strategy.
Clinical Assessment of Treatment Outcomes Following Borago officinalis Extract Therapy in Patients Presenting with Cyclical Mastalgia  [PDF]

Abstract:
In order to evaluate the safety and efficacy of Borago officinalis (900 mg borage oil capsules) in the treatment of patients presenting with cyclic mastalgia, 91 subjects were included in the study. Efficacy assessments were performed based on data obtained prior to the start of treatment (Pretreatment), and after each menstrual cycle (Assessment 2—following 45 days of treatment; and Assessment 3—at the end of the 90-day treatment period). Primary efficacy measures considered the results of the Mastalgia Questionnaire, including a 100 mm visual analog pain scale assessing mean, most intense mastalgia severity, and impact on work, sleep, and sexual activity. Safety and tolerability measures included any changes in vital signs and physical exam in relation to pretreatment, any changes in clinical laboratory exams in relation to pretreatment, and the occurrence of adverse events after the first dose of study medication. The VAS scores of the mean mastalgia and most severe mastalgia both showed statistically significant (p < 0.0001) reductions from Pretreatment to Assessment 3. Mean mastalgia scores improved among 92.3% of the treated patients, while most severe mastalgia scores improved among 93.4% of patients. There were statistically significant improvements in the assessments of mastalgia impact on work (χ2 = 28.24; gl = 4; p < 0.0001), sleep (χ2 = 14.29; gl = 4; p = 0.0006), and sexual activity (χ2 = 16.11; gl = 4; p = 0.0029) during the treatment period. The results of this study indicate a significant improvement in the mastalgia of the treated patients together with an improvement in the quality of life parameters evaluated. In terms of safety, the tolerability of the treatment was good, with the presence of some adverse events, all of which had been previously described with use of the Borago officinalis extract. No serious side effects were reported, and the events that did occur were transitory. Based on the results of this study, we concluded that the Borago officinalis extract was safe and effective in the treatment of cyclic mastalgia among the treated patients.
