Genomic instability influences the transcriptome and proteome in endometrial cancer subtypes

Abstract: DNA image cytometry classified 25 endometrioid cancers to be either diploid (n = 16) or aneuploid (n = 9), and all uterine papillary serous cancers (UPSC) to be aneuploid (n = 8). All samples were subjected to comparative genomic hybridization and gene expression profiling. Identified genes were subjected to Ingenuity pathway analysis (IPA) and were correlated to protein expression changes.Comparative genomic hybridization revealed ploidy-associated specific, recurrent genomic imbalances. Gene expression analysis identified 54 genes between diploid and aneuploid endometrioid carcinomas, 39 genes between aneuploid endometrioid cancer and UPSC, and 76 genes between diploid endometrioid and aneuploid UPSC to be differentially expressed. Protein profiling identified AKR7A2 and ANXA2 to show translational alterations consistent with the transcriptional changes. The majority of differentially expressed genes and proteins belonged to identical molecular functions, foremost Cancer, Cell Death, and Cellular Assembly and Organization.We conclude that the grade of genomic instability rather than the histopathological subtype correlates with specific gene and protein expression changes. The identified genes and proteins might be useful as molecular targets for improved diagnostic and therapeutic intervention and merit prospective validation.Endometrial cancer is the most common malignancy of the female genital tract in the Western world and the fourth common cancer in women [1]. In general it is considered to have a favorable prognosis since it usually becomes symptomatic at an early tumor stage. Thus, about 70% of the affected women are detected at tumor stage I. At this stage, the mean survival of five years has been estimated to be 87%. However, one histopathological subtype, uterine papillary serous cancer (UPSC), presents with an aggressive clinical course characterized by early metastasis, reduced survival rates and inferior prognosis compared to endometrioid carcinomas [

Abstract:

The better understanding of the mechanism in which the immune system responds to the developing cancer provided the outcome in a new era in cancer immunotherapy. The tumor suppressive effect on the immune system is caused by negative T cell receptor signaling that abrogate immunity against the cancer cells. Novel monoclonal antibodies that target co-inhibitory receptors on T cells block the tumor induced inhibition of the immune system and enable the immune system to eradicate the tumors. The development of such antibodies started twenty years ago by the preparation of a monoclonal antibody termed BAT. A single administration of the antibody to tumor bearing mice resulted in striking anti tumor activity that was mediated by the lymphocytes. These studies provided a basis for the new era of cancer immunotherapy. The present review summarizes twenty years to the discovery of monoclonal antibodies harnessing the immune system to eradicate tumors.

Abstract: public higher education institutions (heis) are experiencing circumstantial changes due to increasing financial pressure, higher autonomy and diversified actors on an increasingly challenging "market". these changes have led to forms of "entrepreneurial behavior" in public heis, such as franchising arrangements, which have become prominent in the context of internationalization and the export of study programs. over the last decade, a growing number of franchising arrangements on an intrastate level can be observed between public universities as franchisors and private, non-state recognized heis as franchisees in the german he sector. the german situation is of special interest because the he system is traditionally state-run and emphasizes the university as a prestige and sound institution. students pay almost no tuition fees for a public university degree except those students enrolled in a franchise model, where the fees are about 9 times higher. special organizational structures like intermediary stock companies and either the lack of or not commonly interpreted regulatory instruments lead to critical discussions of these models. the most crucial assumptions are that financial motives dominate decision making rather than pedagogical considerations and that public services are exploited to mask the interests of individuals. although these presumptions cannot be rejected through the empirical results of case study analyses, the opportunities of such strategic alliances are not to be underestimated. with adequate external and internal quality assurance mechanisms, franchising models can be used to strengthen the market position of public universities through a higher number of students and an extended portfolio of products and services. additionally, he programs can be offered more flexibly in regions with minimal he infrastructure and / or with programs particularly designed for "non-traditional students".

Abstract: Bild gebende Verfahren in der Medizin haben Einsichten in den lebenden K rper verschafft, die vorher ohne invasive Eingriffe nicht m glich waren. Mit diesen Methoden hat eine Revolution in medizinischer Diagnose und biomedizinischer Forschung eingesetzt. Dabei treten andererseits problematische Effekte auf, insbesondere durch die hoch konstruktiven Eigenschaften der Bildproduktion, die komplizierte physikalische und physiologische Effekte ausnutzen. Die Bilderzeugung verwendet komplizierte Kombinationen von Technik und mathematischen und algorithmischen L sungen. Dazu werden Eigenschaften der menschlichen Kognition und des visuellen Systems für die Konstruktion von Unterscheidbarkeit ausgenutzt. Kein Wunder, dass die Visualisierungen des K rpers auch Effekte vorzeigen, die keine physiologischen Korrelate im menschlichen K rper haben. Dessen ungeachtet werden diese Bilder h ufig als eineindeutige Abbildungen des K rpers gesehen. Das hat Folgen, beispielsweise für ihren Gebrauch als Standardisierungsinstanzen. Diese führen zu Definitionen des normierten gesunden K rpers, versus von Krankheit oder Pathologien, von M nnlichkeit versus Weiblichkeit, von rassischen Unterscheidungen etc. Dazu werden Determinismen insinuiert, die in Widerspruch zur Plastizit t und Variabilit t des Gehirns stehen, die sowohl zeitlich wie r umlich, inter- und intra-individuell enorm gro sind.

Abstract: Die digitale Archivierung qualitativer Forschungsdaten bietet zusammen mit dem Fortschritt softwaregestützter Datenanalyse die Chance zur Wiederverwendung der Daten für eine forschungsnahe Ausbildung in der qualitativen Sozialforschung. Archivierte Daten k nnen als Demonstrations- und Analysematerialien genutzt werden und damit zu einem besseren Verst ndnis qualitativer Forschungsmethodik verhelfen. Für die forschungsnahe Methodenausbildung kann die Verwendung des Konzepts des "Blended Learning", das Pr senzlehre mit Elementen des E-Learning verknüpft, von Vorteil sein. Es gew hrleistet sowohl die fachliche Anleitung durch eine Lehrperson als auch eigenst ndige forschungspraktische Anwendungen durch die Lernenden. Der Artikel stellt das Konzept und Evaluationsergebnisse eines Lehr-Lernmodells vor, welches das Archiv für Lebenslaufforschung (ALLF) der Graduate School of Social Sciences (GSSS) an der Universit t Bremen für die Nutzung archivierter Interviewdaten in der qualitativen Methodenausbildung entwickelte. Das Pilotmodell, dessen inhaltlicher Schwerpunkt auf der Auswertungsmethode des problemzentrierten Interviews liegt, orientiert sich am Konzept des Blended Learning. Die Erprobung und Evaluation in der Lehr-Lernpraxis erfolgte im Rahmen einer Pilotveranstaltung des mobileCampus-Projekts im Studiengang Psychologie der Universit t Bremen. Abstract: Das Prostitutionsgesetz von 2002 hat das Gesch ft mit der Liebe in Deutschland erstmals aus der illegalen Schmuddelecke geholt. Formal ist die Prostitution damit ein Beruf neben anderen. Eine moralische Neubewertung der Sexarbeiterinnen“ ist damit nicht einhergegangen. Im vorliegenden Buch, das die wichtigsten Beitr ge der Tagung Frauenhandel und Prostitution in Europa“ versammelt, wird zum Paradigmenwechsel aufgerufen: Ja, es gibt sie, die Frauen (und M nner), die in der Prostitution einen alternativen, durchaus gangbaren Lebensweg sehen. Aber: Es gibt sie mindestens genauso h ufig, die wei en Sklavinnen“, Opfer des internationalen Menschenhandels, die zur Prostitution gezwungen werden.
