Cholesterol Depletion Disorganizes Oocyte Membrane Rafts Altering Mouse Fertilization

Abstract: Drastic membrane reorganization occurs when mammalian sperm binds to and fuses with the oocyte membrane. Two oocyte protein families are essential for fertilization, tetraspanins and glycosylphosphatidylinositol-anchored proteins. The firsts are associated to tetraspanin-enriched microdomains and the seconds to lipid rafts. Here we report membrane raft involvement in mouse fertilization assessed by cholesterol modulation using methyl-β-cyclodextrin. Cholesterol removal induced: (1) a decrease of the fertilization rate and index; and (2) a delay in the extrusion of the second polar body. Cholesterol repletion recovered the fertilization ability of cholesterol-depleted oocytes, indicating reversibility of these effects. In vivo time-lapse analyses using fluorescent cholesterol permitted to identify the time-point at which the probe is mainly located at the plasma membrane enabling the estimation of the extent of the cholesterol depletion. We confirmed that the mouse oocyte is rich in rafts according to the presence of the raft marker lipid, ganglioside GM1 on the membrane of living oocytes and we identified the coexistence of two types of microdomains, planar rafts and caveolae-like structures, by terms of two differential rafts markers, flotillin-2 and caveolin-1, respectively. Moreover, this is the first report that shows characteristic caveolae-like invaginations in the mouse oocyte identified by electron microscopy. Raft disruption by cholesterol depletion disturbed the subcellular localization of the signal molecule c-Src and the inhibition of Src kinase proteins prevented second polar body extrusion, consistent with a role of Src-related kinases in fertilization via signaling complexes. Our data highlight the functional importance of intact membrane rafts for mouse fertilization and its dependence on cholesterol.

Abstract: the thesis we intend to defend here, concerning newspapers in their role as representatives of the printed media and their function, at present, in brazil, is that the communication relationship maintained by this medium with its reader is basically determined by the social consumption relationship that the newspaper, being a commercial concern, maintains with readers in general. through a case study, we will be defending this line of thought, taking as our subject matter the newspaper segment that deals with issues of health and disease, to demonstrate that the consumption relationship, by extension, decidedly contaminates the contents and views on health and disease communicated by the newspaper.

Abstract: metropolitan areas have become the new spatial fix of globalised capitalism. however, their economic strength is not matched by their political strength because metropolitan areas remain politically weak. this article reflects upon the process of building metropolitan areas as political spaces. considering this process as a conflicting one because it challenges the power of existing players, it seeks to expose the general failure of metropolitan institution building - including most of the south european urban areas - focusing on two elements: on one hand, decentralisation as a process favouring other territorial scales than the metropolitan one, notably the regional and municipal levels; on the other hand, local democracy favouring the municipal and neighbourhood levels but forgetting the metropolitan scale.

Abstract: this article analyzes a billboard entitled "the sad picture of health", displayed in the city of s?o paulo in may 1993. based on various internal features and considering the surrounding social context, the billboard allows for two radically different interpretations. in the case of a such a "signed" billboard, such ambiguity implies a bad choice of the message intended to be conveyed by the sponsors. the author thus suggests that sponsors of such public announcements be aware of the ambiguity of their messages and forms of discourse.

Abstract: posters prepared by students in the northeast of brazil for the iii national schistosomiasis poster contest, sponsored by the health ministry's special schistosomiasis control program, produced four major types of posters expressing different attitudes toward the problem of schistosomiasis: posters showing a purely negative attitude (ex.; "do not bathe in contaminated rivers"); those showing a purely positive attitude "use the latrine"; and the mixed posters, a combination of the first, divided into two subtypes. the first of these subtypes projected a message about an undersirable behavior and an alternative to it; whereas, the second subtype presented the problem's elements but without relating them to alternatives. the last, that is, the fourth, type gave straight didatic or technical explanations of schistosomiasis. the proposals for confronting schistosomiasis which were projected through the official educational messages by the students' posters present a strategy of fundamental change and, in a short period of time, of primitive behavior. to avoid jolts of cultural shock, a more gradual strategy was suggested - one that would respect the cultural habits and values of the communities affected.

Abstract: the symbolic function drugs as part of the commercial exploitation of health/disease in brazil is analysed. in an immature society like the brazilian one, the drug-in so far as it performs the role of a product able to shorten the distance between the concrete and the abstract, between the desire and its fulfilment - needs, in order to accomplish this task, that health and disease of reduced to their purely organic features, and ignores those causal factors of a social and behavioral nature. what makes the symbolic function of drugs a rather serious public health factor is that, in many cases, it is effective. thus all health professionals with educational responsibilities should contribute, through their activities, to the creation of social maturity, in the attempt to prevent drugs being used in such a way as to inhibit action against the social and behavioral causes of diseases.

Abstract: the symbolic utilization of medicines as implying an abridgement of a long and multidimensional process which is necessary for achieving health status, socially and individually speaking, is discussed from a theoretical point of view. "instant health" is proposed as the desired result or as the meaning of this symbolic process. the politico-ideological dimensions of the problem as strictly relation to the idea of medicines as merchandise is discussed. the paper also considers the false opposition: prescribed versus nonprescribed medicines and offers a new one - symbolic versus nonsymbolic use of medicines. finally, a new field of research is proposed in public health - the supply and demand of symbolic goods which would satisfy the desire for "instant health".
