Isolation and characterization of atrazine-degrading Arthrobacter sp. AD26 and use of this strain in bioremediation of contaminated soil

Abstract: A bacterial strain (AD26) capable of utilizing atrazine as a sole nitrogen source for growth was isolated from an industrial wastewatersample by enrichment culture. The 16S rRNA gene sequencing identified AD26 as anArthrobacter sp. PCR assays indicated that AD26contained atrazine-degrading genes trzN and atzBC. The trzN gene of AD26 only differs from the trzN ofArthrobacter aurescens TC1by one base (A→T at 907) and one amino acid (Met→Leu at 303). The specific activity of trzN of AD26 in crude cell extract was0.28 U/mg, which was 1.2 times that of TC 1. This strain has shown faster growth and atrazine-degradation rates in atrazine-containingminimal media than two well characterized atrazine-degrading bacteria, Pseudomonas sp. ADP and Arthrobacter aurescens TC 1. Afterincubating for 48 h at 30℃, the OD600 of AD26 reached 2.6 compared with 1.33 of ADP. AD26 was capable of degrading 500 mg/Lof atrazine in minimal medium at 95% in 72 h, while the degradative rates by TC1 and ADP were only 90% and 86%, respectively. Abioremediation trial of contaminated soil has indicated that AD26 can degrade as high as 98% of atrazine contained in soil (300 mg/kg)after incubating for 20 d at 26℃, nominating this strain as a good candidate for use in bioremediation programs.

Abstract: As the Stock index futures with Cri 300 index for the subject matter launch, the research to stock options done by China’s financial market is gradually in-depth, which has great significance to the improvement of the financial markets. With the Black-Scholes option formula, this paper attempts to study the sensitivity of single stock’ call option named Industrial and Commercial Bank of China Limited to stock price changes, time changes and the situation that both of them occur. The simulation results were achieved based SAS, which not only has a very important practical significance to the launch of this kind financial derivative and the establish of a perfect pricing model of financial derivatives, but also can help financial market further promote economic growth.
Crayfish Carapace Micro-powder (CCM): A Novel and Efficient Adsorbent for Heavy Metal Ion Removal from Wastewater

Abstract: Crayfish carapace, a plentiful waste in China, was applied to remove divalent heavy metal ions—copper (Cu), cadmium (Cd), zinc (Zn), and lead (Pb)—from wastewater. The adsorption capacities of crayfish carapace micro-powder (CCM) for heavy metal ions were studied with adsorbent dosages ranging from 0.5–2.5 g/L and with initial metal concentrations ranging from 50–250 mg/L. CCM particle size, initial solution pH (from 2.5–6.5), temperature (from 25–65 °C) and calcium level (from 3.5–21.5%) were also varied in batch mode. The results indicated that the adsorption capacity increases with both decreasing particle size and increasing calcium level of the crayfish carapace. The kinetic studies indicated that the adsorption could be complete within 2 h, and that the data correlated with the pseudo-second-order model. CCM recorded maximum uptakes of 200, 217.39, 80, and 322.58 mg/g for Cu, Cd, Zn, and Pb, respectively. The adsorption capacities and removal efficiencies of CCM for metal ions were three-times higher than those of chitin and chitosan extracted from the CCM.

Abstract: 桉树枝瘿姬小蜂在广州地区一年发生4—5代,且世代重叠,其越冬期为11月中旬到翌年3月底,越冬虫态为大龄幼虫和蛹。在广州自然条件下,通过人工接种和自然接种相结合的方法,桉树枝瘿姬小蜂在2010年3月至2011年3月间共完成4个世代。利用生命表的方法分析桉树枝瘿姬小蜂连续世代可知,第1代的净增值率R0最大,而第2代的最小;第3代的内禀增长率rm和周限增长率λ最大,其世代平均历期T最小;第4代的rm和λ最小,其T最大。这说明第3代桉树枝瘿姬小蜂的种群增长能力最大,对桉树的危害最强。该代害虫的发生时间为7—9月,因此在防治上要注意该阶段的防控和监测。
Dexamethasone Ameliorates H2S-Induced Acute Lung Injury by Alleviating Matrix Metalloproteinase-2 and -9 Expression

Abstract: Acute lung injury (ALI) is one of the fatal outcomes after exposure to high levels of hydrogen sulfide (H2S), and the matrix metalloproteinases (MMPs) especially MMP-2 and MMP-9 are believed to be involved in the development of ALI by degrading the extracellular matrix (ECM) of blood-air barrier. However, the roles of MMP-2 and MMP-9 in H2S-induced ALI and the mechanisms of dexamethasone (DXM) in treating ALI in clinical practice are still largely unknown. The present work was aimed to investigate the roles of MMP-2 and MMP-9 in H2S-induced ALI and the protective effects of DXM. In our study, SD rats were exposed to H2S to establish the ALI model and in parallel, A549 cells were incubated with NaHS (a H2S donor) to establish cell model. The lung HE staining, immunohistochemisty, electron microscope assay and wet/dry ratio were used to identify the ALI induced by H2S, then the MMP-2 and MMP-9 expression in both rats and A549 cells were detected. Our results revealed that MMP-2 and MMP-9 were obviously increased in both mRNA and protein level after H2S exposure, and they could be inhibited by MMP inhibitor doxycycline (DOX) in rat model. Moreover, DXM significantly ameliorated the symptoms of H2S-induced ALI including alveolar edema, infiltration of inflammatory cells and the protein leakage in BAFL via up-regulating glucocorticoid receptor(GR) to mediate the suppression of MMP-2 and MMP-9. Furthermore, the protective effects of DXM in vivo and vitro study could be partially blocked by co-treated with GR antagonist mifepristone (MIF). Our results, taken together, demonstrated that MMP-2 and MMP-9 were involved in the development of H2S-induced ALI and DXM exerted protective effects by alleviating the expression of MMP-2 and MMP-9. Therefore, MMP-2 and MMP-9 might represent novel pharmacological targets for the treatment of H2S and other hazard gases induced ALI.

Abstract: We investigated the spin dynamics of two-dimensional electrons in (001) GaAs/AlGaAs heterostructure using the time resolved Kerr rotation technique under a transverse magnetic field. The in-plane spin lifetime is found to be anisotropic below 150k due to the interference of Rashba and Dresselhaus spin-orbit coupling and D'yakonov-Perel' spin relaxation. The ratio of in-plane spin lifetimes is measured directly as a function of temperature and pump power, showing that the electron density in 2DEG channel strongly affects the Rashba spin-orbit coupling.

Abstract: Bacille Calmette-Guérin (BCG) is an attenuated strain of Mycobacterium bovis and is the only available vaccine against tuberculosis (TB). Since 1974, BCG vaccination has been included in the World Health Organization (WHO) Expanded Program on Immunization. It is estimated that more than 3 billion individuals have been immunized with BCG and over 100 million doses of BCG are administered annually. Multiple studies have confirmed that BCG is generally safe and can protect children against disseminated disease, including tuberculosis meningitis [1,2]. BCG also provides cross-protection against leprosy [3]. However, the success of BCG against pulmonary TB in adults is still debated, since randomized clinical trials have reported protection efficacy ranging from 0–80% [4,5]. Several hypotheses for the variation in observed efficacy have been proposed [6-9].One explanation concerns the heterogeneity of the BCG strains [6]. The original BCG was derived from a virulent strain of M. bovis isolated from a cow. From 1908 through 1921, this isolate was subjected to 230 passages on glycerinated potato bile medium, which generated an attenuated strain termed BCG [10]. Distribution and widespread use of BCG started around 1924 and was accompanied by changes in the manufacturing process in production facilities. For instance, while BCG in Sweden was transferred without interruption from bile potato to bile potato medium in accordance with Calmette's original practice [11], BCG production in Denmark involved alternating rounds of growth on potato bile medium and Sauton broth until 1949 when it was grown exclusively in Sauton medium [12]. Prior to the establishment of seed stocks in the 1960s, BCG was passaged continuously, and the changes in media and transfer schedules contributed to the "in vitro evolution" of BCG [6]. Abstract: 【目的】对卡介苗(Bacillus Calmette-Guerin,BCG)美国株(BCG Tice)进行基因组补缺口(补洞)工作,以得到它的基因组完整序列。【方法】首先对BCG Tice进行高通量测序,使用SOAPdenovo软件对得到的数据进行拼接。由于在高通量测序的过程中基因组某些区域测序覆盖度低,测序质量差会使测序结果经拼接后形成众多的重叠群(contig),相邻的位置关系确定的contig形成一个scaffold,contig之间未测到的区域为缺口序列(gap),在contig末端设计引物进行PCR扩增,得到连接相邻contig的PCR产物,对PCR产物进行测序。通过优化PCR引物设计策略,尝试不同的聚合酶进行聚合反应,调整PCR反应条件并结合PCR产物构建克隆测序等方法,补齐contig之间的缺口序列。【结果】完成了BCG Tice的全基因组测序,得到了它的基因组完整序列,序列已提交到美国国立生物技术信息中心(NCBI)的GenBank数据库。【结论】BCG属于高GC含量的革兰氏阳性细菌,其基因组GC含量高达65.65%。本文以BCG Tice基因组补洞为例,对高GC含量基因组补缺口过程中遇到的问题与采取的策略给予概述,望给相关高GC含量基因组的物种全基因组测序补缺口工作提供一些借鉴。

Abstract: 摘要:【目的】寡发酵链球菌(Streptococcus oligofermentans)是从无龋人的口腔中分离到的一株链球菌,好氧条件下产生、同时也耐受高浓度(4.4 mmol/L)的过氧化氢。本研究探讨dpr基因对寡发酵链球菌抗过氧化氢的贡献。【方法】克隆和表达寡发酵链球菌dpr基因,分析Dpr蛋白的功能;构建寡发酵链球菌的dpr基因突变株,比较野生株和突变株对不同浓度过氧化氢的耐受程度;并将寡发酵链球菌dpr基因克隆到对过氧化氢耐受力低的变形链球菌中,分析其对变形链球菌过氧化氢耐受能力的影响。【结果】
