Phenotypic Observation of Mandibular First Molar in Caveolin-1 Gene Knockout Mice.

Abstract: 摘要 目的:观察Caveolin-1基因敲除小鼠(knockout,KO)下颌第一磨牙牙胚组织学形态及牙齿表型,为深入研究Caveolin-1在牙齿发育中的作用提供组织学基础。方法:经聚合酶链式反应(polymerase chain reaction,PCR)鉴定基因型后,HE染色观察不同发育阶段的KO小鼠及同源野生型(wildtype,WT)小鼠的下颌第一磨牙牙胚组织学形态;显微CT观察其牙体硬组织结构。结果:胚胎14.5 d,WT鼠牙胚发育进入帽状期,但KO鼠牙胚仍处于蕾状期,成釉器未见明显分层及原发性釉结形成。胚胎16.5 d,KO鼠成釉器出现细胞分层,但成釉器体积较小,颈环处上皮芽向下方间充质中深入较浅。胚胎18.5 d, KO鼠成釉细胞和成牙本质细胞的细胞极化不明显,且成牙本质细胞缺乏典型柱状形态。出生后1个月,下颌第一磨牙显微CT及三维重建结果显示:KO鼠与WT鼠牙体解剖形态未见明显差异。结论:Caveolin-1基因表达缺失可导致小鼠下颌第一磨牙牙胚发育蕾状期-帽状期过渡出现暂时性滞后,但对牙齿后期发育无明显影响
Cell Therapy: A Safe and Efficacious Therapeutic Treatment for Alzheimer’s Disease in APP+PS1 Mice

Abstract: Previously, our lab was the first to report the use of antigen-sensitized dendritic cells as a vaccine against Alzheimer’s disease (AD). In preparation of this vaccine, we sensitized the isolated dendritic cells ex vivo with Aβ peptide, and administered these sensitized dendritic cells as a therapeutic agent. This form of cell therapy has had success in preventing and/or slowing the rate of cognitive decline when administered prior to the appearance of Aβ plaques in PDAPP mice, but has not been tested in 2×Tg models. Herein, we test the efficacy and safety of this vaccine in halting and reversing Alzheimer’s pathology in 9-month-old APP+PS1 mice. The results showed that administration of this vaccine elicits a long-lasting antibody titer, which correlated well with a reduction of Aβ burden upon histological analysis. Cognitive function in transgenic responders to the vaccine was rescued to levels similar to those found in non-transgenic mice, indicating that the vaccine is capable of providing therapeutic benefit in APP+PS1 mice when administered after the onset of AD pathology. The vaccine also shows indications of circumventing past safety problems observed in AD immunotherapy, as Th1 pro-inflammatory cytokines were not elevated after long-term vaccine administration. Moreover, microhemorrhaging and T-cell infiltration into the brain are not observed in any of the treated subjects. All in all, this vaccine has many advantages over contemporary vaccines against Alzheimer’s disease, and may lead to a viable treatment for the disease in the future.
Impact of Industrial Structure on LUCC in Daqing City of Heilongjiang Province

Abstract: Daqing City,as a typical resource-based city that developed by petroleum,meadow and cultivated land resource,not only is a region that human beings and earth interact intensely,but also an area featured by land use change sharply.Land use and land cover change is the most remarkable symbol of the earth's surface system.It influences the structure and function of landscape ecosystem directly or indirectly,and impacts some landscape processes of the earth's surface.There are so many factors affecting land use and land cover change,including nature and human factors.Many researches in the driving force of land use and land cover change have been done in recent years,but few studies have focused on human factors in a resources-based city.Based on the land use data of Daqing City,urban area and four counties in five periods that gained by remote sense images interpreting,this paper analyzed the correlation between the production of main industries and land use in the periods and calculated the coupling coefficient of industry proportion and land use composite of Daqing City urban area and four counties from 1992 to 2001.At first,land uses were divided into six major types: cultivated land,forestland,grassland,water body,residential site and built-up land,wasted land;Secondly,the land use change data of Daqing City by remote sensing interpreting were obtained;At last,using the land use change data and statistical data,the correlation between industry and land use was analyzed and the coupling coefficient of industry proportion and land use composite were calculated.the factors that impact land use change and industry structure of Daqing City were analyzed according to the results.The results showed that the correlation between GDP of industries and LUCC in Daqing City issignificant,the coupling coefficient of industry proportion and land use of urban area in Daqing is the largest.The land use change in Daqing City is mainly affected by the GDP of industry,especially the industry involved petroleum.
Cloning, expression and preparation of polyclonal antibody for human Dcp2 gene

Abstract: The degradation of eukaryotic mRNAs plays important roles in the modulation of gene expression,and quality control of mRNA biogenesis.Dcp1/Dcp2 is a major component of mRNA decay factors.However,the more detailed mechanisms of Dcp1/Dcp2 in mRNA degradation are unknown.In this study,our object is to amplify Dcp2 gene from human brain cDNA library,and then to prepare rabbit-anti-human Dcp2 polyclonal antibodies.Our results showed that anti-Dcp2 rabbit polyclonal antibody was obtained.The antibody could detect the endogenous Dcp2 expression and its cell localization by both Western Blotting and immunohistochemistry,and Dcp2 protein also could be enriched by immunoprecipitation with this antibody.Therefore,this work provides a useful tool for further functional study of Dcp2 gene.
Josephson junction on one edge of a two dimensional topological insulator affected by magnetic impurity

Abstract: Current-phase relation in a Josephson junction formed by putting two s-wave superconductors on the same edge of a two dimensional topological insulator is investigated. We consider the case that the junction length is finite and magnetic impurity exists. The similarity and difference with conventional Josephson junction is discussed. The current is calculated in the semiconductor picture. Both the $2\pi$- and $4\pi$-period current-phase relations ($I_{2\pi}(\phi), I_{4\pi}(\phi)$) are studied. There is a sharp jump at $\phi=\pi$ and $\phi=2\pi$ for $I_{2\pi}$ and $I_{4\pi}$ respectively in the clean junction. For $I_{2\pi}$, the sharp jump is robust against impurity strength and distribution. However for $I_{4\pi}$, the impurity makes the jump at $\phi=2\pi$ smooth. The critical (maximum) current of $I_{2\pi}$ is given and we find it will be increased by asymmetrical distribution of impurity.
Large-scale estuarine engineering and estuarine habitat fragmentation of water body in the Yangtze River Estuary

Abstract: Estuarine habitat, one of basic components of estuarine ecosystems, is a typical marine habitat. Human activities such as large-scale engineering construction in the estuarine comprehensive regulation have an important impact on estuarine ecosystems, resulting in estuarine habitat fragmentation. In this paper, based on the survey and sampling data of 7 habitat indices carried out during May to June, 2002 in the Yangtze River Estuary, we preliminarily studied and analyzed the impact of the first-stage project of Deep-waterway Regulation on habitat fragmentation of estuarine water body. The estuarine water habitats can be subdivided into 3, 2, 3, 3, 6, 4 and 5 patch types for salinity, DO, COM_(Mn), SiO_3-Si, Chl a, phytoplankton and zooplankton, respectively. The seascape diversity(H), dominance index(D)and fragmentation index(C)of 7 indices in water body differed significantly. H of Chl a is the highest with 0.58 and that of DO the lowest with 0.04; D of COD is the highest with 0.30 and that of phytoplankton the lowest with 0.08; C of Chl a is the highest with 0.30 and that of DO is the lowest with 0.05. Except for salinity, the number of patches, H, D and C of DO and COD in water body of the Yangtze River Estuary in May are higher than that in May 1997. Our results showed that the large-scale estuarine engineering has had obvious impacts on the habitat fragmentation of water body in the Yangtze River Estuary, resulting in locally small-scale changes in oceanic water mass. The results from this study suggest that the first-stage project of Deep-waterway Regulation has caused habitat fragmentation and significant impact on seascape diversity of estuarine water body. The patch patterns of estuarine water body are related to local hydrodynamic condition caused by the first-stage project, which deserve for further research.
Study of denial of service attack & improved deterministic packet marking scheme based on IPv6

Abstract: This paper studied the DoS attacks under IPv6, and according to the new features of IPv6 protocol, proposed the ADPM-v6 scheme based on improved MAC-authentication. By using a new IPv6 feature hop-by-hop options and the improved MAC authentication method, the scheme could quickly locate the source of attacker. The scheme could prevent compromised router which could forge the marking effectively. Simultaneous, analysis of a real attack environment verifying IPv6 packet size distribution, made the algorithm have a strong practicality and more effective. The implementation and evaluation shows that the ADPM-v6 scheme can greatly reduce the reconstruction time and reduce the amount of reconstruction computing and false positive.

Bipolaronic blockade effect in quantum dots with negative charging energy

Abstract: We investigate single-electron transport through quantum dots with negative charging energy induced by a polaronic energy shift. For weak dot-lead tunnel couplings, we demonstrate a bipolaronic blockade effect at low biases which suppresses the oscillating linear conductance, while the conductance resonances under large biases are enhanced. Novel conductance plateau develops when the coupling asymmetry is introduced, with its height and width tuned by the coupling strength and external magnetic field. It is further shown that the amplitude ratio of magnetic-split conductance peaks changes from 3 to 1for increasing coupling asymmetry. Though we demonstrate all these transport phenomena in the low-order single-electron tunneling regime, they are already strikingly different from the usual Coulomb blockade physics and are easy to observe experimentally.
Metallothionein diversity and its ecological service functions of invertebrates: an overview

Abstract: Metallothioneins(MTs) are low molecular mass, cysteine rich metal binding polypeptides, which have been found widely distributed in invertebrates since 1975 and established a central role in the regulation of the essential metals, zinc and copper, and the detoxification of nonessential metals, cadmium and mercury. So far, more than 60 invertebrate MTs have been reported each with two and more than two MT isoforms and variants respectively in most major invertebrate groups including nematodes, annelids, echinoderm, mollusks, arthropods, pogonophora, and crustaceans. It was reported that MTs differed significantly among invertebrate species with plentiful information of biodiversity and many kinds of ecological functions, including the following three aspects: diversity of binding metals to MTs, diversity of MT isoforms and variants, and diversity of MTs ecological service functions. Up to now, however, studies on the diversity of MTs are not enough, especially on MTs ecological service functions, so that few MTs ecological functions have been found and studied in natural populations of invertebrates. For this reason, it is important to indicate the ecological service functions on the basis of the diversity of invertebrate MTs in terms of theory and practice. The diversity of invertebrate MTs are overviewed and analyzed in this paper, and their ecological service functions are also discussed in three aspects: MTs function in detoxification and regulation of heavy metals, MTs as biomarkers in environmental monitoring, purification function of MTs in environmental heavy metal pollution and their roles in the environmental pollution management.
