Abstract: Background and Aim: In this study, it was aimed to examine the cytotoxic effect of temozolomide (TMZ) treatment, on MCF-7 and SKBR3 cell lines, to study the methylation levels of MGMT gene expression and gene promoter region. Methods: The MTT test was performed to determine the effective dose of TMZ. The time-dependent cell survival test was performed after the IC50 value was found. Western blotting was performed to determine MGMT gene expression levels. High Resolution Melting (HRM) technique was used to determine the methylation levels of MGMT gene promoter region. Results: TMZ has been shown to have a high cytotoxic effect on SKBR3 cell line and low cytotoxicity on MCF-7. When MGMT expression levels before and after TMZ treatment were observed by western blotting, the gene expression levels of TMZ treatment were shown to decrease in both cell lines. It was observed that MGMT gene promoter region was hypermethylated in two cell lines, and that the application of TMZ further increased the methylation levels in the promoter region. Abstract: This paper describes the Arhavi Highway Tunnel which has two tubes, its geometrical properties, finite element model, and the nonlinear earthquake behaviour under a huge ground motion considering soil-structure interaction. The Arhavi Highway Tunnel is one of the tallest tunnels constructed in the Black Sea region of Turkey as part of the Coast Road Project. The tunnel has two tubes and each of them is about 1000 m tall. In the study, the modal analyses of the tunnel considering soil-structure interaction are performed and natural frequencies and mode shapes are obtained. Then, nonlinear transient analysis of the tunnel using Drucker-Prager criteria is performed applying acceleration components of 1992 Erzincan, Turkey earthquake's ground motion. In the time history analyses, Rayleigh damping coefficients are calculated using main natural frequency obtained from modal analysis. Element matrices are computed using the Gauss numerical integration technique. The Newmark method is used in the solution of the equation of motion. Because too much memory for the analyses is required, the first 7.5 s of the ground motions, which is the most effective duration, is taken into account in calculations. The displacement and stress results are observed to be the allowable level of the concrete material.
The effect of material properties on the seismic performance of Arch Dams

Abstract: The paper investigates the effect of material properties on the seismic performance of arch dam-reservoir-foundation interaction systems based on the Lagrangian approach using demand-capacity ratios. Type-5 arch dam is selected as a numerical application. The linear time history analyses of the arch dam-reservoir-foundation interaction system are carried out for different material properties. The foundation is taken into account as massless; behaviour of the reservoir is assumed to be linearly elastic, inviscid and irrotational. The north-south component of the Erzincan earthquake in 1992 is chosen as a ground motion. Dynamic equations of motions obtained from 3-D finite element modelling of the coupled system are solved by using the Newmark integration algorithm. The damage levels of the coupled system for the different material properties are demonstrated by using demand-capacity ratios and cumulative inelastic durations. The time histories and maximum values of the displacements and principal stresses, and performance curves, are obtained from linear analyses. It is clearly seen from the study that the different material properties affect the seismic behaviour of the dam.

Materials and methods: Fifty eyes of 48 patients who presented between July 2007 and December 2009 with the diagnosis of stage 2, 3, or 4 macular holes according to Gass Classification who had undergone PPV and ILM peeling were included in this study. Pre- and postoperative macular examinations were assessed with spectral-domain optical coherence tomography. 23 G sutureless PPV and ILM peeling with BB was performed on all patients. Results: The mean age of patients was 63.34 ± 9.6 years. Stage 2 macular hole was determined in 17 eyes (34%), stage 3 in 24 eyes (48%), and stage 4 in 9 eyes (18%). The mean follow-up time was 13.6 ± 1.09 months. Anatomic closure was detected in 46/50 eyes (92%), whereas, in four cases, macular hole persisted and a second operation was not required due to subretinal fluid drainage. At follow-up after 2 months, persistant macular hole was detected in one case and it was closed with reoperation. At 12 months, an increase in visual acuity in 41 eyes was observed, while it remained at the same level in six eyes. In three eyes visual acuity decreased. There was a postoperative statistically significant increase in visual acuity in stage 2 and 3 cases (P < 0.05), however, no increase in visual acuity in stage 4 cases was observed. Conclusion: PPV and ILM peeling in stage 2, 3, and 4 macular hole cases provide successful anatomic outcomes, however, in delayed cases, due to photoreceptor loss, it has no effect on functional recovery. BB, used for clarity of ILM, may be beneficial due to its low retinal toxicity.

All children were under 2 years of age. The main outcome was successful probing. Successful probing was defined as a complete resolution of signs and symptoms. Results: The mean age of the patients was 16.4 ± 4.1 months (range 8–24 months) in group A and 16.8 ± 4.2 months (range 7–24 months) in group B. No canalicular or nasolacrimal sac pathologies were diagnosed during probing. The success rate was 76.2% (32/42) in group A and 91.4% (117/128) in group B. The difference was statistically significant (P < 0.01). Conclusion: Manually curved probes can be used efficiently during probing and increase the success rates.

Abstract: Objective: The aim of the study is determination of myocardial performance index (MPI/Tei index) using pulsed (PD) and tissue Doppler (TD) techniques to show cardiac response in newborns with hypoxic-ischemic encephalopathy (HIE) and healthy newborns and eventually evaluation of the differences between these two techniques. Methods: The study is a prospective observational study. Twenty term newborns diagnosed as perinatal asphyxia during postnatal 24 hours due to the defined criteria and fifty healthy term neonates as control group were included the study. Hypoxic group was divided into two groups with Sarnat stages, Sarnat Stage 1 and 2-3. MPIs (Tei indexes) were calculated with PD and TD echocardiographic techniques in all groups after the 24 hours of birth and one year later. The statistical differences between same techniques were calculated with Kruskal-Wallis test and Z score was used to compare the superiority of two techniques. Results: The MPI values calculated by PD (0.41±0.04, 0.51±0.02) and TD (0.59±0.04, 0.51±0.02) during the first day of life in Sarnat Stage 2-3 in both ventricles were significantly higher than the control group (p<0.01, p<0.02, p<0.03). While the Z score, calculated for MPI measured by PD and TD methods, were found similar in both ventricles in Sarnat Stage 1 and control groups, it was significantly different in other groups of Sarnat stages. Conclusion: The degree of cardiac response in neonates with HIE is associated with the severity of hypoxia. MPI values are not different from the controls in newborns received mild hypoxia while they are higher in the patients who were received moderate or severe hypoxia. Any advantage could not be found between two techniques according to the measurement values, but higher variability in the value of MPI, measured by TD method, calculated from moderate and severe hypoxia group was detected.
