Optima of Trypsin-Catalyzed Hydrolysis and Its Inhibition Determined by SDS-PAGE  [PDF]

Abstract: SDS-PAGE was applied to determine trypsin activity and inhibition. After the hydrolysis by trypsin to substrate bovine serum albulnin (BSA) under different temperatures and pH, the hydrolysis degree of BSA was conducted using SDS-PAGE. From the quantitative analysis to the electrophoresis bands of BSA and its hydrolysis products in SDS-PAGE pattern, the change of trypsin activity was determined, and then the optimum temperature at 40°C and the optimum pH at pH 8.5 - 8.7 for trypsin activity were obtained. All the target bonds in BSA molecule could be hydrolyzed at the same time by trypsin. The inhibition was due to the binding of inhibitor to trypsin, which made it impossible for trypsin to touch the substrate protein. SDS-PAGE was demonstrated to be also an effect method for assaying the characteristics of trypsin activity and its inhibition.
An Effective Vaccine Strategy of Translating Apoptotic Tumor Cells into Cancer Vaccine

Abstract: Cancer vaccine has been regarded as the most attractive treatment for the patients. Our previous work showed that cartilage polysaccharide of short-chain (CPS) could induce apoptosis of L1210 cell lines through mitochondria apoptosis pathway and mice treated with CPS didn’t develop tumors after L1210 cell inoculation when compared to control mice without treatment. In present work, we developed a vaccine based on previous experiments and results showed that four in ten mice vaccinated didn’t show proliferation of ascite tumors while the mice in the control group didn’t show inhibition of tumor growth. We further identified the antibody with immunoflourescence and western blot from the serum of immunized mice which can recognize a protein with molecular weight of 35 kDa. Finally, we used mass spectrometry and identified the target recognized by the antiserum annexin A2, which is involved in cell proliferation, differentiation and apoptosis processes and is tightly associated with cancer progression and chemoresistance.

Research on Influencing Factors of Welfare Level about Left-behind Children in China Rural Areas

Abstract: Based on the survey data of 360 left-behind children at junior school in Xinyang city Henan province in China, the welfare of left-behind children was analyzed by using Amartya.Sen ability method theory and fuzzy mathematics. The results showed that left-behind children's material welfare of the economy increased, but their social mental welfare level declined when their parents went to work at other cities. As a result, the level of total welfare utility is low. At last, in order to increase the general welfare of left-behind children, policies and suggestions were provided according to the analysis.
Inactivation of Baroduric Bacteria Isolated by High Hydrostatic Pressure from Pickled Cowpea

Abstract: In this study, Pickled Cowpea, a typical lactic acid fermented vegetable in Sichuan, China, was used as samples to study both species and inactivation of baroduric bacteria isolated by HHP treatment under different pressure levels and different pressure holding time. 16S rDNA gene sequence, amplified using genomic DNA of 4 baroduric bacteria from Pickled Cowpea as templates, were sequenced and then were identified based on the sequence similarity and homology analysis, as B. licheniformis, B. subtilis, B. sonorensis and B. pumilus. The pressure resistance of the 4 strains are compared under pressure from 300 to 500 MPa with holding time from 3 to 25 min. B. pumilus which has higher pressure resistance can be selected as indicator bacteria for applying HHP treatment to Pickle production.
A Box-behnken Design for Characterizing Chinese Truffles (Tuber indicum) Aroma by HS-SPME-GC-MS

Abstract: The aim of the present investigation is to fully characterize the aroma of Chinese truffles (Tuber indicum) by headspace solid phase microextraction (HS-SPME). To develop an objective method to extract aroma compounds, four different fibers were studied and a Box-Behnken design (BBD) was applied. From the statistical analysis of the experimental result, it was able to determine that the most important factor was the extraction temperature and the optimum extraction conditions were as follows: extraction time 20.6 min, extraction temperature 52.4 oC and equilibrium time 6.8 min, By using gas chromatography mass spectrometry (GC-MS) analysis under the optimal conditions, it identified 24 compounds, three of which were reported for the first time in the Chinese truffle: 2-methylpropanal, 2,3-butanedione, 2-nonanone. And we found that the highest content compound was dimethyl sulfide, followed by 3-methylbutanal, 2-methylbutanal, 2-butanol and 1-pentanol, 1-octen-3-ol, all of those compounds were previously described as characteristic aroma of truffle.
Computational identification of microRNAs and their targets in wheat (Triticum aestivum L.)

Abstract: microRNAs (miRNAs) are a class of endogenous, non-coding, short (~21 nt) RNAs directly involved in regulating gene expression at the post-transcriptional level. Previous reports have noted that plant miRNAs in the plant kingdom are highly conserved, which provides the foundation for identification of conserved miRNAs in other plant species through homology alignment. Conserved miRNAs in wheat are identified using EST (Expressed Sequence Tags) and GSS analysis. All previously known miRNAs in other plant species were blasted against wheat EST and GSS sequences to select novel miRNAs in wheat by a series of filtering criteria. From a total of 37 conserved miRNAs belonging to 18 miRNA families 10 conserved miRNAs comprising 4 families were reported in wheat. MiR395 is found to be a special family, because three members belonging to the same miR395 family are clustered together, similar to animal miRNAs. MiRNA targets are transcription factors involved in wheat growth and development, metabolism,and stress responses.

HMGB1 Accelerates Alveolar Epithelial Repair via an IL-1β- and αvβ6 Integrin-dependent Activation of TGF-β1

Abstract: High mobility group box 1 (HMGB1) protein is a danger-signaling molecule, known to activate an inflammatory response via TLR4 and RAGE. HMGB1 can be either actively secreted or passively released from damaged alveolar epithelial cells. Previous studies have shown that IL-1β, a critical mediator acute lung injury in humans that is activated by HMGB1, enhances alveolar epithelial repair, although the mechanisms are not fully understood. Herein, we tested the hypothesis that HMGB1 released by wounded alveolar epithelial cells would increase primary rat and human alveolar type II cell monolayer wound repair via an IL-1β-dependent activation of TGF-β1. HMGB1 induced in primary cultures of rat alveolar epithelial cells results in the release of IL-1β that caused the activation of TGF-β1 via a p38 MAPK-, RhoA- and αvβ6 integrin-dependent mechanism. Furthermore, active TGF-β1 accelerated the wound closure of primary rat epithelial cell monolayers via a PI3 kinase α-dependent mechanism. In conclusion, this study demonstrates that HMGB1 released by wounded epithelial cell monolayers, accelerates wound closure in the distal lung epithelium via the IL-1β-mediated αvβ6-dependent activation of TGF-β1, and thus could play an important role in the resolution of acute lung injury by promoting repair of the injured alveolar epithelium.
Software Industry Cluster be Disagreement on Theory and Practice  [PDF]

Abstract: In the view of traditional industry cluster theory, it is easy to copy the software industry cluster pattern, or it is easy to copy another Silicon Valley, due to low reliability of the resources and the guidance factors of locations in software industry. But it is much more difficult to copy a Silicon Valley mode practically than imaginatively and the difficulties of bringing up and supporting high-tech initiatives is more than theoretic anticipation. In China, the software companies have just gathered together geographically and therefore no initiative center can be formed. All these above signify that software industry cluster is distinct from the traditional industry clusters, but the cognition of the reasons of software industry cluster is not clear yet. Furthermore, reasonable explanations of the bewilderment in the economical practice of software industry cluster are urgently needed.
