CCL2 upregulation triggers hypoxic preconditioning-induced protection from stroke

Abstract: Adult male SW/ND4, CCL2-null, and wild-type mice were used in these studies. Cortical CCL2/CCR2 message, protein, and cell-type specific immunoreactivity were determined following HPC (4 h, 8% O2) or room air control (21% O2) from 6 h through 2 weeks following HPC. Circulating leukocyte subsets were determined by multi-parameter flow cytometry in na?ve mice and 12 h after HPC. CCL2-null and wild-type mice were exposed to HPC 2 days prior to tMCAo, with immunoneutralization of CCL2 during HPC achieved by a monoclonal CCL2 antibody.Cortical CCL2 mRNA and protein expression peaked at 12 h after HPC (both p < 0.01), predominantly in cortical neurons, and returned to baseline by 2 days. A delayed cerebral endothelial CCL2 message expression (p < 0.05) occurred 2 days after HPC. The levels of circulating monocytes (p < 0.0001), T lymphocytes (p < 0.0001), and granulocytes were decreased 12 h after HPC, and those of B lymphocytes were increased (p < 0.0001), but the magnitude of these respective changes did not differ between wild-type and CCL2-null mice. HPC did decrease the number of circulating CCR2+ monocytes (p < 0.0001) in a CCL2-dependent manner, but immunohistochemical analyses at this 12 h timepoint indicated that this leukocyte subpopulation did not move into the CNS. While HPC reduced infarct volumes by 27% (p < 0.01) in wild-type mice, CCL2-null mice subjected to tMCAo were not protected by HPC. Moreover, administration of a CCL2 immunoneutralizing antibody prior to HPC completely blocked (p < 0.0001 vs. HPC-treated mice) the development of ischemic tolerance.The early expression of CCL2 in neurons, the delayed expression of CCL2 in cerebral endothelial cells, and CCL2-mediated actions on circulating CCR2+ monocytes, appear to be required to establish ischemic tolerance to focal stroke in response to HPC, and thus represent a novel role for this chemokine in endogenous neurovascular protection.Preconditioning occurs when an organism, tissue, or cell is exposed

Abstract: the purpose of this article is to introduce elements that allow building an interface between the academic research and the programs of basic education for youngsters and adults. it discusses contributions to these programs that can be found in the results of qualitative research studies. to this end, results of a five-year long project on teacher education are used, which aim was that of analyzing the interaction between teacher and student in youngster and adult literacy classes. the research project was conducted in natural contexts with the purpose of understanding a given social reality, and not of establishing general laws. therefore, the credibility of its results was built through the observation of multiple contexts, and the gathering of data was made through various methods, from the perspective of several participants observed during a prolonged period of time. this empirical basis was used to evaluate recommendations contained in the report commissioned by unesco to the international literacy institute for presentation at the world forum on education held in 2000 in dakar. this report proposed that the continuous attendance of students to basic education programs is one of the great challenges of the new millennium. with respect to the problem of adult evasion from courses and programs, the article discusses the motivation and accessibility factors, pointed out in official documents as relevant factors to the success or failure of the programs.

Abstract: the rapid development of nanotecnology has allowed finding new optical properties of nanosystems, which give rise to investigation of light-matter interactions at nanoscale. in this work we present a systematic study of surface-localized plasmons coupling in systems of two metallic nanoparticles following shape, size and distance between them. we observed the enormous enhancement of the near field, which is strongly dependent on the plasmon creation and the charge distribution at the surface. we also find out the plasmon frequency dependence on the above parameters and discuss the different physical concepts underlying the maximum near field and maximum frequency.

Abstract: the aim of this paper is to present some contributions from the research perspective known as new literacy studies - the study of reading and writing practices in social life - for teacher education programs. we discuss first the ethical aspects of that approach, related to the qualitative methods used, that protect the researcher from the ever present danger of reproducing teacher stereotypes. next, we analyze the specific knowledge demands being made to literacy teachers in official documents, and finally, based upon our critical analysis of those demands, we explore an alternative teacher education strategy centered arround the participation in social practices.

Abstract: Este trabalho tem por objetivo discutir algumas contribui es dos Estudos do Letramento — o estudo das práticas relacionadas com a escrita em toda atividade da vida social — para a reflex o sobre a forma o do professor de língua materna. Primeiramente, discutiremos aspectos éticos dessa abordagem, relacionados ao método qualitativo da pesquisa, que resguardam o pesquisador do perigo, sempre constante, de seus resultados de pesquisa serem utilizados para a reprodu o de críticas e estereótipos sobre o docente. Em seguida, analisaremos, em documentos que prescrevem, avaliam ou informam as a es do professor, as exigências em rela o aos saberes específicos dos alfabetizadores e professores de língua materna. Finalmente, com base na análise crítica dessas exigências, exploraremos uma alternativa de forma o baseada na participa o na prática social.

Abstract: 0 conhecimento de palavras esta fortemente relacionado à capacidade de compreender o texto escrito. Embora a natureza da rela o seja obscura e as explica es polêmicas (vide, por exemplo, as correla es em testes de aptid o verbal e inteligência, Carroll, 1972, Freebody e Anderson, 1979), vários estudos indicam inequivocamente a existência de tal rela o (Davis,1968, Carroll, 1972). No nível puramente intuitivo, o leitor parece pressupor tal rela c, a julgar pela expectativa bastante comum do aprendiz de segunda língua de que as suas dificuldades a leitura dessa língua ser o minimizadas se ele souber o significado de todas as palavras do texto. Esta vis o unilateral da compreens o do texto é apenas sintomática do nível de frustra o que um texto povoado de palavras desconhecidas cause no leitor.

Abstract: Este trabalho argumenta que o discurso escolar difere substantivamente do discurso letrado, que supostamente seria o modelo de linguagem da escola, uma vez que a fun o desta seria a de introduzir o aluno na cultura letrada. Entretanto, esse discurso escolar também difere significativamente da linguagem da crian a.
