A Novel, Non-canonical Splice Variant of the Ikaros Gene Is Aberrantly Expressed in B-cell Lymphoproliferative Disorders

Abstract: The Ikaros gene encodes a Krüppel-like zinc-finger transcription factor involved in hematopoiesis regulation. Ikaros has been established as one of the most clinically relevant tumor suppressors in several hematological malignancies. In fact, expression of dominant negative Ikaros isoforms is associated with adult B-cell acute lymphoblastic leukemia, myelodysplastic syndrome, acute myeloid leukemia and adult and juvenile chronic myeloid leukemia. Here, we report the isolation of a novel, non-canonical Ikaros splice variant, called Ikaros 11 (Ik11). Ik11 is structurally related to known dominant negative Ikaros isoforms, due to the lack of a functional DNA-binding domain. Interestingly, Ik11 is the first Ikaros splice variant missing the transcriptional activation domain. Indeed, we demonstrated that Ik11 works as a dominant negative protein, being able to dimerize with Ikaros DNA-binding isoforms and inhibit their functions, at least in part by retaining them in the cytoplasm. Notably, we demonstrated that Ik11 is the first dominant negative Ikaros isoform to be aberrantly expressed in B-cell lymphoproliferative disorders, such as chronic lymphocytic leukemia. Aberrant expression of Ik11 interferes with both proliferation and apoptotic pathways, providing a mechanism for Ik11 involvement in tumor pathogenesis. Thus, Ik11 could represent a novel marker for B-cell lymphoproliferative disorders.

Abstract: The history of the land admits of two complementary kinds of inquiry: an analysis of the processes affecting the structures on it (its settlements, productions, etc.), and that of the inhabitants' own perceptions of the places they inhabit. The recent historiography on the land of Romuald, of Peter Damian and of their monastic congregations has developed along these very two directions, the history of structures and that of cultural representations, with combinations of both. Here we will concentrate on cultural aspects. La storia del territorio è passibile di due tipi complementari di indagine: l'analisi dei processi di mutazione delle strutture insistenti nello spazio (sistemi insediativi e di produzione, ecc.), e l'analisi della percezione che dei luoghi hanno coloro che li abitano. La storiografia degli ultimi anni riguardante il territorio di Romualdo, di Pier Damiani e delle loro congregazioni monastiche, si è sviluppata proprio seguendo i due percorsi costituiti dalla storia delle strutture e dalla storia delle rappresentazioni culturali, proponendo delle sintesi di questo binomio interpretativo. In questo articolo vengono proposte alcune riflessioni, concentrando l'attenzione prevalentemente sugli aspetti culturali della storia del territorio.

Abstract: This paper introduces Clement III (Wibert of Ravenna) in the context of the general phenomenon of the antipopes, a vast and fundamentally medieval subject. The theme can be approached in two substantially different ways: from the well-established, official position, which condemns the antipopes as schismatics and subverters of the divine order; or from the perspective of an observer who attempts to examine the phenomenon from the inside. This study opts for the latter vantage point, as do the three papers that it introduces. In them, the antipopes take shape as historical personages who believed in their own legitimacy as popes, who often had large followings, and who received their mark of infamy—that is, the title of antipope—because they were defeated by their opponents. With the first of the two methods (the official, well-established one), history is interpreted in reverse, giving events after-the-fact justifications. With the second analytical strategy, the interpreter instead views history in the historical present and tries to comprehend how events unfolded within the dynamics of the myriad possibilities, changes, and inversions of course that life presents. In this sense, the essays in this collection look not at anti-popes but rather at other-popes reflected in the mirrors of their adversaries—adversaries who won their respective struggles and were thus able to transmit their own visions of events as the sole vehicles of truth. In the same spirit, these essays consider not antipopes but rather individuals who, like the pipe in Magritte’s painting, come down to us not in their authentic dimension but rather through the filters of representation. Questo contributo introduce Clemente III (Guiberto di Ravenna) nel contesto del fenomeno generale degli antipapi, un tema ampio ed essenzialmente medievale. L’argomento può essere affrontato in due modi decisamente diversi: da una posizione ufficiale e ben consolidata, che condanna gli antipapi in quanto scismatici o sovvertitori dell’ordine divino; oppure dalla posizione di un osservatore che tenta di esaminare il fenomeno dall’interno. Questo studio opta per la seconda soluzione, così come i tre articoli che seguono. In questa prospettiva gli antipapi prendono forma come personaggi storici che credevano nella propria legittimità di papi, che spesso avevano largo seguito di aderenti, e che hanno ricevuto il loro marchio di infamia – cioè il titolo di antipapa – perché erano stati sconfitti dai loro oppositori. Changes in Some Blood Haematochemical Parameters of Maltese Goats During Lactation

Abstract: The course of some haematochemical parameters sensitive to variations due to lactation in Maltese goats during the second and the fourth lactation has been investigated. Total cholesterol, HDL-cholesterol, LDL-cholesterol, triglycerides, -hydroxybutyrate, total protein, urea, calcium, phosphorus values were determined in Maltese goats during lactation in two different groups. The purpose was to determine if there are sustained modifications in the levels of these parameters due to lactation between two groups and within groups. Twelve goats were divided into two groups, each consisting of 6 subjects: group A aged 14 months at the second lactation and group B aged 22 months at the fourth lactation. Through a simultaneous monitoring equipment and a blood collection by means of a jugular venipuncture, some parameters were assessed for each subject on the following experimental conditions: at 30th, 60th, 90th, 120th and 150th day after parturition. The Bonferroni s test was applied to determine statistical significances within each group and between groups, respectively. With regards to lactation stage, statistically significant differences were observed for all the studied parameters within each group whereas no statistically significant differences were observed between group A and B. These results suggest that lactation curve is responsible for the changes in concentration of the parameters under investigation.
Ripening-induced changes in microbial groups of artisanal Sicilian goats’ milk cheese

Abstract: Changes in the microbial flora of “Caprino dei Nebrodi”, a raw goat’s milk cheese produced in Sicily, were studied during ripening. From 2 batches of cheese, 4 samples were taken at day 0, 2, 15, and 30 of ripening. Also, samples of curd and milk used in the manufacturing process were analyzed. By the end of the ripening process (day 30), high log10 cfu/g were found for Lactobacilli (7.20), Lattococci (7.10), and Enterococci (7.00), whereas counts of Enterobacteriaceae (3.91), Escherichia coli (3.30), and Staphylococcus (3.89) were found to be lower. The study provides useful information on the microbiological properties of “Caprino dei Nebrodi” cheese, and the results obtained suggest that in order to increase the quality of this artisanal product, it is necessary to improve the sanitary conditions of milking and cheese-making. The study was intended as a preliminary step towards the isolation and identification of bacterial species found in this type of goat’s cheese.
Volume-based Semantic Labeling with Signed Distance Functions

Abstract: Research works on the two topics of Semantic Segmentation and SLAM (Simultaneous Localization and Mapping) have been following separate tracks. Here, we link them quite tightly by delineating a category label fusion technique that allows for embedding semantic information into the dense map created by a volume-based SLAM algorithm such as KinectFusion. Accordingly, our approach is the first to provide a semantically labeled dense reconstruction of the environment from a stream of RGB-D images. We validate our proposal using a publicly available semantically annotated RGB-D dataset and a) employing ground truth labels, b) corrupting such annotations with synthetic noise, c) deploying a state of the art semantic segmentation algorithm based on Convolutional Neural Networks.
Study of different herbicide molecules for the control of durum wheat weed

Abstract: In order to enhance the chances to rotate the herbicide molecules, the effectiveness of a new molecule, pinoxaden, was tested, comparing it with other herbicides used in wheat weed control. The trial was carried out comparing the following herbicide mixtures: 1) no weed control treatment; 2) Tribenuron Methyl (TM); 3) Clodinafop (C); 4) Tribenuron Methyl + Clodinafop (TM+C); 5) Pinoxaden + clodinafop + propargile (PCP); 6) Pinoxaden + clodinafop + propargile + Triasulfuron (PCP+T); 7) Pinoxaden + clodinafop + propargile + absolute Ioxinil and Mecoprop (PCP+IM). The new PCP+T herbicides mixture didn’t differ statistically from the traditional TMC treatment in terms of effectiveness, but the agronomic result of the new mixture was totally satisfactory, even taking into account that the marketing of this mixture is not aimed to compete with other existing herbicides but to widen the chance to rotate active principles in time and space, in order to control the onset of resistance phenomena.

Development and validation of predictive MoSaiCo (Modello Statistico Combinato) on emergency admissions: can it also identify patients at high risk of frailty?

Abstract: the prospective historical cohort study develops and validates a method of identifying patients at high risk of emergency admission to hospital in the population of the province of ravenna (no. = 296 641). the main outcome measure is: emergency hospital admission analyzed using multivariate logistic regression (mosaico - modello statistico combinato). to validate the findings, the coefficients for 30 most powerful variables found on half of the population (derivation data set) were then applied to the rest of the population (validation data set). the key predicting factors included some demographic variables, social variables, clinical variables and use of health/social services. discriminatory power and validation both reached good results. risk score increases when variables indicating the individual vulnerability raise. the predictive frailty risk resulting from mosaico allows to stratify the population, to organize care services, to provide a practical planning tool in the field of case management and management of frail patients.
