Stability of Citrus tristeza virus protective isolates in field conditions

Abstract: the objective of this work was to monitor the maintenance of citrus tristeza virus (ctv) protective isolates stability in selected clones of 'pêra' sweet orange (citrus sinensis), preimmunized or naturally infected by the virus, after successive clonal propagations. the work was carried out in field conditions in the north of paraná state, brazil. coat protein gene (cpg) analysis of 33 isolates collected from 16 clones of 'pêra' sweet orange was performed using single strand conformational polymorphism (sscp). initially, the isolates were characterized by symptoms of stem pitting observed in clones. then viral genome was extracted and used as template for the amplification of cpg by reverse transcription polimerase chain reaction (rtpcr). rtpcr products electrophoretic profiles were analyzed using the jaccard coefficient and the upgma method. the majority of the clones had weak to moderate stem pitting symptoms and its ctv isolates showed alterations in the sscp profiles. however, the stability of the protective complex has been maintained, except for isolates from two analised clones. low genetic variability was observed within the isolates during the studied years.

Abstract: a cross-protection program citrus tristeza disease, caused by citrus tristeza virus (ctv) has been developed in the parana state, to obtain pera orange mother plants. thus, 20 plants were selected in the orange commercial fields of the north and northwest region. comparisons between mild and severe recognized isolates were made. the results showed mild-to-moderate stem pitting symptoms and the great majority of the isolates grouped up with the mild control isolate. these results suggest that the plants selected were infected by ctv mild haplotypes showing that they have mother plant potential.

Abstract: objective: to investigate the effects of early specialization (defined here as concentrated dedication to a single specialty during undergraduate medical school) on the general skills of newly graduated doctors. sample and methods: applicants for admission to medical residency programs at the university of s?o paulo who had recently finished the undergraduate medical course at the same institution (n=163), grouped by specialty of choice. the study focused on applicants' performance in admissions exams and their undergraduate grade point average. intergroup analysis - applicants for a specific career versus other applicants. intra-group analysis - performance in each respective field of knowledge versus mean performance in the other areas studied. correlation coefficients - between each admissions exam, and between undergraduate gpa and exams. significance level: p<0.05. results and conclusions: there was a tendency towards early specialization in obstetrics-gynecology, psychiatry, and pediatrics, with a loss of performance in areas not directly related to the specialty choice. applicants for the residency program in internal medicine showed overall higher grades on the residency admissions exam and undergraduate gpa, possibly due to a broader and more comprehensive interest in medicine.

Abstract: this study aimed to investigate a situation of teaching and learning of concepts related to the theme "floating bodies"students in the early years of elementary school. the activities were conducted in a class of eleven students from the age of about nine years old, students at a private school in uberlandia/mg. starting with activities developed in the classroom, based on a teaching methodology that focused on problem solving, materials handling and interaction among students with diferent skill levels. the results discuss aspects of the learning of this concept in children of this age group.

Abstract: objectives: medical education encompasses globally diverse context and conditions. the brazilian scenario seemed a natural environment to study the influence of medical education programs and internship duration on the entrance exam for medical residency. this investigation evaluates some methods used during the entrance exam for medical residency as a means to make a distinction between candidates with longer clerkships. methods: candidates selected for a residency program performed a multiple-choice (mc), an open question (oq) and osce-like tests, an interview and a curriculum analysis for participation in scientific meetings, papers published and voluntary activities. groups were compared for gender, year of graduation, tests and osce scores. results: participants were distributed into two groups based on clerkship duration: 2 years or less than 2 years. there was no difference for the mct score among groups or any of the activities from interview and curriculum analysis. the 2 years clerkship group showed significantly higher oq (p=0.009) and osce-like affective (p=0.025) and knowledge (p=0.002) scores. conclusion: the osce test identified some aspects related to competence acquisition and assessed basic skills and attitudes essential to the supervised practice of medicine during residency. osce discriminated aspects not perceived by the sole use of knowledge tests.

Abstract: objectives: to estimate the prevalence of cardiovascular risk factors in the adult population of luzerna, in the state of santa catarina. methods: a cross-sectional study with adults of both genders aged 20 to 59 years (n = 411). the prevalence of hypertension, diabetes, dyslipidemia, obesity, increased waist circumference, and smoking was estimated. study variables were checked for frequency distribution, and a chi-square test for association was performed. results: the response rate was 85.9%. the following prevalences were found: hypertension: 14.7%; diabetes: 2.3%; dyslipidemia: 18.7%; obesity: 15.6%; increased waist circumference: 24.1%; and smoking: 15.6%. a total of 52.4% of the subjects had none of the risk factors; 22.4% had one risk factor, and 13.6%, 6.8%, and 4.9% had two, three, and four or more associated risk factors, respectively. conclusion: the study sample showed low prevalence of hypertension and diabetes and less clustering of risk factors in the same individual, as compared with other data reported in the literature.

Abstract: Dengue is an important emerging viruses, posing a threat to one-third of the global human population. In 2002, the introduction of DENV-3 in the state of Bahia produced massive epidemic (about 35,000 cases detected) and the first cases of dengue hemorrhagic fever. To understand the nature of the virus circulating at Bahia, E/NS1 sequence was determined for 31 DENV viruses isolated in Bahia during the 2006 and 2007 transmission season, from patients presenting with different degrees of severity. The carboxi-terminal region of the E gene (220 nt) of 31 viruses, isolated from dengue patients with clinical diagnosis of dengue infection were used to determine the genetic variability of dengue 2 (DENV-2) and dengue 3 (DENV-3). Sequence data were used in phylogenetic comparisons with global samples of DENV-2 and DENV-3. DENV-2 sample grouped in the South East Indian genotype, while DENV-3 samples were grouped within Indian genotype. This study is the first report on Bahia isolates during two transmission seasons. Our data confirms reports from other parts of Brazil and different countries showing the DENV-3 (geno-type III) strains circulating in the Americas are closely related, and cluster within the genotype that has been associated with DHF epidemics in different conti-nents.

Abstract: objective: to evaluate the presence and severity of symptoms of anxiety and depression in individuals with asthma or chronic obstructive pulmonary disease. methods: in order to evaluate symptoms of anxiety and depression, specific instruments of quantification (the state-trait anxiety inventory and the beck depression inventory, respectively) were administered to patients at an outpatient clinic for the treatment of asthma and chronic obstructive pulmonary disease. the population comprised 189 randomly and prospectively selected patients that were divided into three study groups (each with a different therapeutic objective): 40 patients with controlled asthma, 100 patients with uncontrolled asthma, and 49 patients with chronic obstructive pulmonary disease. included among the variables studied, as part of the methodology, were symptoms of anxiety and depression. the data obtained were compared taking into consideration demographic and functional aspects, as well as the severity of the symptoms of anxiety and depression. results: among the asthma patients, the prevalence of moderate or severe anxiety was significantly higher than that observed among those with chronic obstructive pulmonary disease (p < 0.001). the uncontrolled asthma group presented significantly higher rates of depressive symptoms than did the controlled asthma group (p < 0.05). conclusion: the frequency of symptoms of anxiety and depression is greater among asthma patients than among patients with chronic obstructive pulmonary disease, which can make clinical control difficult.

Abstract: the burnout index of 119 elementary public education teachers was assessed. the cbp-r, teacher burnout questionnaire - (brown, garrosa and gonzález, 2000) and a socio-demographic questionnaire were used as data collection instruments. the results showed that 70.13 per cent of the participants presented symptoms of burnout, 85 per cent felt threatened in the classroom, 44 per cent spent more than 60 working hours a week and 70 per cent of them were less than 51 years old. significant differences were found in the following factors: feeling threatened in the classroom, journey to work and age, when related to burnout syndrome. based on the research results, the authors suggested alternatives aimed at humanizing the teacher workplace.
