Cinética do crescimento de Borrelia burgdorferi (Spirochaetaceae) em diferentes meios de cultivo

Abstract: the cinetic of growth of borrelia burgdorferi was studied during a 3-month period, using the following 8 culture media: (1) rabbit serum bsk, (2) swine serum bsk, (3) swine serum bsk+5 fluorouracil, (4) pmr, (5) ctb, (6) dubos, (7) brucella broth and (8) bhi. all media were prepared aseptically and were maintained in culture tubes of 10 ml capacity. for each medium, the inoculum was standardized to contain initially 102 spirochetes for each 0.1 ml of culture. the growth was monitorized by counting the total number of spirochetes in 0.1ml of medium in a dark field microscope, using a 10x30 mm cover slip. for the first 12 days, counting was done each 24 hours, and afterwards once a week during 14 weeks. there occurred growth of b. burgdorferi in all tested media, with the best performance of three of them: bsk with rabbit serum, bsk swine serum + 5 fluorouracil, and ctb medium. growth of b. burgdorferi was seen from the 4th week on, reaching its maximum within 8-12 weeks, depleting the culture medium after this time. cystic forms of b. burgdorferi were observed with all tested media.

Abstract: borrelioses are infectous diseases caused by spirochaetes of the genus borrelia. they are born mainly through ticks at animals and/or human beings. in this review are shown and discussed five groups of diseases determined by borrelia, general characteristics of the spirochaetes, aspects related to transmission by arthropods, biological and pathological aspects of the diseases in domestic and wild animals, lyme disease as an important zoonosis, the association of borrelia with other hematozoa agents, the diagnostic methods and the comparative epidemiology with data obtained from brazil and other countries. the borrelioses have pathological, clinical and epidemiological characteristics which vary according to physiographic regions due to the existence of different species, genospecies and strains of borrelia, of arthropod vectors, vector-agent relationship and of different ecocystems.

Abstract: As borrelioses s o enfermidades infecciosas determinadas por espiroquetas do gênero Borrelia, agentes transmissíveis, principalmente, por carrapatos aos animais e/ou ao homem. Nesta revis o s o apresentadas e discutidas as enfermidades determinadas por borrélias, bem como as características gerais das espiroquetas, os aspectos relacionados a transmiss o por artrópodes, as enfermidades nos animais domésticos e silvestres, quanto aos aspectos biológicos e patológicos, a doen a de Lyme como principal zoonose do grupo, a associa o de borrélia com outros agentes hematozoários e os métodos diagnósticos e a epidemiologia comparativa entre dados obtidos no Brasil com os de outros países. Estas borrelioses possuem características patológicas, clínicas e epidemiológicas variadas de acordo à regi o fisiográfica, devido à existência de distintas espécies, genoespécies e cepas; estes aspectos variam ainda em fun o dos artrópodes vetores, da intera o vetor-patógeno e dos ecossistemas distintos.
Esquizogonia exoeritrocítica plasmática en Plasmodium (Novyella) juxtanucleare (Apicomplexa: plasmodiidae)

Abstract: a research about plasmatic exoerythrocytic meronts of the plasmodium juxtanucleare in crossbred fowls from seropédica municipality, rio de janeiro state, brazil, was made. the hemoscopical exam of fowls with high erythrocytic parasitaemia (>10%) showed free exoerythrocytic meronts with an average size of 7.492 ± 2.308 x 5.926 ± 2.048 μm; containig about 15 ± 16.92 merozoites. the meronts showed to be pleomorphic characteristics from spherical to oval with merozoites of basophilic nucleus. they also showed torn meronts with free merozoites. all the observations reported and discussed about this subject make us conclude that p. juxtanucleare studied strain has a paraerythrocytic merogony. it suggests that p. juxtanucleare is an evolutionary plasmodia species intermediate between the plasmodia of reptiles and the plasmodia of mammals.

Abstract: parasitism of leukocytes and thrombocytes of gallus gallus l. by plasmodium (novyella) juxtanucleare (apicomplexa: plasmodiidae) a research about parasitism of the plasmodium juxtanucleare in crossbred fowls from seropédica municipality, rio de janeiro state, brazil, were done. blood smear were done, and stained by giemsa stain diluted in sorensen buffer ph6.8. the hemoscopical exam of fowls with high parasitaemia (>10%) showed trophozoite and meront forms in the cytoplasm of leukocytic cells; and trophozoite forms in the cytoplasm of the thrombocytic cells. these observation do conclude that p. juxtanucleare strain from seropédica produce phanerozoic meronts. this report constitutes the first finding of p. juxtanucleare forms in the leukocytes.

Abstract: spirochaetes transmitted by ticks are microorganisms of worldwide distribution, which infect wild, domestic animals and human beings. a total of 300 equine sera from four municipalities: ananideua (58), belém (61), castanhal (131), and santa izabel do pará (50), were evaluated for borrelia burgdorferi by an elisa test. there were no significant differences (p<0.05) among municipalities, breed, sex or husbandry. a total of 80 (26.7%) horses were b. burgdorferi positive with titles of 1:800, 72 (90%) horses, 1:1.600, 6 (7.5%) horses, and 1:3.200, 2 (2.5%) horses. the results were similar to those in the usa, where related frequencies ranged from 7 to 75% in asymptomatic seropositive horses. the presence of anti-b.burgdorferi homologous antibodies in horses from four municipalities in the metropolitan mesorregion of belém suggests the possibility of occurrence of human cases in the region.

Abstract: serumprevalence of antibodies against anaplasma marginale was evaluated by the indirect enzyme-linked immunosorbent assay (elisa). sera samples from 532 bovines from nine municipalities in the "norte fluminense" mesoregion, rio de janeiro state, were analysed. the results showed that 485 (91.16%) were positive, of which 55.45% had a titre of 1:500, 22.18% of 1:1000, 6.77% of 1:2000, 3.01% of 1:4000, 1.50% of 1:8000, 0.94% of 1:16000, 0.75% of 1:32000, 0.56% of 1:64000, and 8.84% were negative. the prevalence analysis was done within three age groups: 1 to 3 years (n= 110), 3 to 6 years (n= 241) and > 6 years (n= 181), of which 91.82%, 92.95% and 88.95% were positive, respectively. according to the breed, 91.22% of beef cattle (n= 444) and 90.91% of dairy cattle (n= 88), were positive. regarding the sex, 91.35% of the females (n= 497) and 88.57% of the males (n= 35), were positive. there were no significant differences between the age groups, breeding types and the sexes (p > 0.05); and between the prevalence in the municipalities (p > 0.000). the seroprevalence showed that the region has to be considered enzootically stable, and the infection by a. marginale in this area is high and homogeneous.

Abstract: Se realizó un estudio de los esquizontes exoeritrocíticos plasmáticos de Plasmodium juxtanucleare en gallinas sin raza definida, provenientes del município de Seropédica, estado de Rio de Janeiro, Brasil. Al examen sanguíneo se pudo observar en aves con alto índice de parasitémia eritrocítica (> 10%) formas esquizogónicas exoeritrocíticas libres, con un tama o promedio de 7,492 ± 2,308 X 5,926 ± 2,048 μm; conteniendo una média de 15 ± 16,92 merozoítas. Estos esquizontes se presentaron con características pleomórficas de redondeado a oval, con merozoítos de núcleo basofílico. Se observó también, formas de esquizontes rotos con merozoítas libres. Estas y otras observaciones, reportadas y discutidas, hacen inferir que la cepa de P. juxtanucleare estudiada, realiza un ciclo paraeritrocítico. Se sugiere que P. juxtanucleare es una espécie de plasmodio, evolutivamente, intermediário entre los plasmodios de reptiles y los de mamíferos . Abstract: RESUMEN Se realizó una investigación del parasitismo de Plasmodium juxtanucleare en gallinas sin raza definida, provenientes de criaderos rústicos en el município de Seropédica, estado de Rio de Janeiro, Brasil. Se realizaron frotis sanguineos periféricos, los cuales fueron coloreados con Giemsa diluído en tampon sorensen pH6,8. En el exámen hemoscópico se puede observar en aves con alto índice de parasitemia (>10%) formas parasitarias de trofozoítas y esquizontes en el citoplasma de células de la linea leucocítica y trofozoítas en células de la linea trombocítica. Las observaciones en el presente estudio hacen inferir que la cepa de P. juxtanucleare que ocurre en Seropédica realiza esquizogonia fanerozóica. Soroprevalência de Babesia bovis em bovinos na mesorregi?o Norte Fluminense

Abstract: serumprevalence of antibodies against babesia bovis were studied by the indirect enzyme-linked immunosorbent assay (elisa). sera samples from 532 bovines of nine municipalities in the "norte fluminense" mesoregion, state of rio de janeiro, were analysed. the results showed that 484 (90.98%) were positive by indirect elisa, of which were: 20.30% with a titre of 1:500, 35.72% of 1:1000, 20.87% of 1:2000, 7.33% of 1:4000, 2.82% of 1:8000, 2.63% of 1:16000, 0.56% of 1:32000, 0.75% of 1:64000, and 9.02% were negative. the prevalence analysis was done within three age groups: 1 to 3 years (n= 110), 3 to 6 years (n= 241) and > 6 years (n= 181), of which 98.18%, 90.87% and 86.74% were positive, respectively. there were significant differences (p < 0,05) between the 1 to 3 year-group and other age groups. the young animals are less susceptible to the disease than older animals. according to the breed, 91.44% of beef cattle (n= 444) and 88.64% of dairy cattle (n= 88) were positive. regarding the sex, 90.54% of the females (n= 497) and 97.14% of the males (n= 35) were positive. there were no significant differences between breeding types and the sexes (p > 0.05). there were significant differences (p < 0.0001) between the prevalence in the different municipalities. the infection by b. bovis in this mesoregion is heterogeneous, although the seroprevalence showed that the region has to be considered enzootically stable. the epidemiological situation requires immunization only of animals coming from other areas.
