Effect of MDM2 SNP309 and p53 codon 72 polymorphisms on lung cancer risk and survival among non-smoking Chinese women in Singapore

Abstract: We therefore examined the role of the SNPs in the p53 pathway (p53 codon 72 and MDM2 SNP309) on lung cancer risk and prognosis of a life-time non-smoking female Chinese population, in a hospital-based case-control study of 123 cases and 159 age-matched controls, by PCR analysis.Our findings reveal that the risk of lung cancer among individuals with the MDM2 SNP309 TT genotype was 2.1 (95% CI 1.01-4.36) relative to the GG genotype, contrary to initial expectations that the GG genotype with elevated MDM2 levels will increase cancer risk. Those who had this genotype in combination with the p53 Pro allele had a risk of 2.5 (95% CI 1.2-5.0). There was however no effect of either polymorphism on age at diagnosis of lung cancer or on overall survival.The results thus demonstrate that the MDM2 SNP309 TT rather than the GG genotype is associated with increased risk of lung cancer in this population, suggesting that other mechanisms independent of increased MDM2 levels can influence cancer susceptibility.The TP53 tumour suppressor pathway plays a critical role in cell cycle regulation and apoptosis in many cancers, including lung carcinomas [1], and variation in the genes that regulate this pathway may exert an important influence on tumour development, and hence, cancer risk. Recent interest has focused on the murine double minute-2 protein (MDM2), a nuclear phospoprotein that inhibits p53 activity by promoting its degradation [2]. A single nucleotide polymorphism (SNP309) in the MDM2 promoter has been found to influence transcription of this gene via a greater affinity for the SP1 transcription factor, and hence, individuals with the GG genotype have higher MDM2 levels leading to attenuation of the p53 pathway [3,4]. This has been especially so in the case of females, due to the involvement of the MDM2 SNP in the estrogen receptor signaling pathway [3].Several epidemiologic studies have evaluated this association with varying results. The management of desmoids in patients with familial adenomatous polyposis (FAP)

Abstract: Desmoids are rare in the general population but occurs in between 10 to 20% of patients with familial adenomatous polyposis (FAP). This risk is about 852 times the risk for the population at large. Desmoids are benign neoplasms that are capable of infiltrating locally with a high risk of recurrence (25-65%) even after extirpating surgery. Desmoids in FAP may occur extra-abdominally, or within the abdominal wall or most commonly intra-abdominally within the mesentery or retroperitoneal Desmoids are a major problem in patients with FAP. Mortality from desmoids is high in such patients and ranges from 18 to 31%, compared to peri-ampullary carcinomas at about 22% and cancer in the retained rectum at only about 8%. Simple drug treatment with tamoxifen or NSAIDS like sulindac should be used as first line treatment as it carries a response in 30-50% of patients. Surgery should be reserved for extra-abdominal tumours alone and only when needed. Surgery for intra-abdominal desmoids should really only be attempted for intestinal obstruction or ureteric obstruction. Dacarbazine-Doxorubicin chemotherapy may have dramatic response in some cases. Genetic transfer may unlock this disease in future and may give patients with FAP and severe desmoids hope for the future.
Variant Toll-Like Receptor4 (Asp299Gly and Thr399Ile Alleles) and Toll-Like Receptor2 (Arg753Gln and Arg677Trp Alleles) in Colorectal Cancer

Abstract: The innate immune system recognizes the presence of bacterial products through the expression of a family of membrane receptors known as Toll-like receptors (TLRs). Polymorphisms in TLRs have been shown to be associated with increased susceptibility to diseases such as inflammatory bowel disease. The aim of this study was to determine whether there was a correlation between polymorphisms of TLR4 (Asp299Gly; Thr399Ile) and TLR2 (Arg677Trp; Arg753Gln) genes and risk of colorectal cancer. DNA from 60 colorectal carcinoma patients from 3 major races in Malaysia (22 Malays, 20 Chinese and 18 Indians) and blood from 50 apparently healthy individuals were evaluated. Control group were matched to study group by race and age. The polymorphisms were determined by Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP). Genotyping results showed two out of sixty tumor specimens (3.3%) harbored both variant TLR4 Asp299Gly and Thr399Ile alleles. In contrast, DNA isolated from blood cells of 50 apparently healthy individuals harbored wild type TLR4. In the case of TLR2 Arg753Gln genotyping, all of the fifty normal and 60 tumors were of the wild type genotype. TLR2 Arg677Trp genotyping showed a heterozygous pattern in all samples. However, this may not be a true polymorphism of the TLR2 gene as it is likely due to a variation of a duplicated (pseudogene) region. There was only a low incidence (2/60; 3.3%) of TLR4 polymorphism at the Asp299Gly and Thr399Ile alleles in colorectal cancer patients. All normal and tumor samples harbored the wild type TLR2 Arg753 allele. Our study suggests that variant TLR4 (Asp299Gly and Thr399Ile alleles) as well as TLR2 (Arg753Gln allele) are not associated with risk of colorectal cancer.
Dynamic Programming for Estimating Acceptance Probability of Credit Card Products  [PDF]

Abstract: Banks have many variants of a product which they can offer to their customers. For example, a credit card can have different interest rates. So determining which variants of a product to offer to the new customers and having some indication on acceptance probability will aid with the profit optimisation for the banks. In this paper, the authors look at a model for maximisation of the profit looking at the past information via implementation of the dynamic programming model with elements of Bayesian updating. Numerical results are presented of multiple variants of a credit card product with the model providing the best offer for the maximum profit and acceptance probability. The product chosen is a credit card with different interest rates.

Savoirs mondains, savoirs savants : les femmes et leurs cabinets de curiosités au siècle des Lumières

Abstract: Dans la France des Lumières, la culture de la curiosité est un phénomène de mode mais surtout un jeu social et intellectuel. La présente étude entend retracer l’histoire d’une trentaine de cabinets féminins de curiosité tenus à cette époque. Des femmes fortunées de l’aristocratie et de la bourgeoisie parisienne et provinciale ont alors constitué sous l’emprise de leur libido sciendi des cabinets d’alchimie, de minéralogie, de physique chimie, d’histoire naturelle et d’anatomie naturelle et artificielle. Ces cabinets obéissent à une typologie particulière. Il en existe deux catégories : les cabinets d’amateurs, constitués pour la parade et le spectacle des visiteurs et fonctionnant comme de véritables écoles de plaisirs intellectuels et éducatifs ; les cabinets à finalité scientifique et didactique, formés par des savantes expérimentées qui se livrent dans leurs laboratoires à des recherches personnelles et expérimentales au nom des progrès de la science médicale et de l’instruction publique. La réflexion porte aussi sur le fonctionnement de ces cabinets privés de curiosité, particulièrement sur leur mode de constitution, leur décor intérieur ainsi que sur le contenu des collections qui nécessitaient certaines techniques d’organisation, d’acquisition et de conservation communes à celles de leurs homologues masculins. During the Enlightenment in France, curiosity culture constituted both a fashion and an intellectual and social game. This article explores thirty cabinets of curiosities run by women during this period. Wealthy women from the Parisiain and provincial aristocracy and middle classes organized cabinets in alchemy, mineralogy, physics and chemistry, natural history and biology studies. These cabinets can be divided into two distinct categories. The first represented amateur interests; they were developed for show and served as schools for intellectual and éducational pleasure. The second category were run by experienced women scientists who used their laboratories to conduct experimental research in the name of medical progress and public instruction. This article also describes how these private cabinets functioned, their composition, their interior décor as well as the contents of the collections, which required the organisation, acquisition and preservation techniques used in their male counterparts.

Abstract: Le métier de réalisateur de cinéma a été peu étudié en sciences humaines et sociales. Les études cinématographiques et la presse spécialisée s’intéressent beaucoup moins aux parcours des collaborateurs de l’équipe technique qu’aux comédiens et aux projets des auteurs que l’on pourrait croire retranchés dans leur intériorité créatrice. Pour échapper à ce travers biographique, le sociologue peut rendre compte de l’itinéraire d’un réalisateur en examinant ses liens avec ses collaborateurs. On prend ici l’exemple des liens de travail récurrents qui unissent Dominique Cabrera, réalisatrice de cinéma, à ses proches collaborateurs, en montrant de quelle manière des liens de travail peuvent s’approfondir en dépit d’un contexte sectoriel où l’organisation par projet de l’activité cinématographique favorise a priori la fugacité des relations de travail. On postule ainsi que le contexte pertinent pour éclairer les contours de cet itinéraire de carrière est le réseau des relations socioprofessionnelles au sein duquel naissent les projets de films et qui contribuent à ancrer ce parcours dans le paysage professionnel des pratiques cinématographiques. In France, social sciences and humanities have paid little attention to the profession of film director. Film studies and the press are much less interested in the professional careers of technical teams in comparison with the lives of actors and authors of projects who seem to be cut into their inner creativity. To escape the biographical pitfall, sociologists may account for a director’s itinerary by analyzing his/her ties with colleagues. Here, we examine the recurrent work relations between film director Dominique Cabrera and her close colleagues, to underline the strong relationships which paradoxically unite both the director and her staff in a project-based industry that is rather conducive to short-lived work relations. We postulate that the professional network produced consistently helps to describe and understand the course of a career, and that the relevant context to illuminate its contours is the network of professional relations that engenders film projects and is instrumental in fastening the career in the professional landscape of cinema.
