Determination of in vivo toxicity and in vitro cytotoxicity of venom from the Cypriot blunt-nosed viper Macrovipera lebetina lebetina and antivenom production

Abstract: the venomous levantine viper, macrovipera lebetina lebetina is endemic to cyprus. the objective of this study was to investigate in vitro cytotoxicity, in vivo lethality, and antivenom production followed by a re-immunization schedule in mice against macrovipera lebetina lebetina venom. the ld50 value was estimated as 7.58 mg/kg within 24 hours by different venom doses administrated intraperitoneally in mice. freund's complete and incomplete adjuvants were used for first and second immunization of mice in antivenom production. a cell-based assay was performed to determine the effects of macrovipera lebetina lebetina venom and antivenom neutralizing potency on l929 cell viability. the snake venom toxicity and cytotoxicity were examined and comparison of results showed good correlation, the ld50 value was tenfold higher than the ic50 value. the ic50 value was 0.62 ± 0.18 ml after 48 hours treatment while the calculated value was 1.62 ± 0.25 ml for the culture media totally refreshed after two hours treatment with venom. the in vitro efficacy of antivenom against macrovipera lebetina lebetina venom was found to be low. this is the first report that describes the in vivo and in vitro toxic effects of macrovipera lebetina lebetina venom and antivenom production against this species.
Antimicrobial and antioxidant activity of brown algae from the Aegean Sea

Abstract: The present study was conducted to evaluate the antioxidant and antimicrobial activity of methanol, dichloromethane and hexane extracts, as well as the essential oils of brown algae (Phaeophyta) Colpomenia sinuosa, Dictyota dichotoma, Dictyota dichotoma var. implexa, Petalonia fascia and Scytosiphon lomentaria. The essential oil of the macroalgae was obtained by steam distillation and analyzed by GC and GC/MS. The antioxidant activity of the algal extracts was determined using the procedures of inhibition of b-carotene bleaching and ABTS+ methods. The antioxidant effects of the extracts were compared with those of commercial antioxidants, such as butylated hydroxytoluene (BHT), butylated hydroxyanisol (BHA) and a-tocopherol. The hexane extracts of D. dichofoma var. implexa had a higher phenolic content than the other extracts. The dichloromethane extract of S. lomentaria was found to be more active in the decolorization of ABTS+ than the other extracts and generally the dichloromethane extracts were more active than the methanol and hexane extracts. Antimicrobial activities of the extracts were assessed against Gram (+) and Gram (–) bacteria and one yeast strain by the disk diffusion method. According to the re sults, the dichloromethane extracts generally showed more potent antimicro bial activity than the methanol and hexane extracts at concentrations 1.5 and 1.0 mg/disk.
Electron Microscopic Evaluation of the Secretory Mechanisms of Renin from Juxtaglomerular Cells

Abstract: although the structure and the functions of juxtaglomerular cells (jg) have been well defined, there is still a controversy about the secretory mechanisms of renin from these cells. it has been assumed that exocytosis is the main secretory mechanism in these cells in many studies, while others suggest that secretion occurs in a quite different way in these cells. there are several studies suggesting that diacrine secretion, which is very difficult to visualize, might be the other mechanism for secretion of renin. this study is an attempt to find the answers of these questions by identifying the fine structural features of the secretory granules in juxtaglomerular cells. cyclosporin a (cya) has been used in the current experimental study since it has already been reported that this drug increases the number of jg cells and stimulates secretion of renin. twelve female sprague-dawley rats had daily intraperitoneal injections of cya for ten weeks. tissue specimens from the kidneys of these animals were examined by electron microscopy. fine structural characteristics of the secretory granules of juxtaglomerular cells have been examined. considerable amount of granules, which goes to the exocytotic process, have been observed. additionally, several cells, which their granules had been secreting their contents in a different way, were found. this was interpreted as the secretion type of diacrine secretion. in conclusion, this in vivo study presents morphologic evidences demonstrating that both exocytosis and diacrine secretion might occur in jg cells. we also had a chance to observe secretory granule probably exhibiting "diacrine secretion", which is very difficult to visualize, at electron microscope level for the first time. this report also provides morphologic proof which shows that these two distinct secretory mechanisms might occur simultaneously in the same juxtaglomerular cell.

Abstract: Children should have flexible and adaptable repertoire to accomplish qualified social skills. In order to achieve this, teachers have a significant responsibility to propose crucial opportunities to children during classroom activities. Although teachers prefer to use universal approaches to promote prosocial behaviors rather than to remove maladaptive behaviors, cultural issues in social problem solving skills would be effective on their behavioral pattern of treatment to children. In this study, it was evaluated to see possible differences between Turkish and Flemish preschool children in their behavioral patterns. It was found that there was only significant difference in prosocial dimension of Drexel Early Childhood Behavior Rating Scale (Shure, 2005). In aggression and introversion dimensions, it was not found any significant difference between the two groups. However the mean scores of Flemish preschoolers in aggression dimension of the scale were found less than Turkish preschoolers. Findings were discussed based on literature.

Choice of drugs in the treatment of chronic hepatitis B in pregnancy

Abstract: The selection of antiviral drugs for chronic hepatitis B (CHB) treatment in pregnancy is very difficult since none of the drugs have been approved for use in pregnancy. Transmission from mother to newborn remains the most frequent route of infection in mothers with high viral load and positive hepatitis B e antigen status, even with the use of appropriate prophylaxis with hepatitis B virus (HBV) immunoglobulin and HBV vaccination. We read from the article written by Yi et al that lamivudine treatment in early pregnancy was safe and effective. However, we could not understand why adefovir dipivoxil (ADV) was used in three pregnancy cases, since ADV has been classified as pregnancy category C. In pregnancy, telbivudine or tenofovir should be selected when the treatment of CHB is necessary, since these drugs have been classified as Food and Drug Administration pregnancy risk category B.
