Immunohistochemical assessment of mucosal cytokine profile in acetic acid experimental colitis

Abstract: experimental colitis induced by acetic acid has been used extensively as a model for intestinal inflammatory disease. colonic tissue lesions of intestinal inflammatory disease patients seem to be related to the increased local production of pro-inflammatory cytokines (il-1, il-6, tnf-a, and ifn-g). purpose: to assess the cytokine expression pattern identified through immunohistochemistry in colonic mucosa after experimental colitis induced by acetic acid and establish the relationship between this pattern and the presence of macroscopic lesions. materials and methods: adult male wistar rats (n = 39) were divided at random into 4 groups: nc45 and nc24 (control without colitis; sacrificed at 45 minutes and 24 hours, respectively); and wc45 and wc24 (with experimental colitis induced by acetic acid; sacrificed at 45 minutes and 24 hours, respectively). macroscopic and microscopic alterations in colonic tissue were evaluated, and cytokine expression was assessed through immunohistochemistry. results: after 24 hours, il-1 expression was greater in the groups with colitis when compared to the groups without colitis. il-4 expression was higher in the wc45 group. there was an increase in both inf-g and il-6 related to the presence of necrosis of the colonic mucosa in the groups with colitis for both periods evaluated. conclusion: the immunohistochemical technique was efficient for the analysis of various cytokine expressions in the colonic tissue. there was an increase in the il-1 pro-inflammatory cytokines as well as in il-6 and ifn-g associated with the presence of colonic necrosis. experimental colitis induced by acetic acid is a useful model for the development of studies assessing the role of cytokines in the inflammation of mucosa as well as anti-cytokine therapies.

Abstract: the objectives of this work were to determine the viability of using a selection index based on the sum of standardized variables in common bean breeding programs, and to identify the most promising segregating populations for grain productivity, plant growth habit and resistance to lodging simultaneously. segregant populations obtained by crosses in partial diallel scheme (6x6) were evaluated. parental genotypes were divided in two groups. group i comprised parents of the carioca grain type, with semierect to prostate growth habit. group ii comprised parental lines with erect architecture, but with grains not classified as the standard carioca type. the f2 and f3 generations of the obtained hybrid combinations were evaluated in randomized block design, with three replicates and sowing in november 2007 and february 2008, respectively. data of grain yield, score for plant growth habit and score for lodging were standardized per plot (zij). the selection index for the three characters together was obtained from the zij sum. this selection index makes it possible to choose superior segregating populations considering grain yield, scores for plant growth habit, and for lodging simultaneously. segregating populations cv iii 8511 x brs 7762 supremo, cv iii 8511 x rp 166 and cv iii 8511 x rp 26 are identified as promising for breeding programs for grain productivity, plant architecture, and resistance to lodging, simultaneously.

Abstract: the objective of this work was to verify the nine?hole system efficiency in the evaluation of common bean (phaseolus vulgaris) carioca-type progenies for plant architecture. ninety-seven progenies were evaluated in the f2:3 and f2:4 generations in an randomized block design, with 20 replicates, and nine plants per plot, in which the central plant was evaluated, and the eight neighbor ones were considered together as the control. the evaluated traits were: grain yield per plant, architecture and lodging. the joint selection index (zij) for the three characters was obtained through the sum of the standardized values per plot. data were submitted to the analysis of variance, and from the mean square expected values, the components of variance were estimated. it is possible to identify more upright plants using the nine?hole system, in segregating common bean populations. this system allows for high estimates of experimental accuracy and heritability.

Abstract: the objective of this work was to evaluate the efficiency of phenotypic recurrent selection for the number of days until blooming in common bean, and to verify if this selection affected other traits. one s0 population derived from the mixture of the f2 populations of 11 hybrid combinations evaluated for earliness was used. the plants of the s0 (cycle 0) which bloomed first were crossed to obtain cycle i. to go on with the selection the obtained seeds were used. the process was repeated until the fifth selection cycle. fifty-three s1:2 progenies of each cycle were used for evaluation of the genetic progress. the number of days to flowering, severity of angular leaf spot (phaeoisariopsis griseola), number of days to maturation, yield and grain type were evaluated. a selection gain of 2.2% per year was observed, which indicates that phenotypic recurrent selection reduced the number of days to flowering. the selection for the number of days to flowering had no correlated response with other evaluated traits, indicating that progeny selection for early flowering can be associated with the phenotypic expression of other traits, according to the breeder's interest.

Abstract: duchenne muscular dystrophy (dmd) is among the most serious and incapacitating childhood myopathies. children progressively lose strength and usually die from respiratory infection by the end of their adolescence. the first symptoms are usually noticed in early childhood by teachers or caregivers. this study aimed to analyze the effects of collaborative consultation and planning proposal involving physiotherapy with classroom teachers and physical education teachers of two students with dmd, who presented severely limiting functional abilities; the objective was to improve their participation and comfort in the two most significant contexts of their daily lives: at school and at home. visits to the two students' schools and to one of their homes were undertaken, in order to investigate their actual needs. data was collected through interviews with the students, their parents, classroom teachers and physical education teachers. the strategies that were developed involved the adaptation of school furniture, acquisition of low tech materials and resources for the students' use and promotion of specialized orientations for the teachers involved and parents of the student with greater physical needs. it was possible to observe that the ecologic physiotherapy approach, aimed at environmental adaptations and collaborative planning of activities, provided some help and comfort for the participating students and enabled greater involvement/participation at school, besides contributing to the teachers' development. furthermore, the adaptations of the students' homes and supervision of the parents were demonstrated to be essential measures as they provided some comfort and autonomy to the student with more severe limitations in the home environment.

Abstract: the efficiency of recurrent selection was assessed in obtaining common bean (phaseolus vulgaris l) plant lines resistant to the phytopathogenic fungi phaeoisariopsis griseola. the base bean population was obtained from the partial diallel between seven lines with carioca-type grains and 10 sources of resistance to p. griseola. the plants most resistant to the pathogen were selected in the f2 (s0) generation of the populations (c-0). the best s0:1 plants that presented carioca-type grains were intercrossed to obtain cycle i (c-i). the same procedure was adopted to obtain cycles c-ii to c-v. in each recurrent selection cycle, s0:1 progenies selected were also assessed in experiments carried out in lavras, brazil, always using as check the carioca mg (susceptible to p. griseola) and pérola (tolerant) cultivars. the response to selection for resistance to the pathogen was estimated from the general mean of the s0:1 progenies from each selective cycle compared to the susceptible check carioca mg. the estimated gain was 6.4% per cycle and the indirect response in grain yield by selection for resistance to the pathogen was 8.9% per cycle. the variability detected among the progenies in the last selective cycles enabled the prediction of additional responses to recurrent selection.

Prevalence of c-myc expression in breast lesions associated with microcalcifications detected by routine mammography

Abstract: context and objective: genetic abnormalities in cell proliferation-regulating genes have been described in premalignant lesions. the aims here were to evaluate c-myc protein expression in non-palpable breast lesions associated with microcalcifications, detected by screening mammography, and to compare these results with histopathological, clinical and epidemiological variables. design and setting: analytical cross-sectional study, with retrospective data collection, in a university hospital in s?o paulo. methods: seventy-nine female patients who underwent routine mammography between 1998 and 2004 were studied. lesions classified by the breast imaging reporting and data system (bi-rads) as 4 or 5 underwent percutaneous biopsy using a large-core needle. ninety-eight lesions were studied anatomopathologically. paraffin blocks properly representing the lesions were selected for immunohistochemical analyses using the streptavidin-biotin-peroxidase technique with monoclonal mouse c-myc antibodies. results: among the 98 lesions, 29 (29.6%) contained malignant neoplasia; 40 (40.8%) had a positive immunohistochemical reaction for c-myc. when the groups were divided between lesions without atypias versus atypical lesions plus malignant lesions, 31.03% of the 58 lesions without atypias were positive for c-myc and 55% of the 40 malignant and atypical lesions (p = 0.018). comparing the atypical lesions with ductal carcinoma in situ versus the benign lesions without atypias, c-myc was present in 51.61% of the 31 atypical lesions and 31.03% of the benign lesions without atypias (p = 0.057). conclusion: c-myc protein was more frequently expressed in atypical and malignant lesions than in benign lesions without atypias. c-myc expression correlated with the presence of atypias (p = 0.018).
Genetic control of early grain darkening of carioca common bean

Abstract: To obtain information on the genetic control of early darkening in common bean with carioca grain and verifythe feasibility of selection by means of a grade scale, two contrasting parents were crossed, line VC-3 with cultivar BRSMGMajestoso. The seeds of F2 plants and F2:3 progenies were evaluated 30, 60 and 90 days after harvest for grain darkening, ona 1 - 5 grade scale. Genetic and phenotypic parameters were estimated. The grade scale efficiently discriminated the progeniesin terms of grain darkening. The heritability of the trait increases with the storage time and was high enough to ensure successwith selection after 30 days. The genetic control seems to be monogenic with dominance of the allele for early darkening.
