Abstract: 目的探讨乙型肝炎病毒通过乙型肝炎病毒X蛋白(HBx)对肝细胞衰老的影响。方法对HepG2.2.15细胞、HBx沉默HepG2.2.15细胞(HepG2.2.15HBxsi)及HBx过表达HepG2细胞(HepG2HBx)进行β半乳糖苷酶染色,采用Western Blot对肝细胞衰老相关蛋白进行检测;对比分析HBx对肝细胞衰老的影响。结果β半乳糖苷酶染色阳性细胞比例:HepG2.2.15组为(0.480±0.096),HepG2组为(0.016±0.005),两组比较差异有统计学意义(P<0.001);HepG2.2.15HBxsi组为(0.278±0.065),对照组HepG2.2.15HBxnc组为(0.329±0.044),两组比较差异无统计学意义(P=0.092);HepG2HBx组为(0.319±0.033),对照组HepG2HBxcon组为(0.064±0.012),两组比较差异有统计学意义(P<0.001)。Western Blot检测肝细胞衰老蛋白:HepG2.2.15组和HepG2HBx组肝细胞衰老相关蛋白p53及p21表达量均高于其对照组,HepG2.2.15HBxsi组肝细胞衰老相关蛋白p53及p21表达量低于HepG2.2.15HBxnc组。结论乙型肝炎病毒可通过HBx促进肝细胞衰老
Overexpression of polo-like kinase1 predicts a poor prognosis in hepatocellular carcinoma patients

Abstract: AIM: To elucidate the role of overexpressed polo-like kinase1 (PLK1) in hepatocellular carcinoma (HCC).METHODS: We prospectively collected clinicopathological, immunohistochemical and semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR) data from 135 HCC patients undergoing successful hepatectomy. The correlations between PLK1 mRNA expression and clinicopathologic variables were analyzed by Mann-Whitney U test. Prognostic factors were identified by univariate and multivariate analyses.RESULTS: Immunohistochemical results showed overexpression of PLK1 was mainly found in tumor tissues compared with tumor-free tissue. A similar mRNA result was obtained by semi-quantitative RT-PCR. A total of 111 samples were positive for PLK1 mRNA expression. The positive expression was correlated with venous invasion, tumor nodules and Edmondson grade. Furthermore, 1, 3, 5-year survival rates in the positive expression group were significantly lower than the negative control group. Multivariate analysis showed that positive PLK1 expression was an independent risk factor for HCC.CONCLUSION: PLK1 could be a potential biomarker for diagnosis and therapy for HCC.
Expression of thymidine kinase mediated by a novel non-viral delivery system under the control of vascular endothelial growth factor receptor 2 promoter selectively kills human umbilical vein endothelial cells

Abstract: AIM: To investigate the killing efficiency of a recombinant plasmid containing a thymidine kinase (TK) domain insert driven by the vascular endothelial growth factor receptor 2 (VEGFR2) promoter (KDR) on vascular endothelial cells.METHODS: The KDR-TK fragment was extracted from pBluescript II KDR-TK plasmid by enzymatic digestion with XhoI and SalI. The enhanced green fluorescence protein (EGFP) carrier was extracted from pEGFP by the same procedure. The KDR-TK was inserted into the pEGFP carrier to construct pEGFP-KDR-TK. Using ultrasound irradiation and microbubble, pEGFP-KDR-TK was transferred into human umbilical vein endothelial cells (HUVECs). The transient infection rate was estimated by green fluorescent protein (GFP) expression. Transfected HUVECs, non-transfected HUVECs, and HepG2 cells were cultured in the presence of different concentrations of ganciclovir (GCV), and the killing efficacy of HSV-TK/GCV was analyzed by 3-[4, 5-dimethylthiazol-2-yl]-2, 5-diphenyl tetrazolium bromide (MTT) assay.RESULTS: The recombinant pEGFP-KDR-TK was successfully constructed by inserting the KDR-TK fragment into the pEGFP carrier. Transfected HUVECs showed cytoplasmic green fluorescence, and the transient transfection rate was about 20.3%. Pools of G418-resistant cells exhibited a higher sensitivity to the prodrug/GCV compared to non-transfected HUVECs or non-transfected HepG2 cells, respectively.CONCLUSION: KDR promoter and the suicide gene/prodrug system mediated by diagnostic ultrasound combined with microbubble can significantly kill HUVECs. Such therapy may present a novel and attractive approach to target gene therapy on tumor vessels.

Abstract: 目的探讨直接抗人球蛋白试验(DAT)阳性在慢性重型乙型肝炎患者贫血中的临床意义。方法对30例健康人、30例慢性乙型肝炎(慢乙肝)患者和50例慢性重型乙型肝炎(慢重肝)患者红细胞相关参数及DAT进行检测,并对影响DAT阳性的临床相关因素进行分析。结果慢重肝患者红细胞计数、血红蛋白(Hb)浓度以及红细胞压积(HCT)均低于慢乙肝患者和健康组(P<0.05),而慢重肝患者红细胞体积分布宽度(RDW)高于慢乙肝和健康组(P<0.05);慢重肝患者、慢乙肝患者和健康组DAT阳性率分别为62.82%、13.33%和0。DAT阳性的慢重肝患者红细胞计数、Hb浓度和HCT均低于相应DAT阴性患者(均P<0.05),RDW却高于DAT阴性慢重肝患者(P=0.001);慢重肝患者红细胞毛细管分离后,衰老红细胞的DAT阳性强度高于年轻红细胞DAT阳性强度(P<0.001);在慢重肝患者中,DAT的阳性和阴性的患者在性别、年龄、丙氨酸氨基转移酶、总胆红素、补体C3、C反应蛋白及是否并发糖尿病方面存在差异(均P≤0.05)。结论慢重肝患者贫血的产生可能与衰老红细胞吸附抗体所致的免疫性溶血有关

Abstract: 20世纪90年代的高校扩招使我国高等教育从“精英 教育”走向“大众化教育”,高等医学教育招生规模也大幅增 加。然而,学校的管理、师资等无法与之相匹配,教学质量受 到较大影响。高等医学教育的发展模式由“粗放型”向“内涵 型”转变是我国医学高等教育发展的必然趋势。高等医学教 育内涵发展的价值属性主要通过高等医学教育的公共性、精 英性、实践性等特征得以体现。全面深化医学教育体制改革, 培养医学科研创新能力,实施人文素质教育,统筹协调医学 教育布局,加强师资队伍建设,是实现高等医学教育内涵发 展的有效路径。
Chinese higher education has changed from “elite education” to “public education” by the promotion of enrollment expansion since 1990’s. The enrollment scale of higher medical education also has increased significantly. However, management and lecturers of universities and colleges couldn’t satisfy the high-speed development, education quality has been greatly affected. Therefore, it is an inevitable trend of China’s higher medical education’s development that the development mode of higher education changes from “extensive type” into “connotative type”. The value attributes of the connotation development of higher medical education is mainly reflected by features of publicity, elite and practicality. Comprehensively deepening the medical education reform, training the innovation ability of medical scientific research, carrying out humanistic quality education, coordinating the layout of medical education, enhancing the staff construction are the effective paths to realize the connotation development of higher medical education.
Ethno Literary Identity and Geographical Displacement: Liu Na'ou's Chinese Modernist Writing in the East Asian Context

Abstract: In the course of his short literary life, Liu Na'ou travelled across four geographical areas: Taiwan, Japan, Shanghai and Beijing, as well as five cultural domains: Taiwanese, Japanese, French, English and Chinese. The transnational facet of Liu's modernist writing is not merely literary or cultural but political and historical. The earliest modernist writing in China was initiated on the basis of the colonial experience of Taiwan and the semi-colonial modernity of Shanghai. It became a “contact zone” in which various cultural and political influences to contest with each other, signaling a process of power shifting in East Asia in the early 20th century. This article aims to reflect on theoretical issues ranging from nationalism, cosmopolitanism, colonialism and semi-colonialism, to the colonial modernity of East Asia, drawing upon Liu's modernist writings in 1928.

Abstract: This paper proposes a sequential learning neural net which consists of the bounded weight adjustment algorithm and structure adaptive adjustment method. This network is characterized by efficiently learning the knowledge of new samples in series. After an efficient sequential learning algorithm of this network, is presented its learning feature and recognition performance in detail are analyzed. The effectiveness of this network have also been shown by theoretical analyses and a lot of experiments in this paper.

Abstract: This paper proposed a novel intelligent data processing model which applied to fishery forecasting based on the Indian Ocean bigeye tuna fishery application background. Firstly, obtained the support vectors by training the SVM with the sample data. Secondly, extracted the rules from the support vectors by the fuzzy classifier method. Lastly obtained the fishery dynamic knowledge by the extension transformation for the conditions and the conduction transformation for the conclusions. Experimental results show that the dynamic knowledge is of a forceful interpretive capacity, and it enriches fisheries forecasting methods and reveals the formation mechanism of fishing grounds.

Abstract: With the development of software technology, workflow system is being used more and more frequently in the business process of enterprises. How to integrate JBPM into the Spring framework efficiently becomes one of the key points in the establishment of whole system's architecture. Based on analyzing the existing model of deployment and problems of process file deployment in the development of process component, a new deployment model is proposed, in which the deploy process is controlled by server startup parameters combining with SpringModules's inverse control. This new model makes the deployment of process file independent of J2EE system and provides a more loosely coupled integration.
