The Effect of the PD98059 on H2O2 Induced Apoptosis and Associated Proteins of Human Breast Cancer Cells

目的:探讨细胞外信号调节激酶的上游激酶(MEK)抑制剂PD98059对H2O2诱导的人乳腺癌MCF-7细胞凋亡的影响及其作用机制。方法:H2O2诱导乳腺癌MCF-7细胞凋亡,PD98059干预其细胞凋亡。荧光染料吖啶橙(AO)观察细胞凋亡的形态学变化;流式细胞术法进行凋亡细胞计数;Western blot法检测p53,PCNA,caspase-3、caspase-9和Bcl-2的表达。结果:H2O2刺激细胞4 h、12 h,明显诱导了MCF-7细胞凋亡。PD98059将H2O2诱导的细胞凋亡率分别降低了约4%、14%,与H2O2刺激细胞12 h比较,具有显著的统计学差异(P < 0.05)。H2O2显著提高了p53蛋白的表达,激活了caspase-3与caspase-9蛋白,降低了Bcl-2的表达,而PD98059逆转了H2O2的刺激效应,下调了H2O2导致的p53与PCNA蛋白表达,降低了caspase-3与caspase-9蛋白活性,上调了Bcl-2蛋白表达。结论:MEK抑制剂PD98059抑制了H2O2诱导的MCF-7细胞凋亡,其作用机制与其降低了p53和PCNA蛋白表达和caspase-3与caspase-9蛋白活性以及增加Bcl-2蛋白表达有关。
Objective: to study the effect of MEK inhibitor of PD98059 on apoptosis induced by H2O2 and its mechanisms in human breast cancer MCF-7 cells. Methods: H2O2 inducing apoptosis in MCF-7 cells was set to observe apoptosis and its associated proteins treated in the present or absent of PD98059. The cellular morphology was investigated by AO staining. Apoptosis ratio was analyzed by flow cytometry. The expres-sions of p53, PCNA, caspase-3, caspase-9 and Bcl-2 were observed using Western blot analysis. Results: apoptosis was observed with cells treated H2O2 for 4 or 12 h while the degree of apoptosis was greatly de-creased by 4% or 14% in MCF-7 cells treated with H2O2 and PD98059 together. There was significantly dif-ference between with the treatment of H2O2 and PD98059 in the presence of H2O2 for 12 h (P < 0.05). The increased p53, activated caspase-3 and caspase-9 and decreased Bcl-2 were appeared with H2O2 treatment. However, the levels of p53 and PCNA were downregulated, the extent of caspase-3 and caspase-9 activation was decreased, and the level of Bcl-2 was upregulated in MCF-7 cells treated with PD98059 in the presence of H2O2. Conclusions: MEK inhibitor of PD98059 inhibited apoptosis by downregulating the levels of p53 and PCNA, decreasing the activation of caspase-3 and caspase-9 and upregulating the level of Bcl-2 in MCF-7 cells.

Abstract: 侵袭和转移是恶性肿瘤的基本特征,也是导致肿瘤患者死亡的主要原因。防止和阻断肿瘤的侵袭和转移是降低恶性肿瘤死亡率的重要途径。鉴于目前抗肿瘤转移化疗药物疗效的局限性及其毒副作用,从中药中寻找安全性高、且能够有效预防和控制肿瘤转移的药物越来越受到重视。中药在干预肿瘤转移上具有一定的优势,近几年来对中药及其成分干预肿瘤转移的研究报道较多,并且取得了一定的成果。本文综述近几年的研究成果,为今后的研究提供参考。
Advances in Traditional Chinese Medical Treatment Research in Liver Cancer Postoperation

With the improvement of early diagnosis rate for primary liver cancer (LC), the surgical treatment is more and more increasing. However, how to deal with postoperative adverse reactions, side ef- fects caused by chemotherapy and postoperative recurrence and metastasis, is one of the clinical problems. Traditional Chinese Medicine (TCM) theory and its unique system of diagnosis and treatment practices can improve postoperative and chemotherapy-induced adverse reactions. There is great significance to improve the postoperative quality of life of patients with liver cancer, and prolong survival time in LC. This paper reviewed the advances in TCM treatment research on LC postoperation in recent years and put forward problems and countermeasures, to provide a reference for better TCM treatment on LC postoperation.

Recent Advances in Application and Effective Mechanism of Yinchenhao Decoction Treatment for Damp-Heat Syndrome

As a classic traditional Chinese formula for treatment of damp-heat syndrome, Yinchenhao decoc-tion can no merely be used to treat Damp heat jaundice or liver and gall diseases, but also be ap-plied to the damp-heat syndrome-based treatment for dermatoses, endocrinopathy, gynecological and obstetric diseases etc. The pharmacological mechanism due to regulate and control the main pathological events of damp-heat syndrome in diseases. In this article, according to compatibility law, clinical application, therapeutic effect and mechanism and pathological mechanism of damp- heat syndrome, we reviewed the recent advances in the researches on Yinchenhao decoction to provide a reference for further studies.

Abstract: Chronic disease is a complex process. In this paper, authors analyze the limits of both west medication and traditional Chinese medicines in preventing and treating chronic diseases. It is believed that chronic diseases have a sophisticated behavior of their own, and the treatment shall be tailored to such behavior. Authors suggest that the theory and approaches of systems science be made part of the theoretical basis and approaches for TCM based chronic disease prevention and treatment. They also propose to establish an innovative indicator system for evaluating the therapeutic effects of chronic diseases treatment with traditional Chinese medicine, and associated models and processes.
Study of Traditional Chinese Medicine Syndrome by Partial Least Squares and Similarity Analysis

Abstract: Partial least squares (PLS) and similarity analysis were developed to study the mathematic model of traditional Chinese medicine (TCM) syndrome with post-hepatitis cirrhosis (PHC). The similarity of clinical characteristic information (symptoms) and biological parameters with TCM syndromes were calculated. The resemblance between TCM syndrome and clinical information of disease obtained from similarity analysis is basically in accordance with the clinical practice, so, the technique could be taken as a method for finding the characteristics of TCM syndrome with simplified clinical parameters. Stepwise regression was used to analyze the clinical information, including clinical symptoms and biological parameters. The types of syndromes and the related clinical information which was screened by stepwise regression were set up as dependent variable matrix and independent variable matrix respectively. Then, the models in TCM were obtained for the classification and diagnosis of TCM symptom by Leave-One-Out in PLS program. PLS was definite useful in classification and modeling of TCM complicated syndromes.

Abstract: This paper discussed traditional Chinese medicine(TCM) complex system research based on the previously suggested study aim,mission,methodology,research approaches and research methods.Five aspects of TCM complex system study were explained,which are "relation," "evolvement," "emergent," "nondeterminacy" and "complex network." Current hot-spot issues and research methods in TCM complex system were also explored.And future research questions and directions of TCM complex system have been put forward.
