超声靶向破坏微泡技术介导小鼠肝癌细胞株JNK1基因表达、细胞迁移和侵袭抑制

Abstract: 目的:探讨应用超声靶向破坏微泡(UTMD)技术介导小鼠肝癌细胞株JNK1基因的表达、细胞迁移和侵袭抑制的作用,阐明其作用机制。方法:构建并筛选RNA干扰效果最好的短发夹RNA(shRNA)。将小鼠肝癌细胞株Hca-F分为正常Hca-F细胞组、shRNA质粒组、脂质体组、超声微泡结合超声辐照组及脂质体结合超声微泡加超声辐照组。采用倒置荧光显微镜观察各组细胞转染率,荧光定量PCR和Westernblotting法检测JNK1基因mRNA和蛋白表达水平,CCK-8法检测各组细胞的细胞活性,应用Transwell实验检测各组细胞的体外迁移能力。结果:脂质体结合超声微泡加超声辐照组细胞转染率高于shRNA质粒组、脂质体组和超声微泡结合超声辐照组(均P<0.05),脂质体组和超声微泡结合超声辐照组比较差异无统计学意义(P>0.05)。脂质体结合超声微泡加超声辐照组JNK1mRNA和蛋白表达水平低于其他各组(P<0.05);脂质体结合超声微泡加超声辐照组细胞活性和平均穿膜细胞数均低于其他各组(P<0.05)。结论:UTMD技术结合脂质体转染法可以提高小鼠肝癌细胞株JNK1shRNA的转染效率,增强其对基因表达、细胞活力、迁移和侵袭能力的抑制。

This paper addresses the issue of land water cycling and water system approach that is a coupling system of basic hydrological process with geo-chemical & bio-chemical processes and human action compo- nent under the climate change and human activity. By reviewing international and national research, some of analysis and comments are presented on water system observations, modeling, regulation & control. 介观LC电路不可避免会处于辐射场中,把辐射场看成是由无穷多谐振子组成的,选择介观LC电路为研究对象。本文从介观LC电路与辐射场相互作用的哈密顿量出发,把辐射场看成库,通过对库求迹,推导了介观LC电路在辐射场作用下的密度方程。
Quantum LC circuit locates inevitably in radiation field; the thermal radiation field is a reservoir which is described by infinite harmonic oscillators, and quantum LC circuit is a system to be investigated. From the interaction between mesoscopic LC circuit and radiation field, the radiation is reservoir. We trace out the reservoir and deduce the equation of density.

Abstract:

本文从理论上探讨了各向异性的人工媒介与空气界面的Goos-H?nchen相移,然后通过数值计算证实了理论的分析结果。再研究了人工媒质的耗散对Goos-H?nchen相移的影响。Goos-H?nchen相移的符号和大小取决于不同的电磁参数、入射角以及各向异性人工介质板的厚度。同时不同于无耗散的各向异性的介质板,在弱耗散的情况下,耗散对横向的相移产生了很大的影响,在一定的条件下,介质弱耗散可以产生很大的影响。这种特殊的现象可应用于光学装置设计中。
The Goos-H?nchen shift (the lateral shift) has been investigated from the anisotropic metamaterial slab theoretically and numerically. The sign and degree of the shift can be determined by the choices of the electromagnetic parameters and thicknesses of slab, which is different from the case of the isotropic media. We also find that the weak lossy may produce large positive or negative lateral shift. This special phenomenon can apply to some optical device design.
