The Effect of Garcinia parvifolia Miq (Active Fraction) on Phagocytosis by Peritoneal Macrophages During Plasmodium berghei Infection in Mice

Abstract: The aim of this study was to asses the active fraction of the stem bark of G. parvifolia Miq to P. berghei on total red cells of mice and the number of intraperitoneal macrophage phagocytocide latex after infected by P. berghei. Three doses of active fraction of G. parvifolia at the dose of 25, 50, 100 and 200 mg kg-1 b.wt., respectively were used in this study and chloroquine was used as the positive control, while distilled water was used as a negative control. Phagocyte activity was measured by the ability of mice peritoneal macrophages to phagocytize latex particles in vitro. The result of this research indicated that at the dose of 200 mg/kg/day could reduce the number of parasite in the blood up to 54.90%. From the results indicated that the reduction of the number of parasite which circulated in the blood occurs at the doses of 50 and 100 mg/kg/day, although its activity is much lower than malaria drug of chloroquine. Macrophages activity (87.19%) increased at the dose of 200 mg/kg/day, compared to the control group (52.99%), these indicated that the fraction of G. parvifolia induced immune response. The increase of the phagocyte activity of the macrophage was stimulated by the active fractions of G. parvifolia (dose dependent) which was given to the P. berghei-infected mice.

Abstract: The Effect of Ethanol Extract of Daun Johar (Cassia siamea Lamk.) on the Macropages Activities and Capacyties. Daun Johar (DJ) (Cassia siamea Lamk.) has already been reported to stimulate the immune response. The current study investigates the role of DJ on mice macrophages activities and capacyties. Twenty four Swiss mice were divided into 6 equal groups. The first control group (Group I), received aquadest. The second control group (Group II), was given CMC Na 0,5% . The third control group (Group III), was given phytohemaglutinin. The case group: group IV received 23,78 mg ethanol extract of DJ/20 g BW, group V received 47,56 mg/20 BW, and group VI received 95,13 mg/20 g BW. These were injected orally on day 1 until 7. On day 8, Staphylococcus aureus (SA) were injected intraperitoneally. The macrophages activities and capacyties were counted on slide smears of mice peritoneal fluid. According to enhancement of dose, either the macrophages activities or capacyties were found. The lowest activity encounter on the negative control (369,5±18,1384) followed by CMC Na 0,5% (378,5±13,1783), doses of 23,78 mg/20gBW (466,75±9,4296), doses of 47,56 mg/20 g BW (557±30,2324), positive control/PHA (670,5±10,0830) and doses of 95,13 mg/20 g BW (683,5±12,1244)

Abstract: Characterization of Adenocarcinoma’s Autofluorescence Properties Using Multiexcitation Analysis Method. General purpose of this research is to get an early cancer detection method based on the properties of optical analysis between normal and adenocarsinoma tissue using the multiexcitation autofluorescence method. Observation of autofluorescence properties was done on the biopsy sample of adenocarcinoma tissues, GR mice transplanted by adenocarsinoma, and cell culture SM 1. Excitation on tissue was done by using the lamp Light Emitting Diode (LED) at some visible light wavelength range. This research obtained that the value of Intensity Auto fluorescence (IAF) at range red wavelength of cells and adenocarsinoma tissues tend to lower compared to the cells normal tissues if its were excited by blue LED. On the contrary, the value of IAF at infra red wavelength from cells and carcinoma tissues tend to higher compared to the cells and normal tissues if its were excited by red LED.

Abstract: The development of new medicines as immunostimulator agent has beingincreased according to the development of lethal diseases, for example tumor that suppress the immune responses. Propolis, which is resinous substance produced by honeybees known to have antimicrobial, anti- inflammatory, antioxidant, immunostimulator, and anti-tumor activities. The aims of this experiment were to study the effect of propolis as immunostimulator agent in proliferation of mice-suffered with mammary tumor T lymphocyte and the mammary-tumor cells’ viability after been treated with stimulated T lymphocyte. Three different dozes of propolis used in this experiment were 0.5 μg/mL, 1.5 μg/mL, 4.5 μg/mL, and added in cell culture to measure the proliferation of T lymphocyte. Proliferation activity of T lymphocyte was examined with 3-(4.5-dimethyl-2-thiazolyl)-2.5-diphenyl-2H-tetrazolium bromide (MTT) assay. Data was analyzed byusing Analysis of Variance (ANOVA) then followed by Duncan Multiple Ranges Test (DMRT) 5%. The result showed that the proliferation of T lymphocyte was increased after addition of propolis. The highest OD of T lymphocyte was reached in addition of propolis 4.5 μg/mL (2.363). In the examination of mice-mammary tumor cells’ viability, stimulated T lymphocyte was added to tumor cells culture and the results showed that the viability of tumor cells was decreased according to the increasing of T lymphocyte proliferation. The lowest tumor cells’ viability was reached in addition of stimulated T lymphocyte with propolis 4.5 μg/mL (4.667).

Abstract: The Effect of Ethanol Extract of Ketepeng Cina (Cassia alata L.) on the Macropages Activities and Capacyties. Ketepeng cina (KC) (Cassia alata L.) has already been reported to stimulate the immune response. The current study investigates the role of KC on mice macrophages activities and capacyties. Twenty four Swiss mice were divided into 6 equal groups. The first control group (Group I), received phytohemaglutinin. The second control group (Group II), was given CMC Na 0,5%. The third control group (Group III), was given aquadest. The cases group: group IV received 42 mg ethanol extract of KC/20 g BW, group V received 84 mg/20 BW, and group VI received 168 mg/20 g BW. These were admonished orally on day 1 until 7. On day 8, Staphylococcus aureus (SA) were injected intraperitoneally. The macrophages activities and capacyties were counted on slide smears of mice peritoneal fluid. According to enhancement of dose, either the macrophages activities or capacyties were found. The lowest activity encounter on the negative control (group II and III) followed by Group IV, V, positive control (group I) and group VI.
PHOTOELECTROCHEMICAL DETECTION OF DENGUE-RELATED OLIGONUCLEOTIDE SEQUENCE USING ANTHOCYANIN AS A INTERCALATING AGENT AND ELECTROCHROMIC MATERIAL

Abstract: World Health Organization (WHO) presupposes a confirmation of dengue virus infection diagnosis with two criteria, i.e. clinical and laboratory criteria. One of the basic methods used by most laboratories to diagnose dengue virus is to detect oligonucleotide sequence using a DNA amplification technique. In this research, the measurement of dengue-related oligonucleotide was conducted by photoelectrochemical method. The presence of oligonucleotide sequence in target DNA can be detected by DNA probe that is immobilized on TiO2 electrode. The DNA hybrid is then bound to electrochromic substance like anthocyanin that generates current when it is subjected to light. The photocurrent is directly proportional to the number of target DNA. The aim of this research is to obtain photoelectrochemical system that has sensitivity and high responsiveness toward the change in oligonucleotide concentration, especially the applicability of anthocyanin as a electrochromic substance and intercalating agent. Linearity (R2) generated from the change of current in response to concentration changes of target DNA (in the concentration range of 0.75–3.00 nM) is 0.9611. Thus, this method has the potential to be developed to detect the presence of dengue virus in biological sample.
Design and Molecular Docking Study of Antimycin A3 Analogues as Inhibitors of Anti-Apoptotic Bcl-2 of Breast Cancer  [PDF]

Abstract: In this paper, we report the design and moleculardocking study of analogues of antimycin A3 as inhibitors of anti-apoptotic Bcl-2 of breast cancer. Twenty designed compounds and the original antimycin A3 were docked based on their interaction with breast tumor receptor binding target Bcl-2. The docking resulted in the five top-ranked compounds, namely, compounds 11, 14, 15, 16, and 20, which have a lower \"\"G binding energy, better affinity and stronger hydrogen bonding interactions to the active site of Bcl-2 than antimycin A3. Among those five top-ranked compounds, analogue compounds 11 and 14, which have an 18-membered tetralactone core and 18-membered tetraol core, respectively, exhibited the strongest hydrogen bond interaction, formed high stability conformation, and demonstrated the greatest inhibitory activity on the catalytic site of Bcl-2.
THE POTENTIALITY OF - CAROTENE AS FREE RADICAL SCAVENGER: A STUDY OF THE RAISE OF SUPEROXIDE PRODUCTION IN MITOCHONDRIAL LYMPHOCYTE

Abstract: This is a study on carotenoid extracted from ‘Minyak Buah Merah’ (Pandanus conoideus) in therapy simulation using high dose of β-carotene on two lymphocytes—that of a healthy subject and a leukemia patient. Superoxide production in mithocondrial lymphocyte as a result of β-carotene exposure was examined using a flow cytometer. The test finds that superoxide (a specific mitochondrial free radical) increases easier and faster in leukemic lymphocytes than in normal lymphocytes. The results of the study support previous meta-analysis studies that high dosages of β-carotene and vitamin E supplements as antioxidant should be discouraged because it increases all-cause mortality.
