Antitumor Activities of Kushen: Literature Review

Abstract: To discover and develop novel natural compounds with therapeutic selectivity or that can preferentially kill cancer cells without significant toxicity to normal cells is an important area in cancer chemotherapy. Kushen, the dried roots of Sophora flavescens Aiton, has a long history of use in traditional Chinese medicine to treat inflammatory diseases and cancer. Kushen alkaloids (KS-As) and kushen flavonoids (KS-Fs) are well-characterized components in kushen. KS-As containing oxymatrine, matrine, and total alkaloids have been developed in China as anticancer drugs. More potent antitumor activities were identified in KS-Fs than in KS-As in vitro and in vivo. KS-Fs may be developed as novel antitumor agents.

Abstract: Objective: To explore the antitumor activities of kushen (Sophora flavescens) flavonoids (KS-Fs) in vivo and in vitro.Methods: Cell proliferation was assayed by using methyl thiazolyl tetrazolium (MTT) method. H22 hepatocellular carcinoma and S180 sarcoma were induced in ICR mice. Lewis lung carcinoma was induced in C57BL/6 mice. H460 and Eca-109 tumor were induced in Balb/c nude mice by injecting 5×105 or 5×106 tumor cells in the right flank, respectively.Results: KS-Fs could inhibit the growth of a variety of human tumor cell lines (A549, SPC-A-1, NCI-H460, etc.) in vitro. The antitumor efficacies were confirmed in the mice models of H22, S180 and Lewis lung tumors and the nude mice models of human H460 and Eca-109 xenografted tumors. The oral or intravenous maximum tolerated dose of KS-Fs was more than 2.8 g/kg or 750 mg/kg respectively, far more than the oral medial lethal dose of kushen alkaloids (≤1.18 g/kg). No adverse reactions were observed.Conclusion: These results suggest that KS-Fs or kurarinone may be developed as a novel antitumor agent.
Antioxidant and Immunity Activities of Fufang Kushen Injection Liquid  [PDF]

Abstract: We investigated the effects of Fufang Kushen Injection Liquid (FFKSIL) on gastric immunity and oxidant-antioxidant status during N-methyl-N′-nitro-N-nitroso-guanidine (MNNG)-induced gastric carcinogenesis. The extent of lipid peroxidation and the levels of reduced glutathione (GSH) and activities of the GSH-dependent enzymes superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH-Px) were used to monitor the peroxidative balance. Enhanced lipid peroxidation in the gastric cancer animals was accompanied by significant decreases in the activities of GSH, GPx, GST and GR. Administration of FFKSIL significantly enhanced serum IgA, IgG, IgM, IL-2, IL-4 and IL-10 levels, decreased serum IL-6 and TNF-α levels, lowered the levels of lipid peroxides and enhanced GSH levels and activities of GSH-dependent enzymes. Our results suggest that FFKSIL blocks experimental gastric carcinogenesis by protecting against carcinogen-induced oxidative damage and improving immunity activity.
Antiviral Effect of Matrine against Human Enterovirus 71  [PDF]

Abstract: Human enterovirus 71, a member of the Picornaviridae family, is one of the major causative agent of hand, foot and mouth disease in children less than six years old. This illness has caused mortalities in large-scale outbreaks in the Asia-Pacific region in recent years. No vaccine or antiviral therapy is available. In this study, antiviral effect of matrine against enterovirus 71 were evaluated in vitro and in vivo. Matrine could suppress the viral RNA copy number on rhabdomyosarcoma cells. Moreover, matrine treatment of mice challenged with a lethal dose of enterovirus 71 reduced the mortality and relieved clinical symptoms. The results showed that matrine may represent a potential therapeutic agent for enterovirus 71 infection.

Abstract: reverse micellar extraction has been widely used in the purification of biomolecules. however, reverse micelles formed by ionic surfactants can only be employed for the extraction of biomolecules that are charged in the extraction system with the electrostatic interaction between surfactants and solutes as the driving force. in this study, the extraction of matrine by using reverse micelles formed by non-ionic trpo surfactants was studied. theoretical analysis and experimental results demonstrated that the driving force of the extraction is the coordination forces between matrines and trpos. using this coordination-based reverse micellar extraction, matrine can be efficiently separated from oxymatrine and other components in the raw matrine materials. experimental studies showed that the factors affecting matrine extraction include ph value and trpo concentration. the existence of ions in the system does not affect the partition coefficient significantly and the addition of a small amount of chloroform in the solution of reverse micelles was found to improve the extraction significantly.
三种核素对植物烂种烂芽的影响  [PDF]

Abstract: 为探讨核素及其浓度与植物烂种烂芽的关系,用Sr(NO3)2、CsNO3和UO2(NO3)26H2O,配制成0,0.1,0.5,1.0,2.5,5.0,7.5和10.0mmol·L-1的溶液,研究Sr、Cs和U在发芽试验中对向日葵、大豆、玉米、黄瓜和油菜种子烂种烂芽的影响。结果表明核素、核素浓度和植物对烂种烂芽的影响极显著,互作效应也非常显著。U处理的烂种烂芽率极显著地高于Sr或Cs处理,Sr或Cs处理之间差异不显著。不同Sr2+或Cs+浓度处理间没有显著差异。2.5mmol·L-1及以上U6+处理极显著地增加植物烂种烂芽率。植物之间对核素的反应差异较大,向日葵敏感,黄瓜不敏感,油菜、大豆和玉米介于二者之间。因此,不同核素对植物种子烂种烂芽的影响不同,U的影响大于Sr或Cs的影响,中高浓度U离子易致植物种子和幼芽死亡,不同植物烂种烂芽率对不同种类和不同浓度的核素反应不同。
Matrine and anti-tumor drugs in inhibiting the growth of human lung cancer cell line  [cached]

Abstract: Objective: To study the activities of matrine and anti-tumor drugs on SPCA/Ⅰ human lung adenocarcinoma cell line.Methods: Suppression effects of different concentrations of matrine and matrine combined with anti-tumor drugs on lung cancer cells were measured by methyl thiazolyl tetrazolium (MTT) colorimetric assay.Results: Different concentrations of matrine could inhibit the growth of SPCA/Ⅰ human lung adenocarcinoma cells and there was a positive correlation between the inhibition rate and the drug concentration. Different concentrations of matrine combined with anti-tumor drugs had higher growth inhibition rate than anti-tumor drugs alone.Conclusion: Matrine has direct growth suppression effect on SPCA/Ⅰ human lung adenocarcinoma cells and matrine conbined with anti-tumor drugs shows a significant synergistic effect on tumor cells.

Abstract: 海带(laminariajaponicaaresch),自开展人工养殖以来就不断地发生各种病害。其中发生较普遍,危害较大的有“绿烂病”、“白烂病”和“点烂病”三种。“绿烂病”先发生于叶片尖梢并很快地向叶片中部漫延,发病部分由褐色变为绿色,而后腐烂脱落。“白烂病”通常是先发生于叶片尖梢的叶绿,然后向叶片中部发展,发病部分呈现白色。“点烂病”则先是从叶片中部叶绿上或同时于梢部叶绿上出现一些不规则的小白点,随着白点的增加和扩大,使该部叶片变白腐烂或形成一些不规则的孔洞。

Abstract: A temperature-sensitive matrine-imprinted polymer was prepared in chloroform by free-radical cross-linking copolymerization of methacrylic acid at 60 °C in the presence of ethylene glycol dimethacrylate as the cross-linker, N-isopropyl acrylamide as the temperature-responsive monomer and matrine as the template molecule. Binding experiments and Scatchard analyses revealed that two classes of binding sites were formed on molecular imprinted polymer (MIP) at 50 °C. Additionally, the thermoresponsive MIP was tested for its application as a sorbent material for the selective separation of matrine from Chinese medicinal plant radix Sophorae tonkinensis. It was shown that the thermoresponsive MIP displayed different efficiency in clean-up and enrichments using the SPE protocol at different temperatures.
