Abstract: 在鉴证实务中,一直存在着在法律上无法对鉴证对象进行明确界定的问题,而套用审计准则来界定在范围上又显得过于狭窄,这一直是影响注册会计师开展鉴证业务的一个重要因素。本文从分析鉴证与审计的区别开始,探讨了鉴证准则增加“鉴证对象”的动因以及影响。【关键词】鉴证准则鉴证对象动因影响将于2007年1月1日起施行的《中国注册会计师鉴证业务基本准则》(简称“鉴证准则”)的第五章增加了“鉴证对象”的相关内容。鉴证准则第五章第二十一条规定,鉴证对象主要分为财务业绩或状况、非财务业绩或状况、物理特征、某种系统和过程、行为五大类,而与这五类鉴证对象相对应的信息形式分别为财务报表、反映效率或效果的关键指标、有关鉴证对象物理特征的说明文件、关于系统和过程有效性的认定、对法律法规遵守情况或执行效果的声明。
Characteristics of Streptomyces Flavogriseus -xylosidase and its Use in Xylan Hydrolysis  [PDF]

Abstract: -Xylosidase was induced when Streptomyces flavogriseus strain was grown in shake-culture of a mineral salt medium containing 1 per cent wheat straw hemicellulose for 3 days at 30 C. The intracellular -xylosidase was separated by precipitation with acetone, ammonium sulphate and purified on Sephadex G-150. The enzyme had an apparent molecular weight of 85 Kda as determined by SDS-gel electrophoresis. -Xylosidase showed maximum activity at pH 6.5 and temperature 40 C. It was stable at pH 6.0-8.0. Heat treatment to 45 C for 1 h did not cause any inhibition for enzyme activity. It was stable for 5 months at 4 C. It was also stable against freezing, thawing and lyophilization with 5.65 per cent loss per cycle. It hydrolyzed -nitrophenyl- -D-xylopyranoside readily with a Km. of 1.1 mM. I2, and HgCl2 were the most potent inhibitors for -xylosidase activity. Xylose was inhibitory towards the enzyme and the inhibition constant was 5.2 mM. The activity of -xylosidase and xylanase acting together was greater than the sum of the individual activities, indicating the synergism between St. flavogriseus xylanolytic components in the hydrolysis of insoluble hemicelluloses.

Abstract: Ethanol produced from lignocellulosic biomass is a second-generation biofuel which does not compete with food resources (Sims et al. 2010) and is expected to be an alternative to gasoline that reduces dependence on fossil fuels. Bioethanol can be produced from lignocellulosic biomass via several processes; pretreatment, enzymatic hydrolysis, and fermentation. In these processes, effective enzymatic hydrolysis of cellulose and hemicellulose is the most important step. Therefore, we focused on the enzyme activities that catalyze the saccharification of cellulose and hemicellulose.Cellulose is the primary component of lignocellulosic biomass. After cellulose, hemicellulose is the second most abundant component of the plant cell wall, and accounts for 20-30% of lignocellulosic biomass (Girio et al. 2010). Glucuronoxylans (O-acetyl-4-O-methylglucuronoxylan) are the most abundant type of hemicellulose, and they make up 15-30% of the dry mass in hardwoods (Girio et al. 2010). Although conventional ethanol fermenting yeasts cannot utilize xylose, we have developed efficient xylose-fermentable Saccharomyces cerevisiae strains (Matsushika et al., 2009). Hence, effective saccharification of xylan is practically important for attaining higher yields of monosaccharides.Endo-β-1, 4-xylanase and β-xylosidase catalyze the production of xylooligosaccharides from xylan and xylose from xylooligosaccharides, respectively. β-xylosidase hydrolyzes the non-reducing end of xylooligosaccharides. Although purified β-xylosidase is commercially available, it is too costly for practical large-scale applications.
