Arf4 Is Required for Mammalian Development but Dispensable for Ciliary Assembly  [PDF]

Abstract: The primary cilium is a sensory organelle, defects in which cause a wide range of human diseases including retinal degeneration, polycystic kidney disease and birth defects. The sensory functions of cilia require specific receptors to be targeted to the ciliary subdomain of the plasma membrane. Arf4 has been proposed to sort cargo destined for the cilium at the Golgi complex and deemed a key regulator of ciliary protein trafficking. In this work, we show that Arf4 binds to the ciliary targeting sequence (CTS) of fibrocystin. Knockdown of Arf4 indicates that it is not absolutely required for trafficking of the fibrocystin CTS to cilia as steady-state CTS levels are unaffected. However, we did observe a delay in delivery of newly synthesized CTS from the Golgi complex to the cilium when Arf4 was reduced. Arf4 mutant mice are embryonic lethal and die at mid-gestation shortly after node formation. Nodal cilia appeared normal and functioned properly to break left-right symmetry in Arf4 mutant embryos. At this stage of development Arf4 expression is highest in the visceral endoderm but we did not detect cilia on these cells. In the visceral endoderm, the lack of Arf4 caused defects in cell structure and apical protein localization. This work suggests that while Arf4 is not required for ciliary assembly, it is important for the efficient transport of fibrocystin to cilia, and also plays critical roles in non-ciliary processes.
Arf4 Determines Dentate Gyrus-Mediated Pattern Separation by Regulating Dendritic Spine Development  [PDF]

Abstract: The ability to distinguish between similar experiences is a critical feature of episodic memory and is primarily regulated by the dentate gyrus (DG) region of the hippocampus. However, the molecular mechanisms underlying such pattern separation tasks are poorly understood. We report a novel role for the small GTPase ADP ribosylation factor 4 (Arf4) in controlling pattern separation by regulating dendritic spine development. Arf4+/? mice at 4–5 months of age display severe impairments in a pattern separation task, as well as significant dendritic spine loss and smaller miniature excitatory post-synaptic currents (mEPSCs) in granule cells of the DG. Arf4 knockdown also decreases spine density in primary neurons, whereas Arf4 overexpression promotes spine development. A constitutively active form of Arf4, Arf4-Q71L, promotes spine density to an even greater extent than wildtype Arf4, whereas the inactive Arf4-T31N mutant does not increase spine density relative to controls. Arf4′s effects on spine development are regulated by ASAP1, a GTPase-activating protein that modulates Arf4 GTPase activity. ASAP1 overexpression decreases spine density, and this effect is partially rescued by concomitant overexpression of wildtype Arf4 or Arf4-Q71L. In addition, Arf4 overexpression rescues spine loss in primary neurons from an Alzheimer's disease-related apolipoprotein (apo) E4 mouse model. Our findings suggest that Arf4 is a critical modulator of DG-mediated pattern separation by regulating dendritic spine development.

Abstract: ????在相同条件下将番鸭、樱桃谷鸭和番樱半番鸭饲养至10周龄,测定3种鸭屠宰性能、胸肌营养成分和常规肉品质。结果表明:各周龄樱桃谷鸭、半番鸭体重显著高于番鸭,10周龄半番鸭、樱桃谷鸭和番鸭体重分别为2446.4、3496.5和3440.0g;10周龄樱桃谷鸭屠宰率、半净膛率显著高于半番鸭和番鸭,其瘦肉率最高,半番鸭次之,番鸭最低;樱桃谷鸭和番鸭皮脂率显著高于半番鸭,番鸭腹脂率显著高于樱桃谷鸭和半番鸭;半番鸭胸肌蛋白质含量显著高于番鸭和樱桃谷鸭,樱桃谷鸭胸肌脂肪含量高于番鸭和半番鸭,半番鸭胸肌水分含量显著低于番鸭和樱桃谷鸭,番鸭胸肌剪切力显著高于樱桃谷鸭和半番鸭。

Abstract: ???日遗化武是中日两国的重大历史遗留问题。1945年日本战败投降后,侵华日军将大量化学武器掩埋在中国。迄今为止,已在中国大陆19个省(市、自治区)发现日遗化武,其主要种类有化学炮弹、化学航弹、毒气筒以及散装毒剂桶等,装填毒剂的类型主要包括糜烂性毒剂、刺激性毒剂、窒息性毒剂、全身中毒性毒剂等。为尽快清除日遗化武,减少毒害,中日两国政府组织力量进行了大量的探测、挖掘、回收和销毁等相关作业。目前,中日双方已挖掘回收日遗化武5万余枚(件)。销毁日遗化武的工作也于2010年10月在南京正式启动。处理日遗化武作业的医学保障要点:一是妥善处理新发现日遗化武伤人事件,二是切实做好突发事件应急医学救援,三是积极做好日遗化武作业现场日常医疗保障。

Abstract: ????以法国番鸭为父本,分别以樱桃谷鸭、金定鸭和樱桃谷鸭与金定鸭杂交后代为母本,采用人工授精的方法生产番樱、番金和番樱金半番鸭。测定3种半番鸭的生长性能、屠宰性能、胸腿肌和肝脏营养成分。结果表明:番樱半番鸭10周龄体重、饲料转化率明显优于其他两个组合,番樱金半番鸭其次,番金半番鸭最低。番樱半番鸭屠体重、全净膛重高于番樱金、番金半番鸭。番樱金半番鸭屠宰率、全净膛率最高,番金半番鸭其次,番樱半番鸭最低。番樱杂交鸭胸、腿肌肉中水分和粗脂肪含量高于番金和番樱金半番鸭。
Molecular Cloning, Sequence Identification and Tissue Expression Profile of Three Novel Sheep (Ovis aries) Genes-ARF3, ARF4 and ARF5

Abstract: The complete coding sequences of three sheep genes-ARF3, ARF4 and ARF5 were amplified using the Reverse Transcriptase Polymerase Chain Reaction (RT-PCR). The nucleotide sequences of these three genes revealed that sheep ARF3 gene encodes a protein of 181 amino acids that shares high homology with the ADP-Ribosylation Factor 3 (ARF3) proteins of five species human (99%), zebrafish (99%), African clawed frog (99%), pig (99%) and atlantic salmon (98%). The sheep ARF4 gene encodes a protein of 180 amino acids that shares high homology with the ADP-Ribosylation Factor 4 (ARF4) proteins of third species-cattle (100%), platypus (99%), green anole (99%), pig (99%), rabbit (98%), African clawed frog (98%), red jungle fowl (98%), mouse (98%), zebrafish (96%), human (97%), atlantic salmon (92%), rainbow trout (92%) and Caligus rogercresseyi (89%). The sheep ARF5 gene encodes a protein of 180 amino acids that shares high homology with the ADP-Ribosylation Factor 5 (ARF5) proteins of seven species human (100%), mouse (100%), rat (100%), gray short-tailed opossum (99%), chicken (98%), channel catfish (93%) and zebrafish (93%). Finally, these three novel sheep genes were assigned to GeneIDs; 100302303, 100302304 and 100302305. The phylogenetic analysis indicated that the sheep ARF3 gene has closer genetic relationship with the ARF3 gene of human. The sheep ARF4 gene has closer genetic relationship with the ARF4 gene of cattle and the sheep ARF5 gene has closer genetic relationship with the ARF5 genes of human, mouse and rat. Tissue expression profile analysis was also carried out and results demonstrated that sheep ARF3, ARF4 and ARF5 genes were differentially expressed in detected tissues.

Abstract: 对商品肉用香鸭和北京鸭的胸部肌肉成分进行了对比测定,结果表明,北京鸭带皮胸肌的脂肪含量为273.9g/kg,番鸭为101.8g/kg,去皮胸肌中北京鸭的脂肪含量为62.6g/kg,番鸭为24.2g/kg,去皮胸肌的蛋白质组成,在所测定的9种必需氨基酸中,番鸭有7种必需氨基酸的含量高于北京鸭,1种基本相同,只有苯丙氨酸的含量较低。

Abstract: 目的:建立测定牛黄解毒丸中番泻苷A和番泻苷B含量的方法。方法:AgilentZorbaxSB-C18色谱柱(4.6mm×250mm,5μm),流动相乙腈-0.05%磷酸水,流速1mL·min-1,检测波长280nm。结果:番泻苷A和番泻苷B的分离度良好(R>1.5),2条标准曲线在检测范围内均呈良好线性(r>0.9999),番泻苷A和番泻苷B的检测限均低于2.40ng、定量限均低于6.40ng,高、中、低3个水平日内和日间精密度的RSD均小于3.2%,加样回收率分别高于为93.5%和92.6%。应用所建立的方法测定了10批购自不同地区的牛黄解毒丸中以上番泻苷A和番泻苷B的含量,其结果差别较大。结论:本研究所建立的牛黄解毒丸中番泻苷A和番泻苷B同时定量的方法准确、可靠,可作为牛黄解毒丸中番泻苷A与番泻苷B的含量测定方法,为该药的全面质量评价提供参考。
