昆虫病原线虫对小地老虎的致病力测定及防治效果

Abstract: 为探讨应用昆虫病原线虫防治小地老虎技术的可行性, 采用培养皿滤纸法比较9个品系的昆虫病原线虫对小地老虎幼虫的致病力, 同时通过盆栽试验验证了斯氏线虫属小卷蛾斯氏线虫Steinernema carpocapsae NC116品系、芫菁夜蛾斯氏线虫S. feltiae SF-SN品系和异小杆线虫属嗜菌异小杆线虫Heterorhabditis bacteriphora H06品系的保苗效果和防治效果, 并测定了苦参碱与NC116品系混用对小地老虎3龄幼虫的联合作用效果。结果表明, NC116品系对小地老虎3龄幼虫致病力最高, 其对3龄、4龄和5龄的LD50分别为2.4、7.5、和31.2条/头, 表明随小地老虎幼虫龄期递增, 其致病力下降。盆栽玉米苗上分别施用线虫100~200条/头时, 对玉米保苗效果及对小地老虎3龄幼虫的控制效果依次为NC116 >H06 >SF-SN。0.8 mg/L苦参碱和NC116品系混用后, 可使小地老虎3龄幼虫死亡率提高109.89%, 二者表现增效作用。
To evaluate the potential control effects of nine entomopathogenic nematodes (EPN) strains against Agrotis ypsilon Rottemberg larvae, the filter paper culture method was used to determine their pathogenicity against the larvae of A.ypsilon under laboratory conditions. Protective effects upon seedlings and control efficiency of three EPN strains (Steinernema carpocapsae NC116, S. feltiae SF-SN and Heterorhabditis bacteriphora H06) were also studied by pot experiments. Meanwhile, the combined effects of nematodes strain NC116 and different concentrations of matrine to the third instar larvae were determined. The results showed that strain NC116 had the highest pathogenicity to the third instar larvae of A. ypsilon, and their pathogenicity decreased as the larval instar increased: strain NC116 LD50 values to the third, fourth and fifth instar larvae were 2.4, 7.5, and 31.2 IJs/larva, respectively. Pot experiments showed that the protective effects for seedlings and the control efficacy at the dosage of 100-200 IJs/larva were in the following order: NC116>H06>SF-SN. Mortality of A. ypsilon was raised by 109.89% under the mixed application of 0.8 mg/L matrine and nematode strain NC116, which showed a synergistic interaction.

Abstract: V-ATP酶是一种ATP依赖型的多亚基质子泵,广泛存在于多种生物的内膜和质膜上,参与生物体内多种重要的生理生化反应,为了进一步研究V-ATP酶,克隆小地老虎V-ATP酶A亚基基因。以鳞翅目昆虫小地老虎(Agrotis ypsilon)的中肠组织为材料,利用RACE技术克隆小地老虎V-ATP酶A亚基基因全长。结果表明:克隆得到A亚基基因序列2 693 bp,其中CDS区1 851 bp,编码616个氨基酸,翻译的蛋白质分子质量为68.06 ku,等电点为5.14,含有3个N-糖基化位点。基因同源性分析结果说明该基因与棉铃虫(Helicoverpa armigera)、烟草天蛾(Manduca sexta)、小菜蛾(Plutella xyllostella)均有较高的同源性。A亚基基因序列已提交至GenBank并获得登录号KM434187。
V-ATPase is a multisubunit proton pump relying on hydrolysis ATP and widely exists in many kinds of biological membrane and plasma membrane, and it participates in several important organism physiological and biochemical reactions. To make a further research on V-ATPase , this research aimed at clone the gene of V-ATPase subunit A of Agrotis ypsilon .Total RNA was isolated from the Agrotis ypsilon. Using the RT-PCR and RACE technologies, the V-ATPase subunit A was cloned from the midgut of A.ypsilon.The whole length is 2 693 bp, CDS region was 1 851 bp, coding a protein of 616 amino acid residues, with a predicted molecular mass of 68.06 ku and PI 5.14, and there were three N-glycosylation sites among the gene. The analysis of the system evolution of subunit A showed that it had high homology with H.armigera,M.sexta and P.xylostella,and the sequence had been deposited in GenBank with accession number KM434187.

Abstract: 采用高效液相色谱技术研究了苦皮藤素Ⅴ在粘虫Mythimna separata和小地老虎Agrotis ypsilon幼虫中的穿透及代谢。结果表明:苦皮藤素V均不能从粘虫和小地老虎幼虫的体壁穿透到血腔或从血腔穿透到中肠,但很容易从中肠穿透到血腔,且穿透速率差异;苦皮藤素V在小地老虎幼虫体内的代谢解毒速率远大于其在粘虫幼虫体内的代谢速率,其半衰期分别为5.5和13.1 h。本研究结果表明,苦皮藤素V对粘虫和小地老虎幼虫的选择毒杀作用与药物的穿透能力关,其在试虫体内的解毒代谢差异才是其对昆虫具有选择作用的机理之一。
Penetration and metabolism of celangulin V in the larvae of Mythimna separata and Agrotis ypsilon were studied using high performance liquid chromatography. The results showed that celangulin V can not penetrate into the haemolymph from the body wall or penetrate into the midgut from the haemolymph in both of larvae. Whereas, celangulin V with the same rate of penetration is easily penetrate into the haemolymph from the midgut in the case of both larvae. The metabolic rate of celangulin V in the larvae of A.ypsilon is greater than that in the larvae of M. separata and the half-life of celangulin V was 5.5 h and 13.1 h in the larvae of A.ypsilon and M. separata, respectively. The results indicate that the penetration was not involved in the selective toxicity of celangulin V, whereas, detoxification metabolism in the larvae of A.ypsilon and M. separata was one of the mechanisms of this selective toxicity.

Abstract: This paper deals with the migrations of the black cutworm moth (Agrotis ypsilon Rott.)and the energy source and climative factors. Observation stations were setting up on Baima snow mountain, Yunnan Province, and the throughout the year observed periodically the migratory population dynamics of the black cutworm moth, in 1985 to 1988. 1. We found that these are five mass immigratory on Baima snow moun-tain each year: The first in April; the second in May; the third mass immigratory from June to A.ugust; the fourth in September and October; the fifth in November. 2. The results of the observation of last serveral years showed that the dynamics of the immigratory population had close relation with temperature, humidity, rain, wind and atmospheric pressure. In natural condition, the immigratory density reached peak at 6.1 to 6.8C, air humidity at 72.1 to 78.3%, wind vilocity at 4.8 to 6.5 m/s. a day precipitation at 0.3 to 4.5 mm, and atmospheric pressure 639.5 to 648.6 hectopascal. The results of the observation also showed that the various immigration peak periods of the black cutworm moth had closest correlation with the southwest and north wind of high altitude atmospheric pressure 500 hectopascal airflow court. 3. There are obvious differences in crude fat contents among adults the varions type of population sources. The content of the emigratory type is higher than the local type; the local type is higher than transitionary type; the transilionary type is higher than immigratory type; however, that of the immigratory type is the lowest one. The percentages are 33.4%, 33.2%, 24.2% and 22.0% respectively.

Abstract: 为进一步探讨杠柳新苷类化合物在昆虫体内的代谢机制。比较研究高活性杠柳新苷P(Periploca sepium P,PSP)和杀虫活性的杠柳新苷E(Periploca sepium E,PSE)对6龄粘虫幼虫(Mythimna separata)与小地老虎幼虫(Agrotis ypsilon)中肠3种代谢酶[羧酸酯酶(CarE)、谷胱甘肽-S-转移酶(GST)、细胞色素P450-O-脱甲基酶]活性的影响。结果表明:药后8 h,PSP和PSE使粘虫幼虫GST活性增加,但PSP上升趋势比PSE更高。而2种化合物对试虫CarE活性均明显影响。药后12 h,PSP和PSE使粘虫中肠细胞色素P450-O-脱甲基酶酶比活力显著下降,而小地老虎中该酶活性明显变化。因此,可推测GST可能参与昆虫对杠柳毒素的解毒作用,[JP3]而细胞色素P450-O-脱甲基酶活性被杠柳毒素抑制可能是造成粘虫中毒的机制之一。[JP]
Periplocosides on detoxification enzymes in midgut of larvar has not been reported.In order to reveal the insecticidal activity and the effect of periplocosides on activities of detoxification enzymes in midgut of larvar.We investigated the effects of strong insecticidal periplocoside P (PSP) and periplocoside E (PSE) which had no insecticidal activity on the activities of three metabolism enzymes in midgut of 6th instar larvae of Mythimna separata and Agrotis ypsilon, including carboxylesterase (CarE), glutathione S-transferase (GST) and O-demethylase of cytochrome P450.GST activity could be increased by PSP and PSE when M.separata larvae were administrated orally with both compounds after 8 h, but PSP showed higher rise trend of GST than PSE.Both compounds showed no obvious effect on CarE activity of the tested insects.The activity of O-demethylase of cytochrome P450 on M.separata was significantly declined by PSP and PSE after 12 h, whereas had no obvious changes on A.ypsilon.The cytochrome P450 in the midgut may be relevant to the intoxication mechanism of PSP against M.separata, whereas GST may be involved in detoxification of periplocosides in insects.

Abstract: Se describe e ilustra una nueva especie de Noctuidae: Agrotis leucovenata sp. nov. Esta se distribuye en la estepa patagónica de la provincia de Neuquén, en Argentina. Se discuten los caracteres propios de la nueva especie, que la diferencian de otras especies similares del género. Agrotis leucovenata sp. nov., a new species of Noctuidae, is described and illustrated. This species is distributed in the Patagonian steppe of Neuquén province, Argentina. Characteristic traits of the new species which differentiate it from similar species of the genus are discussed.
