节节麦及人工合成多倍体中Glu—D1位点高分子量谷蛋白亚基组成及表达研究  [PDF]

Abstract: 为了了解节节麦及人工合成多倍体的Glu—D1位点高分子量谷蛋白亚基组成及表达情况,采用十二烷基磺酸钠-聚丙烯跣胺凝胶电泳(SDS-PAGE),分析了4份节节麦及其在此基础上形成的人工合成多倍体高分子量走谷蛋白亚基(high—molecular-weightgluteninsubunit,HMW-GS)的组成。研究结果表明,4份节节走AS60-1、AS60-2、AS77和AS2383在Glu—D1位点上出现了4种不同的亚基类型,分别为210、512、510和2.110;2份节节走人工加倍形成四倍体AS2390、AS2410的HMW—GS分别为2.110和510;3份节节麦-圆锥麦人工合成双二倍体RSP、SHW—L1及SHW—L2的HWM—GS分别为2^*、1718、510;2^*、1718、210和210。谷蛋白王基呈现了共显性遗传,表明节节麦高分子量谷蛋白基因能在人工多倍体情况下得到正常表达。本文还对利用节节麦优良基因的方式作了讨论。
小麦品种周麦22号的分子遗传基础及其特异引物筛选  [PDF]

Abstract: 国审小麦品种周麦22号具有高产、稳产、多抗和广适等突出优点,是目前全国种植面积居于前列的品种,也是优异的育种亲本材料。为研究周麦22号的分子遗传学基础以及筛选周麦22的特异引物,利用覆盖小麦全基因组的340个SSR标记对周麦22号及其亲本周麦12号、温麦6号、周麦13号进行SSR标记分析。结果表明,温麦6号对周麦22号的遗传贡献最大(37.35%),其次是周麦13号(36.14%),周麦12号贡献最小(26.51%) ;周麦22号和其3个亲本间的遗传相似系数的聚类结果与系谱分析不一致,表明在选育过程中亲本遗传物质的传递发生了偏分离;在不同基因组水平上,3个亲本对周麦22号的遗传贡献率差异较明显,在A、B、D三个染色体组上对周麦22号的遗传贡献各有侧重;从340个SSR标记中筛选出28个周麦22号的特异标记,并通过与周麦22号的姊妹系、相似品种、衍生品种及黄淮麦区主推的小麦品种相比较,进一步筛选出1个周麦22号的特异引物Xgwm577,建立了一种能够准确、快速、简便、稳定检测周麦22号品种真实性的检测手段,为周麦22号进一步的遗传改良和推广应用提供了理论参考。

Abstract: BCG Purified Protein Derivative (BCG-PPD)was isolated and purified from BCG Culture filtrate by trichloroacetic acid and ammonium sulfate methods. The purity of BCG-PPD was Similar to PPD-S(international standard) and PPD-C(China), but more than that of PPD-CT68 (Canada)and PPD-RT23(Danish). The Delayed-Type Hyperseusitivity(DTH) to BCG-PPD was more sensitivity than other PPD on BCG vaccinated guinea pigs, but less sensitivity than other PPD on Mycobacterium tuberculosis infected guinea pigs. The conversion rate and induration diameter to BCG-PPD was higher than PPD in 333 of 12 weeks after BCG vaccination newborns, but lower than that of other PPD in 97 tuberculosis patients. It was shown that DTH reaction to PPD was more sensitivity in Mycobacteria homogeneous strain vaccinated individual than Mycobacteria heterogeneous strain vaccinated individual. It was demonstrated that BCG-PPD was better than other PPD on observation conversion rates and induration diameter of BCG vaccinated individual. It maybe help to identification BCG vaccinated or tuberculosis infected with DTH of BCG-PPD and PPD in same individual.
周麦18号小麦生理生化特性及其与产量性状的关系研究  [PDF]

Abstract: 以新麦18号(CK1)和豫麦49号(CK2)为对照,对周麦18号小麦品种生理生化特性及其产量构成进行了研究。结果表明,周麦18号产量构成因素比较协调,其千粒重最高,为48.8g,分别较新麦18号、豫麦49号提高21.1%、32.2%。周麦18号在灌浆期具有较高的叶面积指数、较稳定的净光合速率、较低的冠层温度,灌浆强度达1.28g/(d.千粒),还具有较强的碳、氮代谢活性以及灌浆中后期较强的根系活力,这些特性是周麦18号高产稳产的保证。

栽培措施对周麦23号小麦主要农艺性状及品质特性的影响  [PDF]

Abstract: 为探讨小麦新品种周麦23号的高产优质配套栽培措施,采用三因子二次正交旋转组合设计,研究了播期(X1)、播量(X2)、追肥量(X3)对周麦23号主要农艺性状和品质特性的影响。结果表明,周麦23号的株高主要受播期影响,在10月21日播种时,株高最低(80.7cm),10月10日播种植株最高(90.27cm)。产量主要受播期、播量和追肥量的共同作用,建立的产量模型:Y=10660.49-520.18X1-233.93X3-166.38X22+237.56X2X3,在10月10日播种,216万苗/hm2基本苗,追肥量225kg/hm2时,预测产量将达最大值,为12161.91kg/hm2。播量和追肥量与周麦23号穗数和穗粒数分别表现为二次曲线和线性关系,而千粒重同时受播期、播量和追肥量的共同作用。周麦23号的沉降值、揉混仪峰宽、8min带宽均受播期和追肥量互作影响,和面时间受播期、播量与追肥量的共同作用。建议周麦23号在高肥水地块于10月13日播种,基本苗216万~270万苗/hm2,于拔节期追施尿素225~255kg/hm2,若遇暖冬年份,播种期可适当推迟,增加播量。
The Study Of Booster Effect After Sequential PPD Test

Abstract: Infection by M. tuberculosis can be demonstrated by tuberculin"ntesting. On sequential PPD some person show a marked increase in the size of their"nskin reactions which may not be due to recent or past tuberculous infection. This"nphenomenon (booster effect) can occur as soon as a week after the initial test and"npersist for as long as a year."nMethods and Materials: The purpose of this research is studying the size of"ninduration following repeated PPD tests (booster effect) that studied in children"nwho came to our clinic or imbedded in pediatric unit and the group of student in"ngrades 4 and 5 at elementary school (totally 80 cases) for one year."nResults: Increase of induration in 25% of all (20 cases) was 5 mm or more and in"n75% of all (60 cases) was less than 5 mm and between these two groups,"ndifferences was meaning full (P< 0.05). The increase of induration in 12.5% was 7"nmm or more."nConclusion: Because there is not shown definite increase of induration (more than"n5 mm) on sequential PPD in children, we must to consider to new infection.
小麦品种百农AK58与周麦18的遗传差异性分析  [PDF]

Abstract: 为解析百农AK58和周麦18的遗传差异,利用表型数据分布于小麦全基因组的305个SSR标记进行分析,以期为小麦大面积种植品种的培育提供参考。方差分析表明,百农AK58和周麦18在株高、千粒质量和单位面积穗数上存在极显著差异,其中百农AK58在株高和单位面积穗数上明显优于周麦18。分子标记比较发现,305对SSR引物中有130对引物在百农AK58与周麦18之间扩增出差异带,多态引物比例达到42.6%。2个品种在A、B、D基因组存在差异的引物数基本一致,分别有34,37,37个,多态引物比例表现为B基因组(43.9%)>A基因组(37.1%)>D基因组(33.5%)。百农AK58和周麦18的遗传差异在小麦7个部分同源群和21对染色体上也表现一定的不均衡性。在7个部分同源群之间,二者在第5部分同源群的遗传差异最大,多态引物比例达到49.3%,而在染色体水平上,它们在3B、5B、7B、4A的遗传差异相对较大,多态性引物比例均超过50%。百农AK58和周麦18所不同的SSR位点是遗传研究中需要重点关注的基因组区域。

Abstract: 为了研究强筋优质小麦新品种周麦24号的配套栽培措施,采用四因子二次正交旋转组合设计,研究了播期、密度、追肥期、多效唑对周麦24号产量和品质特性的影响。结果表明,产量主要受播期和追肥期的共同影响,与播期、追肥期呈线性关系。蛋白质含量受播期、密度、追肥期和多效唑共同影响;面筋指数、揉混仪参数8min峰高主要受播期的显著影响。建立了产量模型:Y=554.85-49.07X1-22.55X3,预测产量最大值为10129.5kg·hm-2。综合分析认为,周麦24号在10月10日左右播种、基本苗210万·hm-2、2月20日左右追肥、不喷施多效唑的条件下,产量和品质较好。

Abstract: Tuberculosis (TB) is one of the most important global health problems. Recently the increased rate of tuberculosis-HIV co-infection and formation of multidrug-resistant strains of M. tuberculosis have led to changes in vaccination policies and progression in vaccination trials including BCG and new TB vaccines. BCG is one of the safest and most widely used vaccines worldwide. BCG is produced by passaging from the M.bovis and has been available for more than 80 years During passaging, molecular changes may occur in the genome of BCG strains. There are a number of BCG vaccines produced by different countries. Vaccination policy differs between countries. A wide range of protective efficacy has been reported for BCG vaccine in studies. However it is well known that it protects against the severe form of TB . Among the new BCG vaccines under investigation several are promising but at the phase of clinical trials.their efficacy may change according to the genetic make up of the individual, previous BCG vaccinations and the time and number of previous PPD applications, which may cause changes in response to the PPD test.
