Substratos na aclimatiza o de Pfaffia glomerata (Spreng) Pedersen produzida in vitro sob diferentes doses de sacarose  [cached]

Abstract: Este trabalho teve como objetivo selecionar substratos para a aclimatiza o de plantulas de Pfaffia glomerata produzidas in vitro sob diferentes concentra es de sacarose. Os tratamentos consistiram de uma combina o bifatorial (5x3) entre cinco doses de sacarose (15, 30, 45, 60 e 75g L-1), presentes no meio de cultura in vitro, e três substratos [Plantmax Hortali as, Plantmax + Solo (1:1 v/v) e Vermiculita (granulometria média) + Solo (ARGISSOLO VERMELHO Distrófico arênico) (1:1 v/v)] utilizados na aclimatiza o ex vitro. Foram realizadas determina es das características físicas e químicas dos substratos, bem como avalia es do crescimento e da sobrevivência das plantas tanto durante o cultivo in vitro como no ex vitro. Plantas provenientes do cultivo in vitro na presen a de 45 a 60g L-1 de sacarose apresentaram melhor aclimatiza o ex vitro. As combina es dos substratos Vermiculita + solo (1:1 v/v) e Plantmax + solo (1:1 v/v) proporcionaram maior crescimento às plantas durante a última fase de aclimatiza o (cultivo sob sombrite), provavelmente devido a possuírem maior porosidade total. Entretanto, o uso isolado de Plantmax aumentou a sobrevivência das plantas cultivadas a campo, fato relacionado a esse substrato possuir os maiores valores de capacidade de reten o de água, de água facilmente disponível e de água disponível.

Abstract: this work was aimed at selecting substrates on the ex vitro acclimatization of pfaffia glomerata produced in vitro under different sucrose levels. the treatments consisted of a bifactorial combination (5x3) between five sucrose levels (15, 30, 45, 60, and 75g l-1), present in the in vitro culture, and three substrates [plantmax?; plantmax? + soil (1:1 v/v), and vermiculite (middle size) + soil (paleudalf) (1:1 v/v)] used in the ex vitro acclimatization steps. physical and chemical evaluations of the substrates were carried out as well as evaluations of plant growth and survival for both in vitro and ex vitro cultures. plants obtained from in vitro cultures in the presence of 45 to 60g l-1 sucrose showed better ex vitro acclimatization. the substrate combinations vermiculite + soil and plantmax? + soil gave rise greater growth to plants during the last acclimatization step (partially shady natural environment), probably due to having greater total porosity. however, the isolated use of plantmax? increased the plant survival under field conditions because this substrate showed the greatest values for water retention capacity, easily available water and available water.

Abstract: the present research was aimed at evaluating the growth of brazilian ginseng (pfaffia glomerata (spreng.) pedersen) in vitro cultured plantlets under two levels of nitrogen and sucrose, during six successive subcultures and acclimatization. two nutritional treatments were tested: nitrogen at 100% strength of the ms medium + 30g l-1 sucrose; and nitrogen at 50% strength + 45g l-1 sucrose. during six successive subcultures, the explants of 30-day-old plantlets grown under both treatments were transferred, respectively to the next subculture on the same treatment conditions. at 6th subculture, plantlets were transferred to substrate plantmaxa hortali?as and submitted to acclimatization in a growth chamber. throughout the subcultures, fluctuations in growth response due to nutritional conditions tested were observed. the height of the higher sprout, the average height of sprouts, and the total number of nodal segments per plantlet, respectively for four, four and three subcultures were greater in 100% strength of the ms medium + 30g l-1 sucrose than in 50% strength + 45g l-1 sucrose. on the other hand, dry matter of roots, of shoots and of the whole plant, respectively for two, two and three subcultures, were greater in 50% strength + 45g l-1 sucrose. in conclusion, the increase of sucrose associated with reduction of n level in the culture medium enhanced the biomass production, while the ms at full strength enhanced the growth based in sprout length and number of nodal segments. these differences concerning the biomass production observed during in vitro culture did not persisted in the acclimatization process ex vitro, where ms at full strength give rise greater growth in sprout length and number of leaves per plant.

Abstract: Pfaffia glomerata (Spreng.) Pedersen, conhecida como ginseng brasileiro, é uma planta extensivamente usada na medicina popular em decorrência de possuir propriedades fitoterápicas. Este trabalho teve como objetivo determinar a melhor combina o de concentra o da sacarose (30, 45 e 60g L-1) e do nitrogênio (50, 75, 100 e 125% da concentra o padr o do meio MS) para a multiplica o in vitro de P. glomerata. Aos 30 dias de cultivo, verificou-se que concentra es entre 40 e 45g L-1 de sacarose e 50% de N propiciaram maior crescimento em altura e número de segmentos nodais por plantula. O número de brota es foi maior na concentra o de 55g L-1 de sacarose combinada com 70% de N. A matéria seca de raízes, da parte aérea e do total da plantula foi maior na concentra o de 45g L-1 de sacarose associada com 50% de N. No geral, a redu o da concentra o de N para 50% daquela padr o do meio MS, associada a um incremento na dose de sacarose para 45g L-1, favorece o crescimento em altura, número de segmentos nodais e brota es, bem como a produ o de biomassa de P. glomerata cultivada in vitro, devido ao estímulo ao uso do carbono.

Abstract: pfaffia glomerata (spreng.) pedersen, known as brazilian ginseng, is an extensively used plant in folk medicine due to its phytotherapic characteristics. this work aimed to determine the best combination of sucrose (30, 45 and 60 g l-1) and nitrogen (50, 75, 100 and 125% of the strength of ms medium) on micropropagation of p. glomerata. in 30 days of cultivation, sucrose ranging from 40 to 45g l-1 and nitrogen at 50% increased the height and the number of nodes. the number of shoots was greater at concentration of 55g l-1 sucrose combined with 70% n. the dry matter of roots, aerial parts, and of the whole seedling was increased on 50% n and 45g l-1 sucrose. altogether the halved concentration of nitrogen and the increased concentration of sucrose to 45g l-1 increase the height, the number of nodes and the shoots as well as biomass production improved due to an enhancemednt of carbon use.

Abstract: pfaffia glomerata (spreng.) pedersen, known as brazilian ginseng, is a plant extensively used in folk medicine due to its phytotherapic characteristics. the aim of this study was to evaluate the presence of β-ecdysone in the roots and shoots of in vivo and in vitro cultured plants of two sources (bra and jb-ufsm) of pfaffia glomerata using thin-layer chromatography (tlc). for in vivo cultured plants, tlc plates showed that β-ecdysone was present in the roots and shoots. sources showed no differences concerning tlc plates. tlc analysis did not detect β-ecdysone in the in vitro cultured plants.

Abstract: the plant pfaffia glomerata (spreng.) pedersen, known as brazilian ginseng, is extensively used in folk medicine due to its phytotherapic characteristics. this work aimed to evaluate the effect of bap and tdz concentrations on the in vitro propagation of two sources (bra and jb-ufsm) of this species. nodal segments originated from aseptically grown plants were cultured on murashige and skoog (ms) medium supplemented with 0, 1 and 5 μm bap or tdz. after 30 days, the plants were transferred to ms medium without cytokinin. the in vitro organogenesis of p. glomerata is genotype-dependent. the source bra had greater potential for in vitro propagation than jb-ufsm. the culture of bra nodal segments on medium with 1 μm tdz, followed by subcultivation of sprouts on cytokinin-free medium, showed to be a viable method for in vitro propagation due to high multiplication rate and good plant development. in the source jb-ufsm, the culture of nodal segments on medium without cytokinin was the best method to obtain a large number of high-quality plants.

Abstract: pfaffia glomerata occurs in several states of brazil and its neighboring countries in the south region at riverbanks and gallery forests. it is a hydrophyte and heliophyte species. the roots of the genus pfaffia are used in brazilian folk medicine especially as tonic, aphrodisiac and to control diabetes. the aim of this work was to establish an in vitro germplasm bank for pfaffia glomerata. the experiment was carried out in completely randomized design with six treatments: 1) dm + 2% sucrose + 4% sorbitol; 2) dm/2 + 2% sucrose + 4% sorbitol; 3) dm + 2% sucrose + 4% sorbitol + 2 mg l-1 calcium pantothenate; 4) dm/2 + 2% sucrose + 4% sorbitol + 2 mg l-1 calcium pantothenate; 5) dm + 2% sucrose + 3% mannitol + 2 mg l-1 calcium pantothenate; 6) dm/2 + 2% sucrose + 3% mannitol + 2 mg l-1 calcium pantothenate. results were subjected to analysis of variance and scott knott test for mean grouping. treatments 1, 3 and 4 had a significantly larger number of nodal segments per stem, compared to treatments 2, 5 and 6. treatment 2 was the most appropriate for the in vitro conservation of this species since it led to the lowest growth (3.1±1.9 cm height), high survival rate, 100% explants with sprouting, and the largest number of sprouts per explant after six months of culture. all seedlings produced root and showed no formation of calluses or hyperhydricidity under the evaluated treatments. acclimatized plants showed 100% survival in the ex vitro environment. maintaining p. glomerata accessions in an in vitro germplasm bank is viable both economically and for conservation.

Abstract: brazilian ginseng [pfaffia glomerata (spreng.) pedersen] has expressionable medicinal properties, and therefore, nowadays it is extensively exploited either by the herb collectors as well as the pharmaceutical industry. the aim of this work was to characterize the effect of n and p concentrations from the ms medium on growth of p. glomerata in vitro cultured plants. 1-node segment without leaves, from in vitro cultivated plants, were grown on five levels (0, 25, 50, 100, and 150% of the strength of the ms medium) of n and p. after 15 days of inoculation, root number and rooting percentage are greater on n and p levels of the ms medium with 50% strength. after 40 days of inoculation, growth, based on sprout height, number of nodal segments per plant, leave area index per plant, number of leaves per plant, dry weight of aerial parts, roots and of the whole plant, is greater when the n and p levels of the ms medium is near to 80% strength.

Abstract: pfaffia glomerata (spreng.) pedersen is one of the brazilian ginseng species used as medicinal plant. the aim of this work was to evaluate the effect of carbohydrate concentration (15, 30, 45, and 60 g.l-1) and source (sucrose, fructose, glucose, maltose, and lactose) on the growth of plants grown in vitro. an entirely random statistical design with 15 replications per treatment was used. the experimental unity consisted of a test tube containing 10 ml of ms medium and 1-node segment of 1,0cm length and without leaves. after 41 days of the inoculation the growth parameters analyzed were sprout number, total number of nodal segment per seedling, average length of sprouts, length of the highest sprout, fresh and dry matter weight of aerial parts, and root dry matter weight. the best carbohydrate source for growth of p. glomerata in vitro cultured plants was sucrose, and lactose was found unsuitable. the total number of nodal segment per seedling, average length of sprouts, length of the highest sprout, fresh and dry matter weight of aerial parts, and root dry matter weight increase by sucrose concentrations, and at 30, 45 and 60 g.l-1 all parameters were significantly higher than those obtained by other carbon sources.
