Evaluation of the in vivo mutagenic potential of hydroalcoholic extracts of the northern highbush blueberry (Vaccinium corymbosum L. Ericales, Ericaceae) on peripheral blood cells of Swiss mice (Mus musculus Rodentia, Muridae)

Abstract: the northern highbush blueberry (vaccinium corymbosum l. ericales, ericaceae) is very rich in anthocyanins, natural pigments which have strong antioxidant properties and potential health benefits, resulting in the worldwide use the blueberry as a medicinal plant. we investigated the mutagenic potential of simple hydroalcoholic extracts of v. corymbosum acutely administrated by gavage to swiss mice at doses of 1 g kg-1, 1.5 g kg-1 and 2 g kg-1. peripheral blood cells were collected 4 h and 24 h post-gavage and assessed by the alkaline comet assay, with further blood samples being collected at 48 h and 72 h for assessment using the micronucleus (mn) assay. our results show that the v. corymbosum extracts did not induce any statistically significant increase in the average amount of dna damage in peripheral blood leukocytes. however, we did record a significant increase in the frequency of micronucleated polychromatic erythrocytes at the three doses tested.

Abstract: we evaluated the genotype and maturity effects on antioxidant activity and phenolic compounds of whole, skin and pulp fruits from three highbush blueberry cultivare (cv. brigitta, cv. bluegold and cv. legacy) grown in southern chile. total antioxidant activity (taa) in ripe fruits varied among the cultivare in the order legacy > brigitta > bluegold. we found that taa in unripe green and fully ripe fruits was high and similar between them, whereas the lowest levels were found in intermediate ripe fruits. the same trend was observed for fruit total phenolic content. this could be attributed to the higher concentrations of phenolic acids (mainly chlorogenic acid) and flavonols (mainly rutin) at immature fruit stages; whereas the high taa in mature fruits could be explained by the elevated amounts of anthocyanin. all antioxidant compounds were mostly located in the skin. high amounts of delphinidin aglycone were found. hplc-dad/ms revealed that the main contents of skin anthocyanins are petunidin-3-glucoside and petunidin-3-arabinnoside followed by malvidin-3-galactoside. it is noticeable that highbush blueberry fruits grown in southern chile have exceptionally higher antioxidant activity and anthocyanins contents compared with those cultivated in the northern hemisphere.

Abstract: La producción comercial de arándanos en el país ha sufrido modificaciones que apuntan hacia garantizar el cumplimiento de las normas de calidad impuestas por los países importadores. Con el objetivo de evaluar el efecto de distintas fuentes de calcio sobre la calidad de frutos de arándano alto, en la temporada agrícola 2007/2008, se realizaron aplicaciones de calcio al suelo y foliar en precosecha en plantas de arándano alto (Vaccinium corymbosum L.) cultivar Elliott. El estudio se realizó en un huerto comercial localizado en la comuna de Río Negro, Región de Los Lagos. El suelo correspondió a un andisol de la serie Corte Alto. El ensayo consideró siete tratamientos y las aplicaciones se realizaron en plantas de 15 a os de edad, desde inicio de cuaja hasta desarrollo de frutos. Se llevaron a cabo diez cosechas donde se evaluaron calibre, peso, sólidos solubles y firmeza de frutos de arándano alto. Los datos obtenidos se sometieron a un análisis de varianza (ANOVA) y prueba de comparación múltiple mediante la prueba de Tukey con un nivel de significancia del 5%. No se produjeron diferencias estadísticamente significativas entre los diferentes tratamientos evaluados para las variables de calibre, peso y sólidos solubles. Generation and analysis of blueberry transcriptome sequences from leaves, developing fruit, and flower buds from cold acclimation through deacclimation

Abstract: Efforts to sequence the transcriptome of the commercial highbush blueberry (Vaccinium corymbosum) cultivar Bluecrop and use the sequences to identify genes associated with cold acclimation and fruit development and develop SSR markers for mapping studies are presented here. Transcriptome sequences were generated from blueberry fruit at different stages of development, flower buds at different stages of cold acclimation, and leaves by next-generation Roche 454 sequencing. Over 600,000 reads were assembled into approximately 15,000 contigs and 124,000 singletons. The assembled sequences were annotated and functionally mapped to Gene Ontology (GO) terms. Frequency of the most abundant sequences in each of the libraries was compared across all libraries to identify genes that are potentially differentially expressed during cold acclimation and fruit development. Real-time PCR was performed to confirm their differential expression patterns. Overall, 14 out of 17 of the genes examined had differential expression patterns similar to what was predicted from their reads alone. The assembled sequences were also mined for SSRs. From these sequences, 15,886 blueberry EST-SSR loci were identified. Primers were designed from 7,705 of the SSR-containing sequences with adequate flanking sequence. One hundred primer pairs were tested for amplification and polymorphism among parents of two blueberry populations currently being used for genetic linkage map construction. The tetraploid mapping population was based on a cross between the highbush cultivars Draper and Jewel (V. darrowii is also in the background of 'Jewel'). The diploid mapping population was based on a cross between an F1 hybrid of V. darrowii and diploid V. corymbosum and another diploid V. corymbosum. The overall amplification rate of the SSR primers was 68% and the polymorphism rate was 43%.These results indicate that this large collection of 454 ESTs will be a valuable resource for identifying genes that are potenti

Abstract: 为探讨不同温度对北高丛蓝莓品种叶片气孔特征及其气体交换参数的影响机理,以2年生北高丛蓝莓(Vaccinium corymbosum L.) ‘蓝丰’、‘公爵’ 和 ‘布里吉塔’ 幼苗为试验材料,利用人工气候箱设置4个温度处理,即对照(25℃)、轻度高温(30℃)、中度高温(35℃)和重度高温(40℃),研究不同温度对北高丛蓝莓叶片气孔的结构和功能、空间分布格局及气体交换的影响。结果显示,高温显著提高‘布里吉塔’叶片的气孔密度,但却没有改变‘公爵’和‘蓝丰’叶片的气孔密度。同时,中度高温增加‘蓝丰’和‘布里吉塔’叶片气孔的长度、宽度和面积,但是对‘公爵’叶片气孔开度的影响不大;且高温导致‘布里吉塔’气孔在叶片上的空间分布变得比对照更加规则,而对‘公爵’和‘蓝丰’2个品种的影响却很有限。此外,不同强度的温度处理导致3个北高丛蓝莓品种叶片的净光合反应速率、气孔导度、蒸腾速率和水分利用效率均呈现先升高后降低的趋势,但其最大值随着品种的不同而发生变化。高温条件下北高丛蓝莓主要通过增加气孔的宽度和规则化气孔的空间分布提高光合效率。然而,不同蓝莓品种对叶片气孔结构和功能进行调整和优化的能力存在一定的差异,最终导致蓝莓叶片气体交换对增温的响应存在种间差异。
In order to investigate the effects of temperature on the leaf stomatal structural and function, stomatal distribution pattern, and gas exchange of north highbush blueberry, we examined the changes in leaf stomatal structure, stomatal size, stomatal density, stomatal distribution pattern, and gas exchange of three north highbush blueberry cultivars (‘Bluecrop’, ‘Duke’, and ‘Brigitta’) with four growth chambers controlling different temperatures including control (25℃), mild high temperature (30℃), moderate high temperature (35℃), and severe high temperature (40℃).The results showed that high temperature significantly increased the stomatal density of ‘Brigitta’, but had little effect on the stomatal density of ‘Duke’ and ‘Bluecrop’.Meanwhile, moderate high temperature substantially increased the stomatal aperture width, stomatal aperture length, and stomatal area of ‘Brigitta’ and ‘Bluecrop’, whereas the stomatal aperture size of ‘Duke’ was barely changed.In addition, high temperature resulted in a more regular stomatal distribution pattern on the leaves of ‘Brigitta’, while had little effect on the stomatal distribution pattern of ‘Duke’ and ‘Bluecrop’.Moreover, mild and moderate high temperature significantly increased the net photosynthetic rates (Pn), stomatal conductance (Gs), transpiration rates (Tr), and water use efficiency (WUE) of the three blueberry cultivars, whereas the severe high temperature caused them sharply decreased with the maximum values varying across the three cultivars.Our results suggested that high temperature increased the photosynthetic efficiency by increasing the stomatal width and regulating the spatial distribution pattern of stomata on the leaves of north highbush blueberry.However, the optimized ability on stomatal structure and function was species dependant, and thus resulted in different responses to high temperature.
The changes of antioxidant properties in highbush blueberries (Vaccinium corymbosum L.) during freezing and long-term frozen storage

Abstract: The aim of this work was to investigate the effect of freezing and long-term frozen storage on total phenolic, total anthocyanin contents and antioxidant activity of highbush blueberries (Vaccinium corymbosum L.). In addition, a HPLC method was developed for the determination of individual anthocyanins and chlorogenic acid contents. Blueberries were frozen at –18°C and –35°C and were stored at these temperature conditions for six months. Derivatives of malvidin and delphinidin accounted for 61% and 22% of total anthocyanin contents in fruit, respectively. Measurements of the antioxidant activity and bioactive compounds contents of blueberries showed there were no significant differences between fresh and frozen fruits. Also, at the end of frozen storage period, antioxidant activity remained significantly unchanged compared with the values measured just after the freezing process.

Abstract: The aim of the present study is to investigate the effect of a hydroalcoholic extract of Vaccinium corymbosum Lam. (highbush blueberry) leaves on plasma and cholesterol levels, on the structure of pancreas and on the ultrastructure of liver, in streptozotocin-diabetic rats. Plasma glucose levels were found to drop by about 30% after 10 and 20 days of treatment with the leaf extract, while plasma cholesterol levels slightly decreased only after 20 days of treatment. There was no evident improvement in the streptozotocin-altered structure of the pancreas after 3 and 6 days of treatment with the extract. The extract was effective in improving the shape and structure of the nucleus of hepatocytes altered by streptozotocin.

Kinetic Study of Convective Drying of Blueberry Variety O?neil (Vaccinium corymbosum L.)

Abstract: the aim of this research was to study and to model the drying kinetics of the blueberry vaccinium corymbosum l.) at three temperatures (60, 70 and 80 oc) with an airflow of 2.0 ± 0.2 m s-1. modeling of the desorption isotherm was carried out with the gab (guggenheim, anderson and de boer) equation, showing a good fit to experimental moisture data, giving as a result a monolayer moisture level of 0.084 g water g-1 dm. newton, henderson-pabis, page, modified page and logarithmic mathematical models were applied in the study and in the modeling of the drying kinetics of this fruit. kinetic parameters k of each model showed dependence on temperature, and were evaluated by an arrhenius-type equation, with an activation energy of between 36.2 and 54.5 kj mol-1. logarithmic and modified page models gave the best fits for each drying curve, based on the statistical test determination coefficient, sum square error, root mean sum errors and chi-square. in consequence, both models are excellent tools for estimating the drying time of this product.
