Karyotype Analysis of Anemarrhena asphodeloides Bunge (Liliaceae)

Abstract: A karyotypical analysis of Anemarrhena asphodeloides Bung. of the monotypic genus Anemarrhena Bung. (Liliaceae) was carried out for the first time. The number of chromo- somes in root-tip cell of the species was found to be 22, agreeing with that reported by Sato12], although inconsistent in some other respects, such as position of centromeres, length of chromosomes, and nucleoli, etc. (Table 1 ). According to the terminology defined by Levan et al.[8], the karyotype formula is therefore 2n=22=2sm (SAT)+2sm+18m. Photomicrographs of the chromosome complements and idiogram of the karyotype are given Fig. 1 and 2). The karyotype of Anemarrhena asphodeloides shows explicitly to be asymmetrical, with three pairs of long chromosomes and eight pairs of short chromosomes. This specialized feature, when considered together with the rare occurrence of the basic chromosome number of 11 of the genus within the Tribe Asphodeleae of Liliaceae (see Table 1), suggests that the genus Anemarrhena is probably a rather specialized one, which has scarcely any intimate relationship with the other genera of the above tribe. The fact that this specialized karyotype is associated with certain trends of morphological specialization, such as flowers possessing three stamens only, gives support to the above suggestion. But, it is impossible to draw a more precise conclusion without a more thorough and comprehensive investigation of the species in question.

Abstract: Onion chromosomes (Allium cepa L.; 2n=2x=16) were analysed for karyotype analysis using aceto-orcein staining technique, and classified due to their lengths from long to short arms whether satellite exists or not.The length of arms appeared to be very close to each other and got more force their identification. The length of long arms showed a variation from 5 to 14 m while short arms have 2 to 10 m at the early and late stages of mitotic metaphase. The reason of this variation comes from the stages which observed as late prophase in mitosis. This different chromosome length also was affected in regard to the satellite position seen on long chromosome. The folding rate changed between 1.01 to 2.16 due to the smallest total length of chromosomes.

Abstract: The C-banded karyotype of Heteranthelium piliferum species was studied in a natural population from northwest of Iran using aceto-iron-hematoxilin staining and C-banding technique. Chromosome measurements including long arm, short arm and chromosome lengths, arm ratio index, relative chromosome length, heterochromatin percent per chromosome and per chromosome set were made. It was revealed that the karyotype of this species is symmetric and consists of 7 pairs of metacentric chromosomes. Arm ratio index values ranged from 1.01 in chromosome G to 1.44 in chromosome D. One of the chromosomes had a satellite located on the end of its long arm (chromosome G). The Q genome of this species like A, B, D, S, M and Mt genomes in diploid species of Aegilops-Trticum group, H genome in Hordeum, E genome in Agropyron and R genome in Secale has metacentric or sub-metacentric chromosomes.

Abstract: Chromosomes, with detailed karyotype information (number, shape, total length, relative length, arm ratio and centromeric index) and C-banding patterns in the somatic division of haploid male of Apis florae in Iran are described. Samples were obtained from the colonies in south of Iran. Prior to the swarming season, drone-brood cells were added adjoining to the lower rows of the worker-brood cells. Testes from young larvae were removed, fixed in acetic acid methanol (1:3), and stored at -20°C. The slides pretreatment were made by usual air dry method. C-banding and staining was carried out by barium hydroxide and Giemsa solution. Sixteen chromosomes of this species were observed and divided into two groups, 4 metacentric (no.1, 4, 7, 11), 12 sub metacentric and sub telocentric.

Abstract: The variation of chromosomes of Gansu Black pig was caused by infusion of Duroc blood. The chromosome length ratios of No.1,6,7,13 chromosomes in the line I of Gansu Black pig were higher than those in the line II. It showed that the homogeneity in line I was higher than in line II. The number of Ag-staining nucleolus organizers (Ag-NOR) was between 1-4 in the line I and II, the average No. of Ag-NOR was different obviously between the line I and the line II. This indicated that the activity of translation in rRNA in the line I was different in the line II.

Abstract: The chromosomes of Aprostocetus eurytomae (Nees, 1834) are studied for the first time. The karyotype of this species has five pairs of metacentric chromosomes of gradually decreasing length. Chromosome morphology and number in Aprostocetus eurytomae is very similar to that of Aprostocetus (s.str.) sp. (group epicharmus) as reported by Gokhman (2004).

Abstract: The chromosomes were identified individually and standard karyotype of Lycopersicon esculentum cv. Oxheart was formulated. Roots were collected from germinating seeds and were pre-treated in a saturated aqueous solution of monobromonaphthalene. After iron-alum mordanting, standard hematoxylin method was followed. 2n = 24 chromosomes were found. Karyotype analysis was carried out following a quantitative method using scatter diagram technique. Only two chromosomes were identified individually and the rest were morphologically characterized. One of the two identified chromosomes had mean total length 3.1μ and mean arm ratio 1.3 whereas, the other one had mean total length 2.5μ and mean arm ratio 2.0. The length of the prometaphase chromosomes ranged from 1.3 to 3.1μ. The karyotype of the studied tomato genotype consists of 8 metacentric and 4 sub-metacentric chromosomes. One pair of satellited chromosomes was also observed.

Abstract: The analysis of karyotype of an endangered plant endemic to China, Paeonia delavayi var. lutea is presented. The karyotype formula is K(2n)=10=6m (2SAT) 2sm 2st(SAT). The micro-satellites are on the short arms of 3rd and 5th pairs of chromosomes. According to the Stebbins' classification of karyotype asymmetry, the karyotype belongs to"2A" type. Based on the comparison of propotional length of chromosomes of P. delavayi var. lutea with that of other plants in section Moutan, we discussed the interspecific relationship and presented cytological evidence of subsection division in section Moutan.

Abstract: Using usual method, we got karyotype of hybrid pig (wild soar (male symbol) x domestic pig (female symbol)), C-band and approximate 174 bands of G-band, and we also obtained approximate 258 bands of high resolution G-band by micro-Colchicin method. The result indicate that the diploid chromosome number is 2n=38; there is polymorphism in C-band,and compared with domestic pig in G-band and high resolution G-band there is no distinguish difference. They belong to the same seed.

Abstract: The karyotype analysis based on mitotic cells in rice, Oryza sativa L. (2n=24), was conducted. Root tips were treated with KCl and enzyme and stained with Giemsa. Twelve pairs of chromosomes are arranged according to the length. The karyotype of rice analysed in this work is n =5m+6sm+1st and 2 pairs of satellite chromosomes were observed in numerous cells.
