Abstract: The located culture of cells on patterned surfaces is useful for tissue engineering, biosensor development and fundamental research of cell biology. It is presented here a rapid fabrication method of Biotin-Avidin protein layers m icropattern, which is based on soft-lithography technology. The bovine aortic e ndothelial cells are cultured on the micropatterned surface. It is found that ce ll location can be controlled on the scale of individual cell by this method.

Abstract: This report presents biotin-functionalized semiconducting polymers that are based on fluorene and bithiophene co-polymers (F8T2). Also presented is the application of these polymers to an organic thin film transistor used as a biosensor. The side chains of fluorene were partially biotinylated after the esterification of the biotin with corresponding alcohol-groups at the side chain in F8T2. Their properties as an organic semiconductor were tested using an organic thin film transistor (OTFT) and were found to show typical p-type semiconductor curves. The functionality of this biosensor in the sensing of biologically active molecules such as avidin in comparison with bovine serum albumin (BSA) was established through a selective decrease in the conductivity of the transistor, as measured with a device that was developed by the authors. Changes to the optical properties of this polymer were also measured through the change in the color of the UV-fluorescence before and after a reaction with avidin or BSA.

Abstract: Two-dimensional assmble of biological molecules is one important field in field in the molecular engineering. We.studied the interaction of avidin with phospholipid model membranes con taining biotin-conjugated phospholipids. Using LB technique together with sarface sensitive techniques (such as eilipsometry and sarface plasmon spectroscopy), we studied the nonspecific adsorption and specific binding of avidin to phospholipid lamella, and the effect of the lateral steric hindrance.The primary conclusions of the present work are as following first, the charges of membrane sarface effect the rate of protein adsorption, but do not effect the amount of protein ultimately adsorbed second, the nonspecific adsorption of avidin can be detached by adding a suitable amount of bivalent alkaline-earth metal ions; third, the binding of avidin to membrane bound model receptor is astricted remarkable by the latelal steric hindrence.
Endogenous avidin biotin activity (EABA) in thyroid pathology: immunohistochemical study

Abstract: The immunohistochemical study was performed on paraffin-embedded specimens of surgically resected thyroid tissue from 97 patients with thyroid diseases: 65 patients with papillary carcinoma (PTC), 11 patients with nodular goiter in whom features of benign papillary hyperplasia were found, 9 with lymphocytic thyroiditis (LT), 8 with follicular adenoma, and 4 patients with follicular carcinoma. In PTC immunohistochemical study was performed both in primary tumors and in lymph node metastases. After HIER, incubation with streptavidin from LSAB+ (DakoCytomation) kit was done.Strong cytoplasmic EABA was observed in 56 of 65 (87.5%) PTC and in oxyphilic cells in 8 of 9 cases of LT. Significant correlation between EABA in primary PTC tumor and EABA in lymph node metastases was stated. Normal surrounding thyroid tissues showed absence or weak EABA. Aberrant intranuclear localization of biotin was noted in morules of cribriform-morular variant of PTC. No statistically significant correlation between patient's age, sex, metastases presence and EABA was observed.Among thyroid lesions, false positive reactions are highly probable in papillary thyroid carcinoma and in lymphocytic thyroiditis if immunohistochemical detection is used on systems containing (strept)avidin. The most probable reason is the high endogenous biotin content.For immunohistochemical diagnostics of thyroid diseases, it is very important to choose an optimal visualization method and an appropriate quality control. Entrapment of Avidin in Sol-Gel Derived Silica Glasses

Abstract: Avidin has been entrapped within silica matrices under ambient processing conditions. Properties of the entrapped avidin including stability and its binding with biotin have been investigated by measuring changes in fluorescence. The relative rates of binding to biotin in a range of organically modified silicate were used to compare diffusion properties within gels and their suitability for entrapment of biosensor reagents. Results showed that none of the sol-gel processing conditions used in this study denatured the biomolecules, with the silicate-entrapped generally avidin having better stability towards denaturing conditions than in solution. The avidin retains its binding ability even as the gels aged. Addition of specific organically modified silanes to sol-gel derived materials resulted in a slight improvement in stability and greater accessibility of avidin to external reagents.
Chicken genome analysis reveals novel genes encoding biotin-binding proteins related to avidin family

Abstract: Two separate hits showing significant homology for these N-terminal sequences were discovered. For one of these hits, the chromosomal location in the immediate proximity of the avidin gene family was found. Both of these hits encode proteins having high sequence similarity with avidin suggesting that chicken BBPs are paralogous to avidin family. In particular, almost all residues corresponding to biotin binding in avidin are conserved in these putative BBP proteins. One of the found DNA sequences, however, seems to encode a carboxy-terminal extension not present in avidin.We describe here the predicted properties of the putative BBP genes and proteins. Our present observations link BBP genes together with avidin gene family and shed more light on the genetic arrangement and variability of this family. In addition, comparative modelling revealed the potential structural elements important for the functional and structural properties of the putative BBP proteins.Chickens are known to produce several different proteins which bind biotin in a non-covalent fashion. One of them is avidin, which is expressed by oviduct cells upon progesterone induction and is then transferred to the egg-white where it constitutes a minor fraction of the total protein content of the egg-white [1]. Independently of progesterone, avidin expression is also induced by an inflammation response in almost all of the studied chicken tissues [2]. Another biotin-binder, called literally biotin-binding protein (BBP), is presumably induced by estrogen [3] and secreted from the liver into chicken plasma [4]. From plasma, the BBP is thought to be deposited in egg-yolk [5]. Abstract: In this experiment, a novel biotin-avidin conjugation probe was synthesized and employed in the detection of reverse- phase protein microarray. Firstly, the proportion of the biotin-avidin conjugation probe was optimized. Then the rat IgG and goat anti-rat IgG system was served as a model to optimize the fabrication conditions of reverse-phase protein microarray, including the non-specific absorption of streptavidin-Cy3 molecules, spotting buffer as well as protein activities. At last, the biotin-avidin conjugation probe was applied to the detection of the reverse-phase protein microarray. The results show that the protein microarray prepared by using BSA spotting buffer could prevent non-specific absorptions of fluorescent molecules and improve the sensitivity, effectively. In addition, compared with traditional biotin-avidin system, the detection limit could be improved four times using the biotin-avidin conjugation probe. In conclusion, the biotin-avidin conjugation probe has its merits of easy synthesis, low price and could be further conjugated with other signal amplification techniques, which is promising to be used in the detection of protein microarray.
