ALGINATE DRESSINGS – AN OVERVIEW  [cached]

Abstract: Alginate dressings are highly absorbent, biodegradable material derived from the seaweed. They are used to cleanse wide variety of secreting lesions. They show high absorptivity which is achieved via strong hydrophilic gel formation, this limits the wound secretions, promotes wound healing and also minimizes the bacterial contamination. Alginate dressings are very useful for moderate to heavily exudating wounds. This article throws light on the alginate dressing and its importance medical science. Patents are the richest source of the latest technological information. A review of some patents on alginate dressing is provided that summarizes the recent technical advancements taken place in this area.

Abstract: Alginates are polysaccharides extracted from brown seaweeds. They are used in food industry, pharmaceutical, textile, among other, because of their properties to give high viscous solution and gel forming. This review describes the optimized process at pilot plant level for alginate production. The process includes washing the algae with HCl at pH 4, extraction of the alginate in Na2CO3 solution at pH 10 and heating to 80oC, dilution of the paste and filtrate with a vacuum rotary filter. Alginate precipitation is carried out by adding CaCl2 filtration. The fibers obtained are treated with HCl to obtain alginic acid. The product is neutralized with Na2CO3 to obtain sodium alginate. The product is dried with hot air, milled, and screened at different mesh sizes. We described the different products obtained and their physical and chemical properties. Finally, costs and barriers found that limit the alginate production at commercial level in Mexico are discussed, including the lack of the industrial design, the international cost of the alginates, the policy to give the seaweeds beds concessions, and the role of the investors.

Abstract: The enzyme urease (EC.3.5.1.5) from jack bean meal was immobilized by various techniques, such as entrapment in calcium alginate gel spheres in aqueous suspension; lac impregnated muslin cloth as dry films and by embedding in paraffin wax impregnated muslin cloth. The activity of the free and immobilized enzymes as a function of pH, temperature, storage stability, kinetic parameters and periodic use were compared. The immobilized enzyme showed good storage stability. After repeated use, the alginate beads turned brown and deteriorated, hence the storage stability was not good. The paraffin films were preserved dry because during wet preservation, the film slightly softened and the protein leached out slightly. The alginate beads had moderate mechanical stability. The lac films were tougher than the paraffin wax films in terms of mechanical stability. The Km and Vmax values were altered after immobilization. The Km values for calcium alginate and lac were low, while it was larger in paraffin film as compared to the free enzyme. This may be due to the fact that immobilization on calcium alginate and lac in presence of CaCl2 and methanol exposed certain active sites of the urease. While immobilization on paraffin masks the active sites and may lead to reduced binding of the substrate.

Abstract: In this study, chondrocyte culture was established for the first time in Iran,and calcium alginate was used for longer culture of chondrocyte in vitro. Thestudy was programmed in order to be used for future human chondrocytetransplantation. The cartilage specimen obtained from 50 patients whounderwent total knee and hip operations in Isfahan University of MedicalSciences. Cartilage specimens were used for monolayer as well as suspensionculture in alginate beads. Approximately 12±1 millions cells were harvestedfrom the 3rd passage. The cells were round with large euchromatic nucleusand several nucleoli and small vacuoles. The cells derived from passages 1to 4, which were grown up then, in alginate beads, showed higher stainingwith alcian blue. The harvested cells in some patients were immediately andsuccessfully used for autologus transplantation. This later work will be reportedseparately.

Abstract: OBJECTIVE: To evaluate the cytotoxicity of three different alginate impression materials for orthodontic use. METHODS: Three different brands of alginate were divided into three groups, namely, Group JCO (Jeltrate Chromatic Ortho), OP (Orthoprint) and CO (Cavex Orthotrace). Three control groups were also included: Group C+ (positive control), consisting of detergent Tween 80; Group C- (negative control), consisting of PBS, and Group CC (cell control), consisting of cells not exposed to any material. After manipulating the materials according to the respective manufacturer instructions, samples were made with the use of silicon rings. Then the samples were immersed in Eagle's minimum essential medium (MEM) for 2 minutes. The supernatants were then removed and brought into direct contact with L929 fibroblasts. After exposure to the medium, the cells were incubated for 24 hours. Then 100 μl of 0.01% neutral red dye were added. The cells were incubated again for 3 hours so that the dye could be absorbed. After this 3-hour period, the cells were fixed to perform the viable cell count, using a spectrophotometer (BioTek, Winooski, Vermont, USA) at a wavelength of 492 nm. RESULTS: Statistical differences were found when Groups CC and C- were compared with the other experimental groups. Group JCO had the highest cytotoxicity, followed by Groups OP and CO. CONCLUSION: Based on the results obtained in this work, it was concluded that all alginate impression materials are potentially cytotoxic. Três grupos controle também participaram: controle + (C+), constituído pelo detergente celular Tween 80; controle - (C-) PBS; e controle de célula (CC) onde as células n o foram expostas a nenhum material. Após manipula o dos materiais, seguindo as orienta es do fabricante, foram confeccionados corpos de prova utilizando-se anéis de silicone. Em seguida, esses foram imersos em meio mínimo essencial de Eagle (MEM) por 2min, onde, ent o, procedeu-se à remo o do sobrenadante e à coloca o em contato com fibroblastos L929. Após contato com o meio, as células foram incubadas por mais 24h onde, ent o, foi adicionado o corante vermelho neutro a 0,01%.
