Abstract: 目的 ·构建人源亲环蛋白B(hCypB)的表达载体,通过原核表达体系制备有活性的hCypB。方法 ·通过双酶切法将编码hCypB基因构建到pGEX-6p-1表达载体中,采用亲和层析法一步纯化hCypB,最后通过GST-pulldown验证过表达的hCypB的活性。结果 ·经DNA测序验证,成功构建了hCypB的表达质粒。通过GST柱纯化可以从1 L过夜诱导的大肠埃希菌菌液中获得26 mg,纯度达到90%的GST-hCypB融合蛋白,且GST-pulldown试验证明该重组蛋白可以与脯氨酸-3-羟化酶1(P3H1)及软骨相关蛋白(CRTAP)形成三元复合物。结论 ·利用该实验方法可以在短期内获得大量高纯度、有活性的人源亲环蛋白CypB。
: Objective · To construct an expression vector for human cyclophilin B (hCypB) and prepare active CypB via the prokaryotic expression system. Methods · The DNA sequence enconding hCypB was amplified by PCR and inserted into expression vector pGEX-6P-1 by double enzyme digestion. hCypB was further purified with affinity chromatography and the activity of over-expressed hCypB was verified with GST-pulldown. Results · The recombinant CypB expression plasmid was successfully constructed and verified with DNA sequencing. GST-CypB fusion protein was purified to a purity of 90% by affinity chromatography with a yield of 26 mg of fusion protein from a liter of overnight E. coli culture. GST-pulldown further confirmed that this GST-hCypB fusion protein could form the ternary complex with prolyl 3-hydroxylase 1 (P3H1) and cartilage-associated protein (CRTAP). Conclusion · Large amount of active and highly purified hCypB can be obtained via this experiment method within short time period

Abstract: ?本文通过两个研究探讨社会比较对亲社会行为的影响及其内在机制。研究一中,被试被随机要求与学习成绩排名第一或者最后的人相比,然后测量其助人倾向。结果发现,与向上比较组和控制组相比,向下社会比较会提高个体的助人倾向。研究二通过虚构的智力测验排名反馈操纵社会比较,然后测量被试的捐款意愿。结果发现,得知测验成绩比大多数人好的时候,人们更愿意捐款。两个实验共同表明,社会比较对亲社会行为的影响由个体对弱势群体的同理心所中介。

Abstract: 利用线粒体控制区序列片段(史氏鲟,425bp,达氏鳇,434bp)分析检测了两个养殖场留做后备亲鱼的史氏鲟和达氏鳇的遗传多样性。在所检测的养殖史氏鲟后备亲鱼4个年龄群体共34个体中,发现5个单倍型,共有11个多态位点,占碱基总数的2.6%,无简约信息位点。不同单倍型之间有1~10个变异位点,占碱基总数的0.2%~2.4%。各单倍型之间的遗传距离为0.002~0.024。单倍型多样性hd=0.768,平均核苷酸差异数k=4.367,核苷酸多样性pi=0.011。而分别来自2个养殖场的2个达氏鳇养殖群体都是1个群体仅1个单倍型,2个单倍型之间仅有2个碱基差异,遗传距离为0.005,遗传变异极度缺乏。结果提示在利用史氏鲟和达氏鳇后备亲鱼进行繁殖育苗时要充分注意近交的影响。

Abstract: 在前期研究中分离到噬碳酸盐小球藻(Chlorella sp.X1)的亲环蛋白基因CsCyp1A(登录号:KY207381),编码CsCyp1A蛋白是否具有肽酰脯氨酰顺反异构酶(PPIase)活性功能是进一步研究它参与小球藻耐盐生物学过程的基础。通过His标签的pQE-30-CsCyp1A原核蛋白表达载体,经IPTG诱导它的E.coli M15菌株表达融合蛋白,Nitra-柱纯化后获得纯化的30 kDa左右的His-CsCyp1A融合蛋白质,Western杂交检测到HisCsCyp1A蛋白的杂交信号。酶活性分析显示,HisCsCyp1A融合蛋白中生色基团的产生速度明显快于对照,表明CsCyp1A蛋白可以催化底物N-succinyl-Xaa-Pro-Phe-p-nitroanilide中Xaa-Pro肽键的顺反折叠,说明纯化的CsCyp1A蛋白具有PPIase活性。典型的亲环蛋白家族成员具有肽酰脯氨酰顺反异构酶(PPIase)活性,参与折叠和转运、信号转导、免疫调节、细胞凋亡等生物学过程。真空渗入法侵染转化的拟南芥在35S启动子驱动下超表达CsCyp1A基因,耐盐性研究表明它提高了过表达株系对NaCl胁迫的耐性,揭示小球藻CsCyp1A基因的抗盐性功能,它将成为抗逆育种的基因资源。本研究也为探索小球藻亲环蛋白A的抗盐碱胁迫生物学作用和分子育种奠定了基础。

Abstract: Coronaviruses infect a variety of mammalian and avian species and cause serious diseases in humans, cats, mice, and birds in the form of severe acute respiratory syndrome (SARS), feline infectious peritonitis (FIP), mouse hepatitis, and avian infectious bronchitis, respectively. No effective vaccine or treatment has been developed for SARS-coronavirus or FIP virus, both of which cause lethal diseases. It has been reported that a cyclophilin inhibitor, cyclosporin A (CsA), could inhibit the replication of coronaviruses. CsA is a well-known immunosuppressive drug that binds to cellular cyclophilins to inhibit calcineurin, a calcium-calmodulin-activated serine/threonine-specific phosphatase. The inhibition of calcineurin blocks the translocation of nuclear factor of activated T cells from the cytosol into the nucleus, thus preventing the transcription of genes encoding cytokines such as interleukin-2. Cyclophilins are peptidyl-prolyl isomerases with physiological functions that have been described for many years to include chaperone and foldase activities. Also, many viruses require cyclophilins for replication; these include human immunodeficiency virus, vesicular stomatitis virus, and hepatitis C virus. However, the molecular mechanisms leading to the suppression of viral replication differ for different viruses. This review describes the suppressive effects of CsA on coronavirus replication.

Abstract: Trans/cis prolyl isomerisation is involved in several biological processes, including the development of numerous diseases. In the HIV-1 capsid protein (CA), such a process takes place in the uncoating and recruitment of the virion and is catalyzed by cyclophilin A (CypA). Here, we use metadynamics simulations to investigate the isomerization of CA's model substrate HAGPIA in water and in its target protein CypA. Our results allow us to propose a novel mechanistic hypothesis, which is finally consistent with all of the available molecular biology data.
