Abstract: 为探讨益生真菌对大花蕙兰组培苗生长的促进效应,从野生兰科植物根系分离的内生真菌中筛选出2株真菌菌株C2y1和S1g1,对其进行形态特征和ITS序列鉴定,分析2株真菌共生对大花蕙兰幼苗营养生长、矿质元素吸收以及叶绿素含量的影响,并检测了真菌在兰花根系中的定殖特点。结果表明:2株真菌能促进大花蕙兰植株根系生长、总生物量增长和叶绿素含量增加;C2y1相比于对照能显著促进大花蕙兰幼苗对N、P、K、Fe等矿质元素的吸收,尤其是菌根化苗Fe元素含量高出对照163.24%;菌株S1g1则显著促进植株对Ca、Mn、Zn 的吸收,处理植株的Ca和Zn含量高出对照62.00%和68.09%;形态结构和ITS序列分析鉴定表明C2y1为兰科丝核菌类瘤菌根菌属(Epulorhiza sp.)真菌,S1g1为黄丝菌科Cephalotheca属真菌。显微结构分析表明,C2y1菌丝可侵入兰根皮层细胞,并形成典型的菌丝结,而S1g1仅在兰根的根被组织聚集。菌根化可有效促进大花蕙兰组培苗的营养生长和矿质元素吸收,而不同种类的真菌对其促进效应表现出一定程度的互补性。
The vegetative growth of Cymbidium hybridum plantlets could be promoted through artificial mycorrhizae. In this study, two endophytes(C2y1 and S1g1) obtained from roots of wild orchids exhibited obvious growth-promoting effects on the C. hybridum plantlets through increasing root length,biomass and chlorophyll content.Furthermore,isolate C2y1 increased the N,P,K,Fe contents of the symbiotic plantlets.Especially the Fe content of the treated plants increased by 163.24% compared with the control.Isolate S1g1 significantly increased the accumulation of Ca,Mn,Zn (by 62%,32%,and 68.09%,respectively) in symbiotic plantlets compared with the control.Combining with internal transcribed spacer sequence (ITS) and the morphological characteristics,isolate C2y1 and S1g1 was identified as Epulorhiza sp. and Cephalotheca sp.,respectively.Microscopic studies showed that hyphae of isolate C2y1 infected the root cortex cells and formed typical orchid mycorrhizae,whereas hyphae of isolate S1g1 aggregated in the host velamen cells.It is indicated that artificial mycorrhization can effectively promote the vegetative growth and mineral element absorption for Cymbidium plantlets.Different fungi exhibit some degree of complementary promoting effects.

Abstract: 【目的】香草兰为多年生热带经济作物,随着种植年限的增加,植株长势弱,土壤有益微生物减少,土壤微生物区系失衡,严重制约了香草兰产业的可持续发展。枯草芽孢杆菌作为一种根际促生菌,被广泛应用于促进作物生长,改善土壤微生物环境。本文将枯草芽孢杆菌Y-IVI接种在有机肥上,生产了生物有机肥,并就该生物有机肥对香草兰生长的影响进行了研究。【方法】采用温室盆栽试验,调查施用根际促生菌枯草芽孢杆菌(Bacillussubtilis)Y-IVI及其经固体发酵制得的微生物有机肥料(Y-IVI:3×108cfu/g)后,香草兰植株地上部及根系的生长状况,采用选择性培养基方法研究了Y-IVI在香草兰根际土壤中的定殖能力及对香草兰根茎腐病致病菌-尖孢镰刀菌数量的影响。【结果】施用Y-IVI及BIO4个月后,香草兰根际土壤Y-IVI数量仍可达到106cfu/g土,二者无显著差异,在处理OF和对照中未检测到菌株Y-IVI。施用生物有机肥香草兰地上部干重和根系干重均显著高于对照,分别增加了63.1%和59.4%,与不接种Y-IVI的有机肥处理(OF)相比,地上部干重显著提高了43.2%,根系干重提高了18%,差异不显著;施用Y-IVI菌液的处理植株地上部干重和根系干重均高于对照,但无显著性差异;处理BIO根系直径、根系表面积和总体积与对照相比分别增加了41.9%、88.9%和80.4%,均显著高于对照,总根长与对照差异不显著;处理BIO根系表面积和总体积与有机肥处理OF相比分别显著增加了41.9%和30.8%,根系直径与OF相比增加了10.1%,差异不显著;处理Y-IVI根系直径与对照相比显著增加了25.5%,但根系表面积和总体积与对照差异不显著;与对照相比,施用BIO及Y-IVI的处理根际土壤尖孢镰刀菌数量分别明显降低了52.2%和41.8%,施用有机肥OF的处理降低了10%,差异不显著。【结论】Y-IVI可稳定定殖于香草兰根际土壤对其生长起有益作用,含促生菌Y-IVI的生物有机肥料比单独使用促生菌菌液可以更有效地减少根际土壤中尖孢镰刀菌数量,降低连作生物障碍。施用生物有机肥料比施用化肥和有机肥更有效地促进香草兰地上部及根系生长,因此,施用由根际促生菌枯草芽孢杆菌(Bacillussubtilis)Y-IVI制得的生物有机肥是解决香草兰连作生物障碍和提高收益的有效手段。

Abstract: 根据兰科植物(Orchidaceae)蝴蝶兰(Phalaenopsis)的肌动蛋白基因(Actin)序列设计跨内含子引物,分别以cDNA第一链和基因组DNA为模板,采用RT-PCR和PCR方法从墨兰(Cymbidium sinense)、春兰(C. goeringii)中分离出Actin基因的同源片段。序列分析结果表明:墨兰和春兰Actin基因片段长度均为1335 bp,其中编码区长度均为1086 bp,编码362个氨基酸,分别命名为CsACT1和CgACT1(GenBank登录号分别为GU181353、GU181354)。CsACT1和CgACT1均含有3个内含子,且内含子位置非常保守。其氨基酸序列与其他植物的同源性均在90%以上,具有高度的保守性。CsACT1和CgACT1在根、茎、叶、花梗、花蕾以及花器官中都有表达,且表达量基本一致,因此,推断其为组成型表达的肌动蛋白基因。

Abstract: Abstract: Objective] In order to obtain the effective plant growth-promoting bacteria, we studied the diversity of siderophore-producing endophytic bacteria in the roots of Cymbidium goeringii. Methods] We screened for siderophore-producing bacteria from 189 root endophytic strains of Cymbidium goeringii by using chrome azurol S (CAS) agar plate assay. The diversity of siderophore-producing endophytic bacteria was investigated by 16S rRNA gene sequence analysis. Results] We obtained 47 siderophore-producing strains (24.9% of the total strains). Sequence analysis revealed that 47 strains were members of 31 species of 17 genera in four phylogenetic groups (Alphaproteobacteria, Betaproteobacteria, Firmicutes, Actinobacteria). The dominant group was Actinobacteria (42.6%), and the dominant genera were Bacillus and Variovorax. The strains of Variovorax produced relatively high levels of siderophore. In addition, there may be two novel taxonomic units. Conclusion] The results show that there are abundant species diversity of siderophore-producing endophytic bacteria in the roots of Cymbidium goeringii.

Abstract: The fungal strain CL-3 was isolated from the root of wild Cymbidium goeringii with using conventional isolation method. The inoculum of strain CL-3 was used to study its interaction with its host plant. According to the morphological features examination and rDNA ITS sequence analysis, the closest relative of the strain was Acremonium strictum, with the similarity of 100%. After two months of symbioses culture, the fresh biomass of C. goeringii inoculated with CL-3 increased by 80.5%, and the endophytic CL-3 was re-isolated from the seedlings. The statistics showed that the difference between inoculation group and control group was significant. The presence of the fungus in the tissues was stud-ied with paraffin-cut section. The results showed that the fungus was observed in inner tissues of roots, indicating that the symbiosis relationship between Cymbidium goeringii and fungi could be established.

Abstract: 以樟树油、茶树油、桂皮油、香茅油、估皮油、薄荷油和丁香油7种精油为材料,测定不同浓度精油对香荚兰根腐病尖镰孢菌菌丝生长和孢予萌发的影响。结果表明,桂皮油、丁香油、樟树油和香茅油对病菌孢子萌发和菌丝生长均有很强的抑制作用。其中以16.0μl/ml的桂皮油、丁香油的抑制作用最强,对孢子萌发和菌丝生长的抑制率分别为100%,99.74%和100%,100%。利用桂皮油和丁香油进行温室盆栽防病试验,均表现出较好的防治效果,在二者用量为2.0μl/ml时防效分别为81.18%和80.02%。

Abstract: 应用~(15)N同位素标记菌体和Tn5转座子诱变的突变株研究固氮粪产碱菌对宿主水稻的联合进程,结果表明,粘附作用是结合过程的第一步,接种3h后粘附在根表的菌体数量达最大值,约占接种量的3.7%。粘附的菌体易脱离根表,表明粘附作用是细菌与宿主植物间的弱的相互作用。在根表定位是结合过程的第二步。接种15h后,定位菌体数量达最大值,约占接种量的21%。经振荡处理不能使定位菌体脱离根表。定殖是结合过程的第三步,接种20h后定殖菌体紧密结合于根表,剧烈振荡亦不易使其脱离根表。定殖后菌体开始出现果胶酶活性。采用趋化(Che)和胞外多糖(Exo)突变株研究表明,趋化性和胞外多糖的合成是影响细菌在根表结合的重要因素。粪产碱菌在水稻根表没有专一的粘附部位,但粪产碱菌的定位和定殖主要发生在主根根表,尤其是主、侧根交接处。
