Abstract: 3G11~6C10~CD4+CD8~胸腺髓质细胞是CD4单阳性(SP)胸腺细胞中的亚群之一 ,表型为TCRαβ+CD69loHSAmed/lo,可接受ConA刺激产生增殖应答和细胞因子分泌 所分泌的细胞因子以Th2型为主 ,还有少量IFNγ存在 ,是处于CD4SP成熟过程的中后期 ,Th0向Th2转化的过渡型细胞 为探讨胸腺微环境中基质细胞对这一亚群细胞的作用 ,在研究体系中加入一株胸腺髓质型上皮细胞系MFEC1 ,发现MTEC1可促进ConA刺激的3G11~6C10~CD4+CD8~胸腺髓质细胞增殖和IL 6分泌 ,但IL-4及IL-10的产生部分地受到抑制 ,不增加IFNγ分泌 ,亦不诱导IL-2产生 .
Characterization of a murine thymic CD4+ T cell subset-TCRαβ+ 3G11 6C10 CD4+ CD8 thymocytes

Abstract: The presence of a relatively mature CD4+ CD8 (SP) T cell subset in mouse thymus has been demonstrated. Composing of 10% of total CD4SP thymocytes, this subset is defined by the absence of 3G11 and 6C10 expression with a phenotype of CD69+/ , HSAmed/lo and heterogeneous for Qa-2 expression. The proliferation capability of TCRαβ+ 3Gl l 6C10 CD4+ CD8 thymocytes was high while using Con A stimulus. And Con A stimulation could result in secretion of IL4, IL-10, IL-6 and a little amount of IFNγ. IL-2 was barely detectable. This is distinct from typical Th0 type cytokines. The cells of this subset were NK1.1 negative, but strongly expressed GATA-3 mRNA. The results suggest that the CD4+ subset of 3G11 6C10 NK1.1 phenotype possesses immunocompetent cells with functions characteristic of Th2-like cytokines, which may indicate the cells at transitional status from Th0 to Th2, with a propensity to Th2.

Thymocyte apoptosis and proliferation modeling during rat thymic involution is influenced by ovarian hormones in a thymocyte subset-specific manner

Abstract: The study was aimed to define the putative role of ovarian hormones in shaping thymocyte apoptosis and proliferation during thymic involution. Thymocytes from young adult and middle-aged rats ovariectomized (Ox) before puberty were examined for apoptosis and proliferation. Apoptosis and proliferation were measured in fresh thymocyte suspensions and in their 18-hour cultures, and fresh thymocyte suspensions, respectively. The thymocyte population and the major thymocyte subsets were analyzed following triple staining using anti-CD4 and anti-CD8 monoclonal antibodies and 7-AAD to label apoptotic or proliferating cells. The frequency of apoptotic cells was lower in thymocyte suspensions and cultures from Ox rats of both ages. This reflected in a diminished frequency of apoptotic cells amongst CD4+CD8+ double positive (DP) and CD4+CD8- single positive (SP), and DP cells in young and middle-aged Ox rats, respectively. Additionally, in thymocyte cultures from Ox rats the frequency of apoptotic cells amongst CD4+CD8- and CD4-CD8+ SP cells decreased with age, but increased within DP and CD4-CD8- double negative (DN) subsets, reaching in the former subset from middle-aged Ox rats higher values than in age-matched controls. The frequency of proliferating cells was also lower in Ox rats than in controls. This reflected the lower frequency of cycling cells amongst CD4+CD8- SP and CD4-CD8+ SP thymocytes in young rats, and DP and CD4-CD8+ SP thymocytes in middle-aged rats. Besides, in both SP and DP thymocyte subsets from Ox rats the frequency of proliferating cells declined with age. In conclusion, thymocyte apoptosis and proliferation exhibit ovarian hormone-dependent thymocyte subset specific alterations during thymic involution. Abstract: Phenotypic analysis of the medullary-type CD4 CD8+ (CD8SP) thymocytes has revealed phenotypic heterogeneity within this cell population. The phenotype of mature peripheral CDS+T cells is TCRαβ+CD3+Qa-2+HSA 3G11 6C10, whereas in the medullary-type CD8SP thymocytes, 20% are Qa-2+; 33%, HAS; 30%, 3G11; and 70% are 6C10. The disparate expression patterns of these four cell surface markers suggest that medullary-type CD8SP thymocytes may undergo phenotypic maturation process. According to the distribution of these four cell surface markers, six subgroups of CD8SP thymocytes have been identified. The precursor-progeny relationship along with developmental pathway is postulated as follows: 6C10+HSA+3G11 Qa-2→ 6C10+HSA+ 3G11+Qa-2 → 6C10 HSA+3G11+Qa-2 → 6C10 HSA 3G11+Qa-2 → 6C10 HSA 3G11 Qa-2 → 6C10 HA S 3G11 Qa-2+, the cells in the last subgroup exit the thymus and home into periphery.

Analysis on the types of chemokines expressed by the murine thymic epithelial cell line MTEC1

Abstract: Six kinds of chemokines including SDF-1α, IP-10, KC, RANTES, MCP-1 and Lymphotactin have been amplified using the primers designed by ourselves with RT-PCR from the total RNA of a medullary-type murine thymic epithelial cell line MTEC1 cells. The amplified cDNA fragments of the chemokines have been identified by nucleotide sequencing and Northern blot analysis. The results demonstrate that there are two different splicing forms of SDF-1 mRNA existing in MTEC1. The concentrated culture supernatant of MTEC1 shows an intermediate level of chemotaxis to CD4 CD8 thymocytes, which conforms to the biological function of SDF-1.

The results demonstrate that there are two different splicing forms of SDF-1 mRNA existing in MTEC1. Abstract: 对胸腺髓质型CD4-CD8+细胞进行表型分析,发现这群细胞表型不均一.外周成熟的CD8+T细胞表型为TCRαβ+CD3+Qa-2+HSA-3G11-6C10-.而胸腺髓质型CD4-CD8+细胞Qa2+细胞占20%,HSA-者占33%,3G11-者占30%,6C10-者占70%.这4种表面标志表达的不同提示髓质区CD8单阳性细胞仍需经历一个表型成熟的过程.根据这4种表面标志在细胞表面分布的比例,推测髓质区CD8单阳性细胞可能经历以下表型成熟阶段6C10+HSA+3G11-Qa2-→6C10+HSA+3G11+Qa2-→6C10-HSA+3G11+Qa2-→6C10-HSA-3G11+Qa2-→6C10-HSA-3G11-Qa2-→6C10-HSA-3G11-Qa2+,然后迁出胸腺,到达外周.
